RNA-seq analysis of kidney tissues in KMN and KM mice

ABSTRACT: To explore the possible role of Nsd2 in kidney cancer, the gene expression profile of kisney tissues from KMN (Ksp-Cre; Myc OE/+; Nsd2 OE/+) and littermate KM (Ksp-Cre; Myc OE/+) mice was examined by RNA sequencing(RNA-Seq).

Project description:To explore a possible role of SETD2 in kidney development and tumorigenesis in vivo, we generated Setd2-floxed mice and deleted the Setd2 gene in tubular epithelial cells using a transgenic Ksp1.3/Cre mouse line. Then, we established our own c-MYC-transgene mouse line by overexpressing the human c-MYC under the control of the Ksp promoter to generate kidney disease mouse model. To get an insight into the mechanism of how SETD2 ablation promotes ccRCC formation in a c-MYC-generated PKD model, we performed RNA-seq using the renal tubules isolated from Wild Type, Setd2-KO, MYC-OE and MYC-OE; Setd2-KO mice.

Project description:To explore potential role of SETD2 in PKD-ccRCC transition in vivo, we generated a PKD mouse model by overexpressing oncogene c-MYC under the control of the Ksp promoter (KM mice), and knockout Setd2 gene using the same Ksp promoter in this PKD mice (KMs mice) to establish a ccRCC mouse model. To further characterize biological responses trigered by SETD2 deletion during PKD-ccRCC transition at the mRNA level, we next conducted global transcriptomics studies.

Gene expression profile of KM-H2 cells conditional expressing PU.1

Project description:PU.1 is an Ets family transcription factor that is essential for the differentiation of both myeloid and lymphoid cells. PU.1 is down-regulated in classical Hodgkin lymphoma cells via methylation of the PU.1 promoter. To evaluate whether down-regulation of PU.1 is essential for the growth of cHL cells, we generated KM-H2 derived cell lines conditionally express PU.1 by tet-off system (designated KM-H2tetPU.1). Conditonally expressed PU.1 by tetracycline removal induced complete growth arrest and apoptosis in KM-H2 cells. To elucidate the mechanisms underlying cell cycle arrest and apoptosis induced by PU.1, we compared gene expression profiles of KM-H2tetPU.1 cells 0, 1 and 3 days after PU.1 induction, by DNA microarray.

Transcriptome analysis of KM-H2 cells with different Rhodamin 123 eflux capacity

Project description:Rhodamin 123 is a dye which can be used to detect the activity of ABC transporters. We observed that after staining of KM-H2 Hodgkin lymphoma cells with Rhodamin 123, part of the cells rapidly eliminated the dye, while another part of the cells retained the dye for a longer time. We compared the transcriptome of KM-H2 Hodgkin lymphoma cells with high Rhodamin 123 efflux capacity and KM-H2 cells with low Rhodamin 123 efflux capacity.
