Plexin D1 emerges as a novel target in the development of neural lineage plasticity in treatment-resistant prostate cancer

ABSTRACT: To explore the gene regulatory mechanisms underlying PLXND1 knockdown in neuroendocrine prostate cancer cells. We performed RNA sequencing analyses using PLXND1-knockdown C4-2B-MDVR and H660 cells to identify the gene programs affected by the PLXND1.

Project description:Prostate cancer C4-2B cells were cultured in enzalutamide in a dose-escalation manner. After sixty passages cells were resistant to enzalutamide, with a specific sets of genes been deregulated. We performed global gene expression analysis by cDNA microarrays to identify genes responsible for enzalutamide resistance in C4-2B-MDVR cells. Enzalutamide resistant C4-2B-MDVR cells were selected from C4-2B cells during long time enzalutamide treatment. Genes responsible for enzalutamide resistance were identified using C4-2B vs. C4-2B-MDVR RNA extraction and hybridization on Affymetrix microarrays.

Project description:The overall goal of this study was to identify genes that were differentially-expressed in C4-2B MDVR cells treated with Indocin. C4-2B MDVR were either treated with Indocin (20 uM) or DMSO vehicle control for 3 days, followed by isolation of total cellular RNA. Transcriptome analysis was performed with RNA-Sequencing (RNA-Seq) in order to identify differentially-expressed genes (DEGs) induced by Indocin treatment.

A MYC Family Switch: L-MYC Drives and Maintains Neuroendocrine Lineage Programs in Prostate Cancer

Project description:Neuroendocrine prostate cancer (NEPC) is an aggressive and therapy-resistant subtype that emerges through lineage plasticity following inhibition of the androgen receptor (AR) signaling pathway. In this study, MYCL was overexpressed in prostate adenocarcinoma C4-2B cells and knocked down in neuroendocrine prostate cancer NCI-H660 cells to investigate its role in lineage regulation. RNA-sequencing analysis revealed that MYCL overexpression suppresses AR signaling and MYC target signaling while inducing neuroendocrine-like transcriptional reprogramming. In contrast, MYCL knockdown disrupts neuroendocrine lineage identity and restores adenocarcinoma-associated gene expression programs, including reactivation of MYC. These findings suggest that MYCL plays a key role in regulating lineage plasticity and maintaining neuroendocrine transcriptional programs in prostate cancer.

Towards this goal, next-generation sequencing (NGS)-based gene expression profiling (RNA-Sequencing; RNA-Seq) was performed on castration-resistant prostate cancer (CRPC) C4-2B MDVR cells that were treated with vehicle control or one of the AR/AR-V7 inhibitors (ARVib), ARVib-7 or ARVib-31.

Transcriptional profiles of castration resistant prostate cancer cells treated with ITRI-148 PROTAC targeting AR and AR-V7

Project description:Androgen receptor (AR) signaling remains a key driver of castration-resistant prostate cancer, with AR splice variants like AR-V7 mediating resistance to second-generation antiandrogens. Targeting the AR N-terminal domain (NTD) offers a strategy to bypass ligand-binding domain-mediated resistance. We developed a VHL-based (ITRI-90) and a CRBN-based (ITRI-148) PROTAC degrader capable of targeting AR and AR-V7 via NTD. These compounds demonstrate potent antitumor activity towards castration-resistant and enzalutamide resistant xenograft models. Transcriptomic analyses confirm robust suppression of AR and AR-V7 target genes by ITRI-148 in VCaP cells. The transcriptomic effects of these AR-NTD degraders in enzalutamide-resistant C4-2B/MDVR cells closely mirror that of AR knockdown.
