ABSTRACT: Streptococcus pyogenes (group A Streptococcus, GAS) responds to environmental changes in a manner that results in an adaptive regulation of the transcriptome. Global transcriptional regulators are able to integrate important extracellular and intracellular information and are responsible for modulation of the transcriptional network. The roles of several global transcriptional regulators in adaptation and virulence gene expression have been described. In this study we used microarray to investigate the regulatory roles of CodY and CovRS played in Streptococcus pyogenes. keywords: genetic modification

Transcriptomic comparison of serotype M1 S. pyogenes

Project description:S. pyogenes strains were compared with the intact covRS form of the globally disseminated M1T1 clone to track transcriptomic changes engendered during the emergence of the M1T1 clone. The mutant covRS form of the M1T1 clone was included as a transcriptomic outlier and to provide a context for the magnitude of transcriptional shifts detected within the isolate set examined. Microarray was performed on RNA extracted from mid-logarithmic phase S. pyogenes grown in Todd-Hewitt with 1% yeast extract in vitro. Experiments were performed using a single color method. Each sample was labelled with Cy3 and hybridized to separate arrays. Each strain was analysed in 3 biological replicates. cDNA hybridized to JCVI PFGRC Streptococcus pyogenes v2 oligo arrays. Only probed representing the core M1 genome were used for analysis.

RNA-seq to investigate differential expression of serotype emm1 Streptococcus pyogenes strains S119 and S126

Project description:The human pathogen Streptococcus pyogenes, or group A streptococcus, is responsible for mild infections to life-threatening diseases. We previously have performed the transcription start site profiling of a Streptococcus pyogenes emm1 strain, strain S119, an invasive strain isolated from a blood culture. Here, we perform strand-specific RNA-seq experiments to complete this characterization and analyze the global coverage and the differential expression in growth medium complemented or not with 15 mM MgCl2. In addition we compare these results to those obtained with a related strain, strain S126, corresponding to a colonization sample, that differs from S119 by only one mutation in the two-component regulator of virulence CovRS.

Dual regulation of nitrogen metabolism by CodY and GlnR in Streptococcus mutans

Project description:CodY and GlnR are two major transcriptional regulators in nitrogen metabolism in Gram-positive bacteria. CodY regulates genes involved in the adaptive response to poor growth conditions, especially to nutrient limitation. GlnR controls nitrogen utilization according to the availability of nitrogen source. In this study we used microarray to investigate the regulatory roles that CodY and GlnR play in nitrogen metabolism in Streptococcus mutans. Streptococcus mutans UA159 wild-type cells, ΔcodY, ΔglnR, and ΔcodYglnR strains were grown in a chemically-defined medium until the mid-log phase. The nitrogen source was 1% tryptone. Twenty millimolar sodium thiosulfate was added to the medium to support cysteine biosynthesis. The transcriptional profile of the whole genome was examined with microarray.
