H4K16ac Cut&Tag profiling of Bone Marrow Derived macrophage stimulated with R848

ABSTRACT: Purpose: Genome-wide identification of genes with H4K16ac, we performed Cleavage Under Targets and Tagmentation (CUT&Tag) analysis of wild-type (WT)and KAT8-deficient BMDMs. Methods: Mouse BMDMs were generated from bone marrow cells in RPMI-1640 medium with recombinant mouse M-CSF (20ng/ml). BMDMs were stained to confirm the surface expression of CD11b and F4/80. Cells with purity >97.5% were used for subsequent experiments. BMDMs were stimulated with R848 (100ng/ml) for 2 hours. Results: We carried out a CUT&Tag assay using antibodies against H4K16ac（CST）, and found the global influence of H4K16ac in regulating genes of transcription regulatory regions of key genes for psoriasis.

Project description:Purpose: The purpose of this study is to detect activated or silenced genes during wild type (WT) and KAT8-deficient bone marrow derived macrophages (BMDMs). Gene expression differences between two samples could be found using transcriptome profiling (RNA-seq) analysis. Methods: Mouse BMDMs were generated from bone marrow cells in RPMI-1640 medium with recombinant mouse M-CSF (20ng/ml). BMDMs were stained to confirm the surface expression of CD11b and F4/80. Cells with purity >97.5% were used for subsequent experiments. WT and KAT8-deficient BMDMs were stimulated with R848 (100ng/ml) for 0 and 2 hours, of which RNA profiles were generated by deep sequencing, using Illumina. Results: We mapped about 10 million sequence reads per sample to the mouse genome, identified hundreds of genes with significant mRNA between WT and KAT8-deficient BMDMs.

BMDMs were stained to confirm the surface expression of CD11b and F4/80,and the use of BMDMs with purity greater than 97.5% for subsequent experiments.BMDMs pretreated with DMSO or KAT8 inhibitor were stimulated with R848 (100ng/ml) for 2 hours, of which RNA profiles were generated by deep sequencing, using Illumina. Results: We mapped about 10 million sequence reads per sample to the mouse genome, identified hundreds of genes with significant mRNA between DMSO or KAT8 inhibitor-treated BMDMs.

KAT8-Low Osteosarcoma suppresses Cisplatin Sensitivity via H4K16ac-Mediated Cell Apoptosis

Project description:Cisplatin (DDP), a core chemotherapeutic agent for osteosarcoma (OS), induces DNA cross-linking to cause damage, yet the underlying regulatory mechanisms remain elusive. Previous studies have revealed that in DDP-resistant osteosarcoma (OS) cells, both the expression level of the lysine acetyltransferase KAT8 and its mediated site-specific acetylation of histone H4K16ac are significantly decreased. Here, using CUT&Tag technology and proteomic analysis, we demonstrated that the KAT8–H4K16ac axis modulates dynamic histone acetylation to epigenetically regulate DNA repair pathways. The biological inactivation of KAT8 results in a targeted decrease in the epigenetic mark H4K16ac at the promoter regions of genes associated with DNA repair, leading to diminished chromatin accessibility and inhibition of the p53 signaling pathway. Consequently, the DNA damage response is compromised, promoting cisplatin resistance in OS. In contrast, targeted restoration of H4K16ac reverses chemotherapy resistance through the reinstatement of chromatin dynamics and transcriptional activation of DNA repair programs. This hypothesis emphasizes the need to clarify the molecular mechanisms and signaling pathways associated with KAT8-H4K16ac, especially in relation to apoptotic pathways, which could offer new insights and justifications for treatment strategies in osteosarcoma.
