Project description:Murine pancreatic cancer cells (HY15549 cells) established from genetically engineered mouse models (GEMM) of PDAC (p48-Cre+, KrasLSL-G12D/+, Trp53lox/+; KPC mice) were transfected with the GFP-LC3-RFP autophagy flux reporter from Mizushima lab, wherein reduction in the GFP/RFP ratio indicates increase in autophagy flux. Organoids derived from these cells were dissociated into single cells and sorted into autophagy-high (AThi) and autophagy-low (ATlo) populations according to GFP/RFP signal ratio. To validate this reporter, RNA was extracted from these two populations and expression of autophagy/lysosome related genes was examined.

Project description:Small cell lung cancer (SCLC) is characterized by significant heterogeneity and plasticity, driving its aggressive progression and resistance to therapy. Understanding the underlying mechanisms of these features is crucial for improving treatment outcomes. Autophagy, a conserved cellular process, is involved in many cancers, but its role in SCLC remains unclear. Using a genetically engineered mouse model (Rb1fl/fl;Trp53fl/fl;GFP-LC3-RFP-LC3△G), we tracked autophagic flux in vivo to assess its effects on SCLC biology. Tumor subpopulations with high autophagic flux exhibited increased proliferation, enhanced metastatic potential, and neuroendocrine (NE) characteristics, whereas subpopulations with low autophagic flux exhibited more immune-related signals and non-NE traits.

Monitoring Autophagic Flux in vivo Revealed its Physiological Response and Significance of Heterogeneity in Pancreatic Beta Cells

Project description:Autophagy plays an important role in preserving cellular homeostasis in pancreatic beta cells. However, the extent of autophagic flux induced in various physiological settings in vivo is unclear. In this study, we generated transgenic mice expressing pHluorin-LC3-mCherry reporter for monitoring systemic autophagic flux. Our findings revealed that autophagic flux in pancreatic islets enhanced after starvation, although suppression of the flux after short-term refeeding needs more prolonged restarvation in islets than in liver and skeletal muscle. Furthermore, heterogeneity of autophagic flux in beta cells manifested after increasing insulin resistance and intracellular calcium influx by glucose stimulation increased more in high- than low-flux beta cells, with differential gene expression based on the flux. Thus, our monitor mouse enables us to reveal physiological response and biological insight of heterogeneity in autophagic flux in pancreatic beta cells.

KNSTRN knockdown impairs autophagy flux and inhibits bladder cancer progression through ROS- mediated lysosomal dysfunction

Project description:Bladder cancer (BLCA) is a prevalent cancer affecting the urinary tract, the mitochondria-localized Astrin-binding protein (KNSTRN) has been shown to be strongly associated with the development of several cancers. In addition, abnormal levels of autophagy have been shown to have a dramatic impact on tumor development. However, the potential mechanisms of BLCA autophagy regulation by KNSTRN remain poorly understood. In this research, we found that knockdown of KNSTRN inhibited the autophagic flux of BLCA. Mechanistically, knockdown of KNSTRN leads to reactive oxygen species (ROS)-dependent disruption of the lysosomal acidic environment and reduction of protease activity, hindering autophagosome-lysosome fusion. Clioquinol reactivates lysosomal activity by normalizing lysosomal pH, which in turn reinstates autophagic flux. Intracellular ROS production is also critical for this process, as ROS scavengers (NAC) reverse lysosomal dysfunction and reactivate autophagic flux. Additionally, the autophagy activator rapamycin (Rapa) effectively protected against KNSTRN knockdown-induced cell death in vitro and in vivo. Thus, we demonstrate that knockdown of KNSTRN leads to intracellular ROS accumulation and lysosomal dysfunction, which impairs autophagic flux and inhibits BLCA progression.

Gene expression of non-apoptotic pancreatic acinar cell death with autophagy

Project description:Serine protease inhibitor Kazal type 3 (Spink3) is a trypsin inhibitor in the pancreas. Spink3-/- pancreatic acinar cells are dead with excessive autophagy at birth (p0.5). To prove the role of nonapoptotic cell death with autophagy, we generated by transgenic technology the pancreas of Spink3-/-;XKI/+ mice contained both normal and dying acinar cells with autophagy. In this new mouse model, chronic inflammation occurred in the pancreas, indicating that some signals from nonapoptotic dead cell induce chronic inflammation in the pancreas. All samples were the pancreas at p0.5. Sample 1 and 2 are the pancreas from wild type (Spink3+/+, control) mice. Sample 3 and 4 are the pancreas from Spink3-/-, which all pancreatic acinar cells show induced nonapoptotic cell death with autophagy. Sample 5 and 6 are the pancreas from Spink3-/-XKI/+, about half acinar cells are normal, but other acinar cells show induced nonapoptotic cell death with autophagy.

Transcriptional profiling of autophagy proficient and compromised cell lines, determined by expression levels of Atg5

Project description:Autophagy is activated in pancreatic ductal adenocarcinoma (PDAC) and is currently being considered a promising therapeutic target in clinical trials. PDAC is a highly lethal disease with incidence rate equalling mortality rate. Main reasons for PDAC lethality are late-stage diagnosis, high agressiveness and metastatic rate, lack of effective treatments as well as specific diagnostic markers. Here we show that varying levels of the Autophagy related gene 5 (Atg5) determine pancreatic tumor formation and malignancy. While homozygous deletion of Atg5 blocks tumor progression in an in vivo model of PDAC, heterozygous deletion increases tumor aggressiveness and metastasis. Further analyses reveal that monoallelic loss of Atg5 affects mitochondrial homeostasis, changes intracellular calcium oscillations, heightens extracellular cathepsin activities , and promotes a pro-tumorigenic inflammatory microenvironment collectively enhancing tumor cell migration, invasion, and metastasis. Future treatments should take into account that variations in the autophagy pathway may have opposing effects on pancreatic tumor load, especially considering the multitude of autophagy inhibitors currently tested in clinical trials.
