Gene expression profiling of cidofovir treatment and resistance in SiHa cells

ABSTRACT: Cidofovir is an acyclic nucleoside phosphonate with strong antiviral activity against a broad spectrum of DNA viruses. Although it has previously been shown that cidofovir exerts an antiproliferative effect on HPV positive cells by the induction of apoptosis, the exact mechanism of action remains to be unraveled. In order to study the activity of cidofovir against HPV, gene expression profiling was performed in cidofovir-treated and cidofovir-resistant SiHa cells by means of microarrays (HG-U133 Plus 2, Affymetrix).

In order to study the activity of cidofovir against HPV, gene expression profiling was performed in cidofovir-treated and cidofovir-resistant HeLa, HaCaT, and PHK cells by means of microarrays (HG-U133 Plus 2, Affymetrix). Controls (untreated HeLa, HaCaT, and PHK wild-type cells): 72h of growth. Cidofovir treated (50 M-BM-5g/ml) HeLa, HaCaT, and PHK cells: 72h of growth. Cidofovir resistant cells (HeLa #52 and HaCaT #51): 72h of growth. All samples were performed in triplicates.

Altered global gene profile in KSHV-infected siHa cells

Project description:The Kaposi sarcoma-associated herpesvirus (KSHV) is the causative agent of Kaposi sarcoma (KS), the most common HIV/AIDS-associated tumor worldwide. Transmission routes of KSHV in the general population are poorly understood. Whereas sexual transmission appears to be common in homosexual men, the evidence for heterosexual transmission is less convincing. In fact, KSHVDNA sequences have been detected in the prostate, semen, and in the female genital tract. Persistent infection with high-risk human papillomavirus (HPV) is the major risk factor and is a requirement for the development of cervical cancer. However, it remains unknown the interaction between KSHV and HPV, and the contribution of KSHV to cervical cancer development and pathogenesis. In the present study, we used Illumina microarray to detect the global gene profile altered in KSHV-infected siHa cervical cancer cell-line containing integrated HPV16 genome.

Transcriptome analysis of SiHa cells

Project description:This study assessed the transcriptional profile of SiHa cells. SiHa is a cervical cancer cell line with integrated HPV16, and was used as a model to study human gene expression in the context of integrated virus. Gene expression in SiHa, calculated by Cufflinks, was scored in windows around the locations of known viral integrations in patients or cell lines to determine if there was an association between gene expression and viral integration. We found that SiHa gene expression was higher near loci of integration for HPV18 vs. HPV16, cervical tissues vs. head and neck cancers, and cervical cancers vs. in vitro integrations. This study provides insight into the factors that may influence where viruses integrate in the human genome.
