LY6H is a specific marker of human pancreatic delta cells

ABSTRACT: The identification of cell surface markers specific to pancreatic islet cell subsets is important for both the study of islet biology and for investigating the pathophysiology of diseases in which these cell types are lost or damaged. Analysis of publicly available single-cell RNAseq data identified expression of LY6H specifically in the delta cells of the pancreas. This finding was confirmed using immunofluorescence analysis of histological sections of human pancreas, and flow cytometric analysis of human islet preparations. We found that expression of LY6H was restricted to pancreatic delta cells in both control and diabetic donors. Furthermore, we demonstrate that antibodies against LY6H can be used to isolate live delta cell populations from preparations of human islets. This study identified LY6H as a novel cell surface marker of human pancreatic delta cells – a finding that will aid in the identification and characterisation of this important cell type.

Integrin avb5 heterodimer is a specific marker of human pancreatic beta cells

Project description:The identification of cell surface markers specific to pancreatic beta cells is important for both the study of islet biology and for investigating the pathophysiology of diseases in which this cell type is lost or damaged. Following analysis of publicly available single-cell RNAseq data, we identified specific integrin subunits, integrin av and integrin b5, that were expressed in beta cells. Flow cytometry analysis showed that the vast majority of islet cells expressed some level of both the av and b5 subunits. Using our analysis of single-cell sequencing data as a guide, we isolated populations expressing both subunits from isolated human islets and subjected these to bulk RNAseq analysis. This analysis suggested that the avlob5lo population was enriched for endocrine cell types including alpha, delta and gamma cells, whilst the avhib5hi population was enriched for pericytes and stellate cells . Interestingly, the av+b5neg reference population appeared to be enriched for myeloid cells. This finding was further elaborated using immunofluorescence analysis of histological sections derived from donor human pancreas. Despite the broad expression of specific integrin subunits, we found that expression of integrin avb5 heterodimers was restricted to beta cells and that this complex persisted in islet remnants of some type 1 diabetic individuals from which insulin expression had been lost. This study identifies avb5 heterodimers as a novel cell surface marker of human pancreatic beta cells, a finding that will aid in the identification and characterisation of this important cell type.

Identification of genes enriched in putative stem/progenitor cells from mouse embryonic pancreas

Project description:Identification of genes enriched in putative stem/progenitor cells (CD133highPDGFRb- cell population) from the mouse embryonic pancreas that are purified by fluorescence activated cell sorting (FACS). Success in islet transplantation-based therapies for type 1 diabetes mellitus and an extreme shortage of pancreatic islets has motivated efforts to develop renewable sources of islet-replacement tissue. Only a few attempts have been made at prospective isolation of pancreatic stem/progenitor cells, due to the lack of specific markers and the development of cell culture method. This study demonstrates the isolation of pancreatic stem/progenitor cells from the embryonic pancreas by cell sorting. RT-PCR and microarray analysis demonstrated that pancreatic stem/progenitor cells are enriched in CD133highPDGFRb- cell population. During in vivo differentiation, these cell populations have the ability for self-renewal and multipotency, including the formation of insulin-producing cells. Since the strategy is based on the cell sorting using cell surface markers common to human and rodents, it may promote strategies to derive transplantable islet-replacement tissues from human pancreatic stem/progenitor cells. Gene expression profiling of a mouse model of islet dysmorphogenesis

Project description:In the past decade, several transcription factors critical for pancreas development have been identified. Despite this success, many of the cell surface and extracellular factors necessary for proper islet morphogenesis and function remain uncharacterized. Previous studies have shown that transgenic over-expression of the transcription factor HNF6 specifically in the pancreatic endocrine cell lineage resulted in the disruption of islet morphogenesis, including dysfunctional endocrine cell sorting, increased islet size, and failure of islets to migrate away from the ductal epithelium. We exploited the dysmorphic islets in pdx1PBHnf6 animals as a tool to identify factors important for islet morphogenesis. Genome-wide microarray analysis was used to identify differences in the gene expression profiles of late gestation and early postnatal pancreas tissue from wild type and pdx1PBHnf6 animals. We report the identification of genes with an altered expression in HNF6 Tg animals and highlight factors with potential importance in islet morphogenesis. Whole Exome Sequencing of Pancreatic Neuroendocrine Tumors

Project description:<p>Pancreatic neuroendocrine tumors (PNETs), often referred to as "islet cell tumors", are neuroendocrine neoplasms that arise from cells of the endocrine and nervous system within the pancreas. These rare tumors which originate from the pancreatic islet are divided into many categories and are often classified by the hormone most strongly secreted. With the collaboration of the Elkins Pancreatic Center, the Human Genome Sequencing Center (HGSC) at Baylor College of Medicine had access to approximately 30 untreated tumor specimens and matched normal blood samples. Since the onset and progression of cancer is driven by extensive mutation of the genome, we are combining enrichment of exonic DNA with next generation sequencing to detect and characterize the somatic mutation profile of patients with pancreatic neuroendocrine cancer.</p> <p>This work was done at the Human Genome Sequencing Center (HGSC) in collaboration with the Elkins Pancreatic Center at Baylor College of Medicine in Houston, TX and was supported by grant number 5U54HG003273 from National Human Genome Research Institute (NHGRI).</p>

Regulated expression and function of the GABAB receptor in human pancreatic beta cell line and islets

Project description:G protein-coupled receptors (GPCRs) are seven transmembrane signaling molecules that are involved in a wide variety of physiological processes. They constitute a large protein family of receptors with almost 300 members detected in human pancreatic islet preparations. However, the functional role of these receptors in pancreatic islets is unknown in most cases. We generated a new stable human beta cell line from neonatal pancreas. This cell line, named ECN90 expresses both subunits (GABBR1 and GABBR2) of the metabotropic GABAB receptor compared to human islet. In ECN90 cells, baclofen, a specific GABAB receptor agonist, inhibits cAMP signaling causing decreased expression of beta cell-specific genes such as MAFA and PCSK1, and reduced insulin secretion. We next demonstrated that in primary human islets, GABBR2 mRNA expression is strongly induced under cAMP signaling, while GABBR1 mRNA is constitutively expressed. We also found that induction and activation of the GABAB receptor in human islets modulates insulin secretion.

NTPDase3 antibody targets adult human pancreatic β-cells for in vitro and in vivo analysis

Project description:Identification of cell surface markers specific to human pancreatic β-cells would allow in vivo analysis and imaging. Here we introduce a biomarker – ectonucleoside triphosphate diphosphohydrolase-3 (NTPDase3) – that is expressed on the cell surface of essentially all adult human β-cells, including those from individuals with type 1 or type 2 diabetes (T1D, T2D). NTPDase3 is expressed dynamically during postnatal human pancreas development, appearing first in acinar cells at birth, but several months later its expression declines in acinar cells while concurrently emerges in islet β-cells. Given its specificity and membrane localization, we utilized an NTPDase3 antibody for purification of live human β-cells as confirmed by transcriptional profiling, and, in addition, for in vivo imaging of transplanted human β-cells. Thus, NTPDase3 is a cell surface biomarker of adult human β-cells and the antibody directed to this protein should be a useful new reagent for β-cell sorting, in vivo imaging, and targeting.
