Indole-3-carboxaldehyde (ICA) mitigates morbidity and mortality in a mouse model of Graft-versus-Host-Disease

Project description:ICA-treatment following allogeneic bone marrow transplant in mice limited GvHD pathology and GVHD mortality, augmented the integrity of the intestinal mucosal barrier, and limited production of inflammatory cytokines, but did not compromise donor T cell-mediated Graft vs. Leukemia (GvL) responses. Following protracted exposure, ICA treatment also induced donor-strain-specific tolerance of engrafted T cells. We used microarray to assess gene expression changes induced by ICA in the small intestine in animals with GvHD.

Deconvolution of melanoma transcriptomes and miRNomes by independent component analysis

Project description:The repositories for various “omics” data collected from different cancer types are constantly growing. However, robust diagnostic and/or prognostic conclusions can often not be extracted from mixed transcriptomes or other heterogenous datasets obtained from large cohorts as important signals or single features can be masked. Here, computational microdissection of bulk transcriptome data was applied to gain insights into patient prognosis and to investigate important processes and cell subtypes within new samples in silico. We developed parallel consensus ICA that decomposes many whole transcriptomes into independent signals (components), some of which originated from distinct cell subtypes while others accounted for technical biases. We show that the weight of components allows for prediction of clinically relevant patient characteristics, which was further validated on an independent patient cohort. Moreover, ICA components can be linked to biological functions, thus new samples could be classified by presence of respective biological properties such as immune signals, angiogenic activity and proliferation. Finally, through integration of different data types (transcriptomes and miRNomes) by ICA, biological functions of miRNAs were deduced, which would otherwise not be possible. Taken together, ICA represents a versatile tool to dissect complex data cohorts into individual components allowing for better use of such datasets.

Gut-derived metabolites drive Th17 cell pathogenicity in Multiple Sclerosis

Project description:The gut-brain axis is an emerging factor in promoting multiple sclerosis (MS). However, the underlying mechanisms and strategies to target this axis are not established. Here, we investigated how the gut environment influences myelin-specific Th17 cell pathogenicity. We used the adoptive Th17 cell transfer experimental autoimmune encephalomyelitis (EAE) model and antibiotic treatment to modulate the intestinal microbiome. We observed a reduced pathogenic Th17 cell signature in the colon of antibiotics-treated mice. Treatment with fecal filtrates enhanced myelin-specific Th17 cell encephalitogenic properties both in vitro and in vivo. Feces metabolomic profiling identified altered tryptophan-derived metabolites, including indole-3-carboxylate (I3CA). Oral I3CA supplementation accelerated EAE development and I3CA concentration in blood samples from persons with MS (PwMS) was associated with increased disease severity mirrored by increased serum neurofilament light chain levels. Altogether, our study shows that microbiota-derived metabolites play a key role in the intestine during neuroinflammation, offering potential therapeutic insights for PwMS.

Human epithelial cell response to bacterial signal indole

Project description:Indole is a bacterial signal secreted by pathogenic and commensal Escherichia coli in the stationary phase at high concentrations (~600 ÂµM). Prior work from our lab has shown that indole decreases E. coli O157:H7 (EHEC) chemotaxis, motility, attachment to epithelial cells and biofilm formation. However, its effect on epithelial cells is not known. We hypothesized that indole induces gene expression changes in epithelial cells that lead to decreased pathogen colonization and infection. Changes in gene expression with the human enterocyte cell line HCT-8 exposed to indole suggested down-regulation of Toll-like receptor signaling and coordinated changes in Jak-STAT and p38 MAPK pathways. Corresponding changes in the expression of cytokines, chemokines, and their receptors also suggested that indole functions as a modulator of inflammation in intestinal epithelial cells. In addition, the expression of genes involved in tight junction organization (claudins, and tight junction proteins) and mucin production were also up-regulated by indole. Keywords: Inter-kingdom signaling interactions between human cells and bacterial signal indole Human intestinal epithelial cells were exposed to 1 mM indole for 4 h or 24 h. RNA was isolated from the control cells and cells exposed to indole for 4 h and 24 h. The experiments were performed in triplicate. A total of 9 microarrays were performed, three for each condition. The gene expression data was analyzed using significance analysis of microarrays (SAM) where a false discovery rate cut-off of 1% was used. The genes were then classified and importnat regulated pathways were reported.

Intrinsic cardiac adrenergic (ICA) cells contribute to LPS-induced myocardial dysfunction

Project description:The role of ICA cells in septic cardiomyopathy is unknown. Here we show that norepinephrine (NE) secretion from ICA cells is increased through activation of Toll-like receptor 4 (TLR4) to aggravate myocardial TNF-α production and dysfunction by lipopolysaccharide (LPS). In ICA cells, LPS activated TLR4-MyD88/TRIF-AP-1 signaling that promoted NE biosynthesis through expression of tyrosine hydroxylase, but did not trigger TNF-α production due to impairment of p65 translocation. Our findings suggest that ICA cells may be a potential therapeutic target for septic cardiomyopathy.
