Analysis of androgen receptor signaling in human osteoblastic, muscle and neuronal cells

ABSTRACT: Androgen signaling through the androgen receptor (AR), a ligand-dependent transcription factor within the steroid receptor superfamily, plays an important role in development and maintenance of multiple tissues including bone, muscle and brain. However, a key unanswered question is the mechanism through which AR-mediated gene expression leads to these effects. To gain further insight into the tissue specific mechanism of AR action in human multiple cells, we investigated gene regulation in three human cells, U2OS, RD and SH-SY5Y (SS5) or cells overexpressing AR using microarray method. In order to investigate the effect of androgen in human cells, we performed gene expression analysis in three human cell lines, U2OS, RD and SS5. We treated cells with vehicle or dihydrotestosterone (DHT).
