Expression data from NC/Nga mice with HDM-induced dermatitis

ABSTRACT: Six applications (twice per week) of HDM cream (Biostir AD) to the back skin and ears of NC/Nga mice resulted in robust phenotype of dermatitis (skin inflammation, itch and barrier function impairement). Expression of an array of genes are altered. IPA and upstream regulator analysis showed similarities to human atopic dermatitis. We used microarray to detail the gene expression to understand the relevant pathways and upstream regulators activated in HDM-induced dermatitis in NC/Nga mice

We evaluated the expression data of five models and their respective controls, and one previously published murine dataset (GSE50400). The included models were groupe 1 (Lesional: Flaky tail and FLG mutant; Control: C57BL/6NCrSlc), Group 2 (Lesional: NC/Nga(+mite); Control: NC/Nga(-mite)), Group 3 (Lesional: OVA-ODT 24h; Control: C57BL/6NCrSlc), and Group 4 (Lesional: OXA 2h; Control: BALB/c,Acetone).

Gene expression changes in mouse skin after exposure to SspA and house dust mite extract

Project description:The skin reprograms its gene expression during dermatitis. This transcriptome profiling examines dermatitis-associated gene expression changes in the skin of mice epicutaneously treated with the Staphylococcus aureus protease SspA, also known as protease V8, and house dust mite extract (HDM). Treatment with SspA and HDM combination results in skin disease in mice that closely resembles atopic dermatitis. The genes whose expression changes in this dermatitis model include cytokines, chemokines, and other molecular mediators of intercellular communication in inflamed skin.

Der f/SEB-induced skin inflammation

Project description:Atopic dermatitis (AD) is a chronic pruritic inflammatory skin disease. We recently described an animal model in which repeated epicutaneous applications of a house dust mite extract and Staphylococcal enterotoxin B induced eczematous skin lesions. In this study we showed that global gene expression patterns are very similar between human AD skin and allergen/staphylococcal enterotoxin B–induced mouse skin lesions, particularly in the expression of genes related to epidermal growth/differentiation, skin barrier, lipid/energy metabolism, immune response, or extracellular matrix. In this model, mast cells and T cells, but not B cells or eosinophils, were shown to be required for the full expression of dermatitis, as revealed by reduced skin inflammation and reduced serum IgE levels in mice lacking mast cells or T cells (TCRb-/- or Rag1-/-). The clinical severity of dermatitis correlated with the numbers of mast cells, but not eosinophils. Consistent with the idea that T helper type 2 (Th2) cells play a predominant role in allergic diseases, the receptor for the Th2-promoting cytokine thymic stromal lymphopoietin and the high-affinity IgE receptor, FceRI, were required to attain maximal clinical scores. Therefore, this clinically relevant model provides mechanistic insights into the pathogenic mechanism of human AD. A total of six samples were analyzed. Back skin samples from healthy or AD-induced C57BL/6, PLC-beta 3 KO (C57BL/6 background), and NC/Nga mice were collected for total RNA extraction. Pooled RNA from 2-4 mice per condition were used for analysis.

Project description:Type 2 inflammation contributes to the pathology of skin diseases such as atopic dermatitis and urticaria. Itch is a common symptom accompanying type 2 skin inflammation. Yet how chronic type 2 inflammation affects itch sensory neurons innervating the skin is poorly understood. The goal of this study was to characterize transcriptional changes in sensory ganglia innervating the skin in the setting of chronic type 2 skin inflammation.

Mouse skin CD45 cells in house dust mite dermatitis

Project description:Despite robust literature associating IL-31 with pruritic inflammatory skin diseases, its influence on cutaneous inflammation and on the interplay between inflammatory and neurosensory pathways remain unmapped. Here, we examined the consequences of disrupting Il31 and its receptor Il31ra in a mouse model of house dust mite (HDM) allergic dermatitis. Il31-deficient mice displayed an increased number and proportion of cutaneous type 2 cytokine-producing CD4 T cells and serum IgE in response to HDM. Single cell RNA-sequencing analysis of skin CD45+ populations from HDM-treated skin revealed that Il4ra+ monocytes and macrophages capable of fueling a feedforward type 2 inflammatory loop were selectively enriched in Il31ra-deficient HDM dermatitis skin. Thus, IL-31 is not strictly a pro-inflammatory cytokine, but rather an immunoregulatory factor that limits the magnitude of type 2 inflammatory responses in skin.
