RNA sequencing (RNA-seq) of Streptococcus pyogenes - group A Streptococcus (GAS) S119 strain in the presence or absence of asparagine (Asn) grown in a chemically defined medium (CDM) (van de Rijn, I. & Kessler, R.E. (1980).

ABSTRACT: The study was conducted to test the global effect of Asn on the transcriptome of the GAS S119 strain. Transcriptome levels in the presence or absence of Asn of cultures harvested at OD600 = 0.7 were compared. We report here that a global differential expression of genes belonging to diverse functional categories, including virulence, metabolism, transporters, regulatory proteins, kinases, hypothetical proteins, and other categories.

Polysome profiling of mRNA translation in neuroblastoma cells in response to asparagine/alanine supplement

Project description:Neuroblastoma, the most common extracranial solid malignancy in infants and young children, originates from aberrant neural crest cells and accounts for 10-15% of childhood cancer deaths. However, the underlying mechanisms regulating neuroblastoma progression remain to be determined. To investigate the mechanisms regulating tumorigenesis and aggressiveness, we employed the Illumina HiSeq X Ten as a discovery platform to analyze the polysome profiling of mRNA translation in neuroblastoma cells in response to asparagine/alanine (Asn/Ala) supplement. The results showed that 618 and 566 translating mRNAs were respectively elevated or reduced in SK-N-BE(2) after Asn/Ala treatment. Furthermore, we validated the polysome profiling results by western blot with high identity. Overall, our results provided fundamental information about the mRNA translation changes in response to Asn/Ala treatment in human neuroblastoma cells, and these findings will help us understand the pathogenesis of neuroblastoma progression.

Microarray analysis for Streptococcus pyogenes M1-CdhA (-) mutant

Project description:A total of 163 genes were found to be differentially expressed; 38 genes of them were up regulated and 125 genes were down regulated. Most notably the functional categories that were affected include Virulence genes (18 genes 11% of the total significantly differentiated genes), carbohydrate metabolism and cell envelop realted genes ( 31, 19.1% of the total), and aminoacid transport genes (21genes, 12.9%). Among the virulence-related genes, the most notable one were those belong to the production of capsule, the M protein, exotoxin, NAD glycohydrolase and Streptolysin S. Some of the capsule related genes were down regulated by more than 64 fold. These results indicate that CdhA (group A Streptococcal Cell division controlling and Chain-forming cell wall hydrolase) plays an important role in the regulation of virulence.
