CD73 enhances hepatic Treg functions by enzymatic and non-enzymatic pathways in NAFLD development [CUT&Tag]

ABSTRACT: CD73 is known as a functional molecule highly expressed by Treg and increasing after FFA (free fatty acids) stimulated. We found that FFA upregulated the transcription factor Gata2 expression in Treg. CUT&Tag assay was performed to investigate the Gata2 binding promoter sites in Treg.

GPSM1 restricts CD73+CD103+ Treg cells in adipose tissue, critical for promoting obesity-related metabolic deterioration

Project description:Regulatory T cells (Tregs) have been considered as a key regulator of adipose homeostasis and metabolic health. However, the underlying regulatory mechanisms remain unclear. Here, we report G-protein-signaling modulator 1 (GPSM1) expression in CD4+ T cells of peripheral blood or visceral fat is significantly correlated with human obesity and glucose dysregulation. CD4+ T or Treg cell-specific deletion of GPSM1 mice exhibit increased numbers of Treg in adipose tissues, restrained inflammation and improved insulin and glucose tolerance under a high fat diet. These effects are mediated through maintaining a new CD73+CD103+ Treg subpopulation we identified that mainly accounts for enhanced adipose thermogenesis and suppressive inflammation after GPSM1 deletion, which is revealed by single-nucleus RNA sequencing. By contrast, CD4+ T cell-specific GPSM1 overexpression mice have less numbers of Tregs and are more prone to adipose tissue dysfunction and metabolic deterioration. Mechanistically, RHOA-cell stiffness-TAZ axis mediates the effects of GPSM1 on the abundance of Treg. Furthermore, adoptive transfer of GPSM1-deficent Tregs promotes energy expenditure and improve metabolic performance in Rag1-/- mice, suggesting a therapeutic potential for Treg against obesity and related disorders.

Expression data from CD73 positive adipose derived mesenchymal stem cells (CD73+ AD-MSCs) and CD73 negative adipose derived mesenchymal stem cells (CD73- AD-MSCs) of mouse

Project description:MSCs are a heterogeneous population and the specific population of MSCs may exhibit a selective ability for tissue repair. The aim of our research was to adapt the CD73+ subgroup of adipose derived MSCs (AD-MSCs) for the therapy of myocardial infarction (MI). Our results revealed that CD73+ AD-MSCs played more effective role in the acceleration function of cardiac recovery by promoting angiogenesis in a rat model of MI compared to mixed AD-MSCs and CD73- AD-MSCs. Microarray analysis shows differences between CD73+ and CD73- AD-MSCs when transcription profile of these two subgroups were compared, especially in VEGF pathway.

Preventive Treatment with a CD73 Small Molecule Inhibitor Enhances Immune Surveillance in K-Ras Mutant Pancreatic Intraepithelial Neoplasia

Project description:Immunoprevention is an emerging consideration for solid tumors, including pancreatic ductal adenocarcinoma (PDAC). We and others have shown that Kras mutations in genetic models of spontaneous pancreatic intraepithelial neoplasia (PanIN), which is a precursor to PDAC, results in CD73 expression in the neoplastic epithelium and some populations of infiltrating immune cells, including macrophages and CD8 T cells. CD73 is an ecto-enzyme that converts extracellular adenosine monophosphate (AMP) to adenosine, a critical immune inhibitory molecule in PDAC. We hypothesized inhibition of CD73 would reduce the incidence of PanIN formation and alter the immune microenvironment. To test our hypothesis, we used the KrasG12D; PdxCre1 (KC) genetically engineered mouse (GEM) model and tested the utility of AB-680, a small molecule inhibitor targeting CD73, to inhibit PanIN progression. AB-680, or vehicle control, was administered using oral gavage delivery three days/week at 10mg/kg, beginning when the mice were two months old and lasting three months. We euthanized the mice at five months old. In the KC model, we quantified significantly less pancreatitis, early and advanced PanIN, and quantified a significant increase in M1 macrophages in AB-680-treated mice. Single Cell RNA sequencing (scRNA-seq) of pancreata of AB-680 treated mice revealed increased infiltration of CD4+ T cells, CD8+ T cells, and mature B cells. The scRNA-seq analysis showed that CD73 inhibition reduced M2 macrophages, acinar, and PanIN cell populations. CD73 inhibition enhanced immune surveillance and expanded unique clonotypes of TCR and BCR, indicating that inhibition of CD73 augments adaptive immunity early in the neoplastic microenvironment.

Expression profiling of WT and p110delta D910A Treg

Project description:The PI3K pathway has emerged as a key regulator of regulatory T cell (Treg) development and homeostasis and is required for full Treg-mediated suppression. To identify new genes involved in PI3K-dependent suppression we compared the transcriptome of WT and p110delta D910A Tregs. Among the genes that were differentially expressed was CD38. Here we show that CD38 is expressed mainly by a subset of Foxp3+CD25+CD4+ T cells originating in the thymus and on Tregs in the spleen. CD38high WT Tregs showed superior suppressive activity and CD38low Tregs failed to upregulate CD73, a surface protein which is important for suppression. However, Tregs from heterozygous CD38+/- mice were unimpaired despite their lower levels of CD38 expression. Therefore, CD38 can be used as a marker for Tregs with high suppressive activity and the impaired Treg function in p110delta D910A mice can in part be explained by the failure of CD38high cells to develop.

CD73 knockdown effect in pancreatic cancer cell lines

Project description:Recent studies have shown that non-enzymatic function of CD73 play a key role in tumor progression, but this function of CD73 in pancreatic cancer cells has not been studied. In the present study, PANC-1 cell lines were transfected with CD73 siRNA, and proliferation ability and cell cycle was significantly inhibited. However, little is known about the mechanisms involved in CD73 regulation in tumors.
