AFF4 regulates cellular adipogenic differentiation via targeting autophagy

Project description:AFF4 was discovered as a scaffold protein of Super Elongation Complex (SEC) and exhibites an essential function on osteogenesis, tumorigenesis and odontogenesis. However, the adipogenic biological functions of AFF4 need further explorations. Here, we demonstrate that knockdown of AFF4 inhibits adipogenesis in both human mesenchymal stem cells (hMSCs) and lineage-committed 3T3-L1 preadipocytes. Conditional knocking down of Aff4 in transgenic mouse presents a thin body phenotype and impeded adipogenesis. Mechanistically, we define autophagy signaling as a crucial downstream of AFF4 through the combination of RNA-seq and ChIP-seq analyses. Depletion of AFF4 mediates the transcription of crutial autophagy genes ATG5 and ATG16L1. Simultaneous overexpression of ATG5 and ATG16L1 rescues the lineage commitment of AFF4-deficient cells. Our data elucidate that AFF4 is indispensable for adipogenic differentiation and reveal a novel mechanism for adipogenesis at transcriptional level.

Identification of Nrf2-regulated genes in A549 lung cancer cells by oligonucleotide microarray

Project description:To elucidate the mechanisms by which Nrf2 regulates cell growth, we performed global gene expression profiling of A549 lung cancer cells with knockdown of Nrf2. Gene networks associated with carbohydrate metabolism and drug metabolism were significantly downregulated in Nrf2-depleted A549 cells. Gene Set Enrichment Analysis revealed significant enrichment of genes associated with carbohydrate catabolic processes, positive regulation of metabolic processes, PPP, and arachidonic acid metabolism. In summary, this analysis revealed that Nrf2 positively regulates transcription of genes that play key roles in central carbon metabolism. A549 cells were transfected with non targeting NS siRNA or siRNA targeting Nrf2. Mock transfected A549 cells were treated with transfection reagent alone. We had 3 biological replicates for each of the 3 groups. Ninty six hours post transfection, cells were lysed and total RNA was isolated.

GAPDH inhibition in A549 and H358 lung cancer cells

Project description:Human lung cancer cell lines A549 and H358 were obtained from (Biobank of Medical University of Graz) and cultured in RPMI 1640 (R0883; Sigma) supplemented with 10% fetal bovine serum (FBS; Gibco), 5 mM glutamine and 5 mM Pen Step, and sub-cultured regularly. For siRNA mediated knock-down of GAPDH, 150,000 A549 and H358 cells were seeded in triplicates of a six-well plate and transfected upon reaching ca. 60% confluency with silencerTM siRNA kit targeting GAPDH or non-target control (AM4605; Thermo), using RNAiMax (Thermo) and according to the manufacturer’s instructions for 6-well plate scale. For GAPDH inhibition, 150.000 A549 and H358 cells were seeded in six replicates per condition, then treated with 10 µM koningic acid (KA) or vehicle control (dmso). 48 h post transfection or 24 h post KA treatment, cells were harvested in 500 µl of harvesting solution (80% methanol in 50 mM ammonium acetate with 2.5 mM N-ethylmaleimide) and processed according to one-pot redox approach.

MicroRNA expression data from Bcl-xL silenced A549 lung adenocarcinoma cells

Project description:Bcl-xL is an anti-apoptotic protein that is frequently found to be overexpressed in non-small cell lung cancer leading to an inhibition of apoptosis and poor prognosis. Recently, the role of miRNAs in regulating apoptosis and cell survival during tumorigenesis has become evident, with cancer cells showing perturbed expression of various miRNAs. We utilized miRNA microarrays to determine if miRNA dysregulation in bcl-xL silenced lung adenocarcinoma cells could be involved in regulating apoptotic behavior, and identified dysregulated miRNAs with putative targets involved in signal transduction pathways regulating apoptosis, cell proliferation and cell progression. Short interfering RNA-based transfection of A549 was carried out inducing a reduction in bcl-xL expression levels. 24 hours post-transfection total RNA was isolated using TRIzol reagent and hybridized onto Affymetrix GeneChip miRNA Arrays. A global miRNA expression profile was then established, which compared total RNA, extracted from siRNA-transfected and non-transfected A549 cells. All experiments were carried out with three independent biological replicates.

Comparison of gene expression profiles of A549 cell treated with NRF2 siRNA and control siRNA

Project description:To examine the role of NRF2 in accelerating cell proliferation and to identify the target genes responsible for this function, transcriptome analysis was performed using A549 cells, in which NRF2 is constitutively activated. NRF2 was knocked down by siRNA against NRF2, and the gene expression profile was compared with that of A549 cells treated with control siRNA. To exclude off-target effects, three different siRNAs against NRF2 was independently applied. NRF2 siRNA or control siRNA was transfected into A549 cells. Cells were harvested 24 hours after transfection, and total RNA was purified.
