Gene expression profile at single cell level of colorectal tumors from Col1a2-Cre/ERT2 and Col1a2-Cre/ERT2;TXNDC5fl/fl mice

ABSTRACT: Cancer-associated fibroblasts (CAFs) are reported to remodel the extracellular matrix to modulate immune escape and tumor progression. We performed single-cell RNA sequencing (scRNA-seq) to evaluate whether aberrant expression of TXNDC5 in CAFs modulates the tumor immune system.

Next Generation Sequencing analysis of Hes1+/+ Cre-ERT2 and Hes1fl/fl Cre-ERT2 bone marrow derived macrophages transcriptomes.

Project description:To evaluate the potential role of Hes1 in regulation of type I IFNs and the downstream ISGs expression, we generated the bone marrow-derived macrophages (BMDMs) from Hes1+/+ Cre-ERT2 and Hes1fl/fl Cre-ERT2 mice. RNA-sequencing analysis of Poly(I:C) [(polyinosinic: polycytidylic acid), a mimic of viral double-stranded RNA]-treated BMDMs revealed that 49 genes were induced more than two-fold by Poly(I:C) for 3 hrs in Hes1-deficient BMDMs relative to their expression in Hes1+/+Cre-ERT2 cells. Notably, 67.3% of these upregulated genes were ISGs by screening the Interferome database, suggesting that Hes1 inhibits expression of TLR-mediated ISGs,these results indicated that Hes1 negatively regulates TLR3-mediated induction of ISGs in macrophages.

Single Cell RNA Sequencing of immune cell populations from WT and Cdh5-cre Myct1 fl/fl mice

Project description:We have examined single-cell sequencing using 10x Genomics scRNA-seq to study changes in the immune cell population from tumor-bearing Cdh5-cre Myct1 f/f mice. Previously, we identified that Myct1 is expressed in endothelial cells, and Myct1 deficiency reduces tumor angiogenesis, thereby enhancing the anti-tumor immune environment. This scRNA-seq provides a transcriptomic analysis of immune cells in the tumor microenvironment, showing how tumor angiogenesis affects immune cells in the tumor.

High sensitivity receptor screen and global phosphoproteomics identifies podoplanin as a regulator of cancer associated fibroblast physiology

Project description:Cancer-associated fibroblasts (CAFs) are increasingly recognized as key regulatory cells in the tumor microenvironment, with the ability to engage in crosstalk both with tumor and immune cells. Still, the molecular determinants that orchestrate the interactions between CAFs and infiltrating immune cells remain poorly understood. To elucidate the surface interactome that underlines cell communication, we build an extensive library of single transmembrane (STM) receptors coupled to high avidity cell-based screens, and identify the neutrophil and Treg surface protein CD177 as a novel functional modulator of the mechanoreceptor podoplanin (PDPN). Functionally, CD177 exerts a profound effect on the phosphoproteome of CAFs that results in modulation of actomyosin contractility and energy homeostasis. In human colorectal tumors, CD177+ cells can interact closely with PDPN+ CAFs, indicating that this interaction may have effects on the local tumor environment. Starting from a powerful technology for target discovery, our study defines a molecular bridge between CAFs and immune cells in the tumor microenvironment and reveals PDPN as a central modulator of fibroblast mechanophysiology.

Stiffness-induced cancer-associated fibroblasts are responsible for immunosuppression in a PDGF ligand-dependent manner

Project description:Pancreatic ductal adenocarcinoma (PDAC) is associated with a vast stromal reaction that arises mainly from cancer-associated fibroblasts (CAFs) and promotes both immune escape and tumor growth. Here, we used a mouse model of KrasG12D mutation (KC) without or with deletion of the activin A receptor ALK4 (4KC), which strongly drives collagen deposition that leads to tissue stiffness. To understand this process, single cell transcriptomes were obtained from purified cancer associated fibroblasts (CAFs) and ductal tumor cells from KC and 4KC tissues. These analyses led to the identification of increased number of CAFs/Ducts receptor:ligand interactions under stiffness conditions and a population of stiffness-induced CAFs with a role in immunosuppression.
