ABSTRACT: Eupalinolide B (EB), a compound isolated from Eupatorium lindleyanum, has been evaluated for its anticancer effects on pancreatic cancer cells and xenograft models. Our research presents a multi-faceted investigation of EB, including assessments of its impact on cell viability, proliferation, migration, invasion, and potential mechanisms of action.

Integration of whole genome sequencing analysis with unique patient-derived models reveals clinically relevant drug targets in TFCP2 fusion-defined intraosseous rhabdomyosarcoma

Project description:Here we present comprehensive, multi-omic and functional analysis, to discover and preclinically validate novel actionable molecular targets for a rare case of intraosseus rhabdomyosarcoma with TFCP2-rearrangement. Whole genome sequencing analyses of matched patient tumour and xenograft material revealed alterations in gene networks associated with the oncogenic, potentially targetable PI3K/AKT pathway. Preclinical assessments revealed that targeting the pathway with a small molecule PI3K/mTOR inhibitor dactolisib presents a promising treatment approach for this rare cancer, decreasing cancer cell viability in vitro and significantly reducing tumour growth in vivo. Parallel identification of the codeletion of adjacent genes CDKN2A and MTAP in these tumours, led us to further explore PRMT5 inhibition as a potential therapeutic approach.

Gene expression data of nasopharyngeal carcinoma (NPC) cell and Thiostrepton treatment

Project description:Thiostrepton, a natural antibiotic, has recently been shown to be a potential anticancer drug for certain cancers, but its study in nasopharyngeal carcinoma (NPC) is still limited. The aims of this study were to investigate the anticancer effect of thiostrepton on NPC cells and to explore its underlying mechanism. Our results showed that thiostrepton reduced NPC cell viability in a dose-dependent manner. Thiostrepton inhibited the migration and invasion of NPC cells in wound healing and cell invasion assays. Thiostrepton effectively suppressed NPC cell proliferation, migration, and invasion, likely by several mechanisms. Thiostrepton may be a potential therapeutic agent for treating NPC in the future.

Antipsychotic Drug Trifluoperazine as a Potential Therapeutic Agent against Nasopharyngeal Carcinoma

Project description:Trifluoperazine (TFP), a typical antipsychotic primarily used for treating schizophrenia, exhibits anticancer effect on several types of cancer in recent years. Nevertheless, the effect of TFP on na-sopharyngeal carcinoma (NPC) is still unknown. In this study, we aimed to evaluate if TFP can be the potential therapeutic agent against NPC and to identify its underlying molecular mechanisms. We used four NPC cell lines, namely TW01, TW03, TW04, and BM, to assess the anticancer effects of TFP using cytotoxicity, wound healing, colony formation, and cell invasion assays. RNA se-quencing combined with Ingenuity Pathways Analysis was performed to identify the mechanism by which TFP influences NPC cells. Our data revealed that TFP decreased NPC cell viability in a dose-dependent manner. The invasion and migration of NPC cells were inhibited by TFP. RNA sequencing showed several anticancer molecular mechanisms after TFP administration. It is promising that the antipsychotic drug TFP can be used as a potential therapeutic regimen in NPC treatment in the future.

TOV112D cells treated with NSC319726

Project description:Rescuing the function of mutant p53 protein is an attractive cancer therapeutic strategy. Using the NCI anticancer drug screen data, we identified two compounds from the thiosemicarbazone family that manifest increased growth inhibitory activity in mutant p53 cells, particularly for the p53R175 mutant. Mechanistic studies reveal that NSC319726 restores WT structure and function to the p53R175 mutant. This compound kills p53R172H knock-in mice with extensive apoptosis and inhibits xenograft tumor growth in a 175-allele specific mutant p53 dependent manner. This activity depends upon the zinc ion chelating properties of the compound as well as redox changes. These data identify NSC319726 as a p53R175 mutant reactivator and as a lead compound for p53 targeted drug development. We utilized gene expression microarrays to examine the transcriptional activity of p53 targets in TOV112D cells after treatment with NSC319726. Triplicate cultures of TOV112D cells untreated, treated with NSC319726. Total RNA was extracted and hybridized to Affymetrix human U133 plus 2.0 microarrays.

Gene expression profiles of BxPC-3, MiaPaCa-2 and PANC-1 cell lines treated with DMSO and THZ1 respectively

Project description:By screening an epigenetic-related compound library, we identified THZ1, a covalent inhibitor of CDK7, as a promising candidate. Multiple long-established and patient-derived PDAC cell lines (PDC) were used to validate the efficacy of THZ1 in vitro. In addition, patient-derived xenograft (PDX) models and animal models of PDAC were utilized for examining THZ1 efficacy in vivo. Furthermore, RNA-Seq and H3K27Ac-based Super-Enhancers (SEs) analyses were performed to reveal the molecular mechanism of THZ1 treatment. Lastly, PDAC cell lines with primary or acquired resistance to THZ1 were investigated to explore the potential mechanism of THZ1 susceptibility.

Downregulation of chemokine (C-C motif) ligand 5 (CCL5) induced by a novel 8-hydroxyquinoline derivative (91b1) suppresses tumor invasiveness in esophageal carcinoma

Project description:Esophageal squamous cell carcinoma (ESCC) is a particularly aggressive form of cancer with high mortality. In this study, a novel 8-hydroxyquinoline derivative (91b1) was investigated for the anticancer activities in ESCC alongside the associated mechanisms. The in vitro cytotoxic effect of 91b1 were evaluated across five ESCC cell lines using MTS assay, with cisplatin serving as a comparative standard. Changes in gene expression profile were identified by cDNA microarray and further validated by qPCR and immunostaining. Additionally, protein levels of the most significantly downregulated target in archival ESCC samples was also studied. 91b1 demonstrated comparable anticancer effect with cisplatin. Notably, CCL5 was identified as the most substantially downregulated gene, with its suppression at both mRNA and protein expression in ESCC cells, exhibiting a dose-dependent manner. The recombinant human protein of CCL5 enhanced the invasion of ESCC cells using the trans-well assay. The upregulation of CCL5 protein was also detected in 50% of ESCC cell lines. CCL5 was also overexpressed in 76.9% of ESCC specimens. The overall results indicated that 91b1 could effectively induce anticancer effect on ESCC cells through downregulating CCL5 expression with suppression of tumor invasion. Overall, these findings suggest that 91b1 effectively inhibits ESCC cell proliferation and tumor invasion by downregulating CCL5 expression, highlighting its potential as a therapeutic agent for ESCC treatment. We used microarrays to identify the most regulated gene expression of KYSE 150 cells treated with compound 91b1.
