A single cell epigenomic atlas of adjuvant-mediated innate immune responses [scRNA-seq]


ABSTRACT: The contribution of adjuvants to long-term development of innate immune responses remains understudied. We constructed a time-course single-cell chromatin accessibility and transcriptomic atlas of bone marrow (BM) progenitors and innate immune cells in humanized NOG (hNOG) mice following intramuscular injection of the highly effective AS01 adjuvant and compared it with intravenous live BCG (Bacille-Calmette Guérin) vaccine administration. We find that AS01 induced persistent and durable epigenetic changes in myeloid cells and their progenitors with greater accessibility at antigen-presenting genes and type 1 interferon signaling pathway genes. Furthermore, monocytes and macrophages from the lungs of AS01-educated mice elicited a trained immunity phenotype, demonstrating enhanced heterolozygous protection against Mycobacterium infection. These striking data suggest the potency of the AS01 adjuvant emerges from its ability to trigger two key epigenetic functions, the induction of aspects of trained immunity and the ability to target key loci in the induction of adaptive immune responses.
