Project description:Human granulosa cells are follicular cells surrounding the oocyte. Human granulosa cells are retrieved during in vitro fertilization a process where patients undergo hormonal stimulation including FSH and LH/hCG stimulation. Under the influence of the luteinizing hormone (LH) a process called luteinization they differentiate to luteal cells and contribute to the corpus luteum. Therefore, this cellular system is a good model for human corpus luteum (CL). To study processes within the human CL, IVF-derived GCs from patients were cultured for two to five days and then analyzed with mass spectrometry based shotgun proteomics.

Project description:Administration of GnRH antagonist in IVF has several advantages. However, studies have shown that GnRH antagonist protocol cycles have lower implantation and clinical pregnancy rates than GnRH agonist long protocol cycles. Endometrial receptivity rather than embryo quality is thought to account for this phenomenon, however the mechanism is still largely unknown. This microarray analysed human endometrium during ‘implantation window phase’ between three groups: 1) untreated control, 2) GnRH agonist long protocol, and 3) GnRH antagonist protocol. The differential gene between groups uncovered the molecular mechanism occurred in endometrium affected by the intake of GnRH agonist or GnRH antagonist. Further study of these differential gene promises to find the reasons for lower endometrial receptivity in patients treated with GnRH antagonist and to improve the implantation rate in GnRH antagonist protocols in IVF.

Project description:Premature progesterone (P) rise during GnRH antagonist cycles for IVF is a frequent phenomenon and has been associated with lower pregnancy and implantation rates. Different thresholds of progesterone have been used so far to define its premature rise during the follicular phase of an IVF stimulated cycle. In this study, we evaluated endometrial gene expression on the day of oocyte retrieval according to the level of serum progesterone on the day of hCG administration in GnRH antagonist cycles.Endometrial biopsies from eleven patients were taken with a Pipelle de Cornier (Prodimed, Neuilly-en-Thelle, France) on the day of oocyte retrieval in a GnRH antagonist/rec-FSH stimulated IVF cycle with fresh embryo transfer. Biopsies were analysed for gene expression with Affymetrix Human Genome (HG) U133 Plus 2.0 Arrays and GCOS software (Affymetrix, Santa Clara, CA, USA). Patients were divided into three different groups according to their progesterone serum concentration on the day of hCG administration (A) P <= 0.9 ng/mL, (B) 1 < P < 1.5 ng/mL, and (C) P > 1.5 ng/mL. Serum P was measured with the automated Elecsys immunoanalyser (Roche Diagnostics, Mannheim, Germany). Selected differentially expressed genes were validated with quantitative real-time PCR (QPCR) with TaqMan Gene Expression Assays (Applied Biosystems, Foster City, CA, USA). Project description:BACKGROUND: Molecular signaling in in vitro fertilization procedures seems to play a crucial role for embryonic development. In a prospective blinded cohort study, we investigated the role of microRNAs (miRNA) and extracellular vesicles (EV) as potential carriers of miRNAs in embryonic culture media. METHODS: Embryonic culture media from 56 female partners undergoing infertility treatment. miRNA microarray and RT-qPCR assays in combination with bioinformatics were used for the screening, validation and analysis of identified miRNAs. Nanoparticle tracking analysis was applied to measure EV in culture media. RESULTS: The overall miRNA repertoire in culture media was astonishingly complex: 621 miRNAs were present in embryonic culture media. Samples with positive outcome had an overall decreased miRNA repertoire, with the largest effects for miR-29c-3p. RT-qPCR measurements were largely consistent with the microarray results. Comparison to human spermatozoa profiles highlighted that a large part of spermatozoa miRNAs (101 miRNAs) are also present in culture media following embryonic transfer. As potential carrier molecules that could be of relevance for the decreased miRNA repertoire in samples with positive outcome we investigated EV in a small proof-of-concept study. While the size distribution was similar between both cohorts, the total number of EV in pure culture media was zero, in culture media of patients with positive outcome 3.8 billion per ml and in culture media of samples with negative outcome 7.35 billion per ml, correlating well with the results for the miRNAs. CONCLUSIONS: Embryonic culture media of pre-implantation embryos contains a stable population of highly specific to ubiquitous miRNAs that are correlated to embryogenesis and spermatogenesis processes. These miRNAs are potentially harbored by EV.

Project description:The objective of the study was to compare the microRNA content in uterine fluid from patients with recurrent implantation failure (RIF) to that of healthy fertile women. It is a descriptive laboratory study including healthy fertile women and patients with RIF, defined as three failed in vitro fertilization cycles with high quality embryos. Study subjects were instructed to monitor their menstrual cycles using a luteinizing hormone test kit. Uterine fluid was collected on day LH+ 7-9 by flushing with saline. Samples were processed for small RNA sequencing and results were analysed using bioinformatics. The main outcome measure was to identify differentially expressed miRNAs between patients with RIF and healthy fertile women.
