Single Nuclei sequencing of mouse model of MBL1 and 2 KD

ABSTRACT: This study looks at a new model of MBL1 and MBL1 KD to understand how these proteins funciton in muscle development and contribute for DM1 pathology. Animals were injected into the tibialis anterior (TA) muscle at 10 weeks of age. Sampling was performed approximately 10 weeks after injection, i.e. at around 20 weeks of age. The left TA was injected with an AAV encoding shmir directed against Mbnl1 and Mbnl2, and the left TA was injected with an AAV encoding shmir directed against LacZ (control). Both muscles were therefore injected with AAV.

Effect of AAV-mediated non-fibrogenic TBX18 expression in mouse hearts

Project description:Gene therapy-based biological pacemakers have been proposed as an alternative to their hardware-based counterparts. In this context, short-term ectopic expression of the T-box transcription factor 18 (TBX18) in the ventricle reportedly generated potent short-term pacemaker function in various animal models. Here, we investigated the effect of adeno-associated virus (AAV)-mediated long-term expression of TBX18 in mouse hearts. Mice were injected with AAV vectors containing GFP-labeled Nanoluciferase or GFP-labbeled TBX18 at the left ventricular apex. Left ventricular tissues were harvest 2 weeks later. The results suggest that at non-fibrogenic level, TBX18 expression suppress working myocardial genes but does not induce pacemaker phenotype.

Gene expression profiling of HDAC3-depleted mouse liver rescued with wild-type exogenous HDAC3

Project description:We have shown that intravenous injection of HDAC3 floxed mice with adeno-associated virus (AAV) expressing Cre depletes hepatic HDAC3, upregulates lipogenic gene expression, and causes fatty liver. When AAV-Flag-HDAC3 wild-type (WT) is co-injected along with AAV-Cre, the exogenous HDAC3 is expressed at endogenous levels and can completely rescue fatty liver phenotype. Here we profile transcriptome of the rescued WT livers in comparison with HDAC3-depleted (KO) livers. 4-months old C57BL/6 male mice were co-injected with AAV-Cre or AAV-Cre plus AAV-Flag-HDAC3. Mice were fed ad libitum and harvested at 5 pm (ZT10) at 2-weeks post-injection. Liver total RNA was extracted and hybridized to Affymetrix Mouse Gene 1.0ST array.

Liver transcriptome analysis of mice with ASirs2 suppression

Project description:Purpose: We show that the Irs2 locus, a critical regulator of insulin actions, encodes an antisense transcript ASIrs2, whose expression increased after refeeding in liver, reciprocally to that of Irs2. In order to elucidate the functions of ASIrs2, the liver transcriptome was analyzed under ASIrs2 suppression in wild type mice after refeeding. Methods: C57BL/6 mice were injected with AAV encoding shRNA for ASIrs2 via the via tail vein at 8 weeks of age, and the mice were sacrificed for organ collection after overnight fasting and subsequently 6 hours of refeeding on the 27th or 28th day after AAV injection.

Project description:The experiment evaluates the therapeutic effect brain injected AAV9-IDS (Adeno-associated virus 9 encoding IDS enzyme) treatment in a mouse model of Mucopolysaccharidosis Type II (MPSII). Microarrays were performed in order to compare the transcriptional profiling after the treatment. Three groups were analyzed, wild type (WT) mice, MPSII mice treated with AAV-NULL (AAV vector alone) and MPSII mice treated with AAV-IDS (vector encoding IDS enzyme). Four months after vector administration (at 6 months of age), animals were sacrificed and tissues were harvested and processed. RNA isolated from the encephalon of three groups of mice was analysed using the Affimetrix® microarray platform

Transcriptomic analysis of hearts from high fat diet fed mice with over expression or knock down of microRNA-322

Project description:Cardiac function of mice fed with a 60%fat diet (high fat diet, HFD) for 16 weeks was analyzed. Hearts showed an alteration in ejection fraction and an increase in left ventricular en systolic diameter. However mice injected with an AAV overexpressing microRNA-322 (miR-322) were protected: ejection fraction was less decreased and LVESD less increased than in animals treated with an AAV control or an AAVsponge anti miR-322 (which decrease miR-322 availability and action in the heart). To better understand globla changes induced by miR-322 modulation in the heart, a transcriptomic analysis was performed to compare gene expression in three hearts from HFD+AAVmiR-322 mice (showing cardiac protection) and gene expression in three hearts from HFD+AAvsponge (were miR-322 action is decreased and showing the most altered cardiac function).

Protein propionylation in Pcca deficiency

Project description:To determine the prevalence and identify any enrichment of lysine propionylation in PCC deficiency, an analysis comparing the propionylated liver proteins of control and liver-specific Pcca knockout mice was performed. Liver-specific knockout of PCC activity was induced in mice with floxed axons for the Pcca gene (Pcca ff) at 5 weeks of age using iCre-AAV with a TBG promoter. For controls, Pcca ff mice were injected with eGFP-AAV under the same promoter.

Sex specific role of KDM5 demethylases in Alcohol-associated Liver Disease

Project description:Alcohol-associated liver disease (ALD) is a major cause of alcohol related mortality. The specific mechanisms responsible for ALD development and progression are not fully understood, and there is limited therapy for any stage of ALD. Sex differences are often disregarded in in genetic and mechanistic studies. We aimed to take an unbiased approach to define sex specific pathways in livers exposed to alcohol. METHODS: Mice were fed LieberDeCarli alcohol liquid diet for 3 weeks. To identify the role of Kdm5b and Kdm5c we injected mice with AAV-shControl at 2x10^11 gc per mouse or AAV-shKdm5b and AAV-Kdm5c vectors at 10^11 gc per mouse. To assess the role of AhR we injected mice with AAV-shControl or AAV-shAhR vectors at 10^11 gc per mouse. RESULTS: We found several pathways affected by alcohol in sex specific way and identified KDM5 demethylases as contributors to that specificity.

Single-cell RNA transcriptome shows IFITM1 regulates stem cell activation in limbal epithelium in response to alkali burn.

Project description:To examine whether the upregulation of Ifitm1 is essential for the expansion of stem/early TA cells in response to corneal injury, wild type mouse eyes were topically treated with AAV-shRNA-Ifitm1 or AAV-empty vector (control) and subsequently were subjected to corneal NaOH burn. AAV-shRNA-Ifitm1 treatment downregulated Ifitm1 expression in mouse limbal epithelium . A scRNA-seq assay was conducted using cells isolated from corneal/limbal tissues at three days after corneas were injured. scRNA-seq assay identified stem/early TA cell population using Gpha2, a limbal epithelial stem /eTA cell marker. In AAV-EV treated mouse eyes, NaOH injury increased the numbers of the stem/eTA cells compared to uninjured eyes. Such expansion of stem/eTA cell population following corneal injury was markedly reduced in AAV-shRNA-Ifitm1 treated eyes. This suggests a positive role of Ifitm1 in stem/early TA cell expansion after corneal injury.
