Single-Cell RNA-Seq of PS2APP mouse hippocampi

Project description:Non-neuronal responses in neurodegenerative disease have received increasing attention as important contributors to disease pathogenesis and progression. Here we utilize single-cell RNA-sequencing to broadly characterize responses of twelve cell types in three different mouse models of Alzheimer’s Disease, capturing the effects of tau-only, amyloid-only, or combined tau- amyloid pathology. We characterize microglia, oligodendrocyte, astrocyte, and T cell responses and compare them across these models. Notably, we identify two distinct transcriptional states for oligodendrocytes induced during disease and determine their spatial distribution. Furthermore, we explore the impact of Trem2 deletion in the context of combined pathology. Trem2 knockout mice exhibit severely blunted microglial responses to combined tau and amyloid pathology, but responses from non-microglial cell types (oligodendrocytes, astrocytes, and T cells) are relatively unchanged. These results delineate core transcriptional states that are accessible in the degenerating brain and how they are influenced by a key Alzheimer’s Disease risk gene, Trem2.

However, most existing models lack essential cellular components, limiting their utility for studying brain physiology and disease. Here, we introduce Cerebroids (CBRs), a standalone, self-organising system that endogenously gives rise to all four major brain cell types: neurons, astrocytes, oligodendrocytes, and microglia, while being amenable to the formation of autologous vasculature, accompanied by scRNAseq data to confirm the cellular component fidelity to primary brain tissue.

Microglial proteomic signatures in APPPS1 and APP-KI mice

Project description:Microglial dysfunction is a key pathological feature of Alzheimer´s disease (AD), but little is known about proteome-wide changes in microglia during the course of AD pathogenesis and their consequences for microglial function. Here, we performed an in-depth proteomic characterization of microglia in two AD mouse models, the overexpression APPPS1 and the knock-in AppNL-G-F (APP-KI) model. Proteome changes were followed from pre-deposition to early, middle and advanced stages of amyloid plaque pathology, revealing a large panel of Microglial Amyloid Response Proteins (MARPs) that reflect a heterogeneity of microglial alterations triggered by Adeposition. We demonstrate that the occurrence of MARPs coincided with the deposition of fibrillar A, recruitment of microglia to amyloid plaques and phagocytic dysfunction. While the proteomic and functional microglial changes were already markedly seen in 3 months old APPPS1 mice, they were delayed in the APP-KI model that generates substantially less fibrillar A. The identified microglial proteomic fingerprints of AD provide a valuable resource for functional studies of novel molecular targets and potential biomarkers for monitoring AD progression or therapeutic efficacy.

CSF and brain proteomics of Trem2-mKate2 KI/wt.APPSAA/SAA.hTfR KI/KI mice

Project description:Triggering receptor expressed on myeloid cells 2 (TREM2) is a central regulator of microglial activity and loss-of-function coding variants are major risk factors for late onset Alzheimer’s disease (LOAD). To better understand the molecular and functional changes associated with TREM2 signalling in microglia, we generated a TREM2 reporter mouse model. Experimental mice expressed the reporter heterozygous while AppSAA and the hTfR knock-in were homozygous (Trem2-mKate2 KI/wt.APPSAA/SAA.hTfR KI/KI) and were treated with the antibody transport vehicle coupled TREM2 agonist antibody ATV:4D9. After the treatment CSF and brain samples were colleceted for proteomics analyses.
