Next Generation Sequencing Facilitates Quantitative Analysis of COL,uvr8 mutant treated with ABA or UV-B Transcriptomes

ABSTRACT: To expore whether UV-B induce the expression of ABA-responsive genes, we compared transcriptomes of 10-d-old Col-0 and uvr8 seedlings ± 50 uM ABA treatment and 3-h exposure to 5 mol m–2 s–1 UV-B or white-light control.

iSB09 and iCB elicit ABA-responsive gene expression in Solanum lycopersicum seedlings

Project description:To compare the genome-wide transcriptional effect of ABA and iSB09 in tomato plants, we performed RNA-seq analysis of mock-, 10 uM ABA- or 20 uM iSB09-treated plants. Differential gene expression analysis between mock- and ABA-treated or iSB09-treated seedlings was done with DESeq2 and genes with an absolute value of log2 fold change (log2FC) > 1 or (log2FC) < -1 and p-adjusted value (padj) < 0.05 were selected. iSB09 upregulated and downregulated genes represent a subset of the ABA-responsive genes, which reflects the activation of PYL1-like and PYL4-like ABA receptors in tomato seedlings. Additionally, to compare the genome-wide transcriptional effect of ABA and iCB in tomato plants, we performed RNA-seq analysis of mock-, 10 uM ABA- or 10 uM iCB-treated plants. Differential gene expression analysis between mock- and ABA-treated or iCB-treated seedlings was done with DESeq2 and genes with an absolute value of log2 fold change (log2FC) > 1 or (log2FC) < -1 and p-adjusted value (padj) < 0.05 were selected. iCB mimics ABA transcriptional response through activation of the three subfamilies of ABA receptors.

Transcriptome analysis of Arabidopsis thaliana photoreceptor mutants impaired in UV and blue light signaling

Project description:Plants use sunlight as a source of energy for photosynthesis but also as an important environmental cue to regulate growth, development and light acclimation. Wavelengths in the UV-B (280-315 nm) and UV-A/blue (315-500 nm) regions of the spectrum are perceived by UV RESISTANCE LOCUS 8 (UVR8) and cryptochromes (CRY1 and CRY2), respectively. Despite recent advances in our understanding of how these photoreceptors promote photomorphogenesis, very little is known about the molecular mechanisms regulated by UVR8 and CRYs in sunlight exposed plants. Here, a factorial experiment was designed to assess the roles of UVR8 and CRYs in regulating transcriptome wide changes, hormone accumulation, and growth competence of Arabidopsis thaliana plants exposed to solar UV-B, UV-A, and blue radiation.

Flavonoid pathway intermediates implicate UVR8 in functions beyond canonical UV-B signaling

Project description:Plants integrate light signals to regulate development and produce protective compounds such as flavonoids under excessive light stress. While these metabolites protect against light damage, their roles in modulating photoreceptor responses remain unclear. Here we show that naringenin chalcone (NGC), a flavonoid precursor that accumulates in the Arabidopsis chalcone isomerase mutant tt5 but which is also present in wild type plants under solar light, promotes UVR8 monomer accumulation beyond its canonical UV-B response. A genetic suppressor screen reveals that loss of UVR8 restores normal growth of tt5 mutants grown under high light, irrespective of the presence of UV-B. Biochemical and transcriptomic analyses show that NGC binds to monomeric UVR8, stabilizing its active form and triggering gene expression changes, even in the absence of UV-B. This work identifies a mechanism for stabilizing activated UVR8 and reveals crosstalk between light-induced metabolic intermediates and photoreceptor-mediated developmental regulation.
