Transcriptomic profile of monocytes/ macrophages from injured bone, muscle, skin or heart tissues of Treg-depleted mice

ABSTRACT: Mice were depleted of Tregs, and bone, muscle, skin or heart injuries were performed. On day 4 and 7 post-injury, the tissues were harvested and the endogenous monocytes/macrophages were sorted using FACS. These were then used for bulk RNA sequencing, to compare the transcriptomic profiles of injured tissue monocytes/ macrophages in Treg-depleted mice versus control mice post-injury.

Effect of dexamethasone treatment on the transcriptome of injured mouse hamstring muscle

Project description:Healthy muscle heals and repairs after injury. However, when mice are treated with dexamethasone, an agonist of glucocorticoid receptors, heterotopic ossifications (HO) develop in the injured muscle within fibrotic areas that fail to fully regenerate functional muscle. To understand the mechanisms driving heterotopic ossification in response to glucocorticoid receptor agonists, we performed RNA seqencing analysis on injured muscle. Mice underwent muscle injuries by intramuscular injection of cardiotoxin (CDTX). All mice were treated with either vehicle or dexamethasone (10mg/kg, intraperitoneal injection) daily from day 0-4. polyA+ RNA was isolated from whole muscle 4 days post injury, libraries generated and sequenced.

Project description:Gene expression of macrophages from injured muscles of bDCRE mutant mice was compared with that of Cebpb wt mice. Skeletal muscles were injured by cardiotoxin injection and infiltrating macrophages were isolated 6 days post-injury by FACS sorting of Mac-1+F4/80+ cells. 4 biological replicates were generated for each genotype and expression profiles were determined by hybridization to Affymetrix Moe430_2 arrays.

Project description:Patients with anterior cruciate ligament (ACL) tears have a significantly increased risk for developing knee osteoarthritis. These injuries often result in a knee effusion in response to the injury. Early changes in these effusions could be informative regarding initial steps in the development of post traumatic osteoarthritis. The purpose of this study was to test the hypothesis that the proteomics of knee synovial fluid changes over time following ACL injury.

Project description:Mice with either a cranial bone defect, skin biopsy punch injury, quadriceps volumetric muscle loss defect or myocardial infarction (MI) were treated with Tregs, either via local hydrogel-mediated delivery (for bone, muscle and skin), or with systemic (intravenous) delivery (for heart). Control mice were treated with hydrogel alone (for bone, muscle and skin) or PBS (for heart). On day 4 and 7 post-MI, the injured tissues were harvested and the endogenous monocytes/macrophages were sorted and used for mini-bulk RNA sequencing, to compare the transcriptomic profiles of injured tissue monocytes/macrophages in Treg-treated and untreated mice post-injury.

Gene expression in experimentally injured and control mouse spinal cords

Project description:This laboratory works on selectins and their carbohydrate ligands in lymphocyte homing and inflammation. The lab also studies heparin-degrading endosulfatases and their roles in regulating the interactions of growth factors and morphogens with proteoglycans. The purpose of the experiment is to examine gene expression profiles in spinal cords of injured as a function of time after a contusion injury. The tissue will be generated in the lab of Linda Noble, Professor in the Department of Neurological Surgery at UCSF who is an expert on the pathogenesis of spinal cord injury. Wild-type male C57B/6 mice (8-10 weeks of age) were used. Injuries were produced by contusion. Spinal cords from control mice (uninjured) and experimental (injured) mice were processed (4 and 7 days after injury). A 3 mm length of spinal cord (centered at the injury) and a 3 mm segment from a distant site were isolated from each animal. Tissue from 3 mice were pooled for each sample. Three replicate samples per treatment group were processed for RNA. Thus, a total of 18 RNA samples were hybridized to 18 gene chips. The analysis will determine whether specific glycosylation changes accompany spinal cord injury. Of particular interest are changes in the sulfation profile of proteoglycan GAG chains (e.g., chondroitin sulfate) and in the appearance of potential carbohydrate ligands for infiltrating leukocytes bearing L-selectin or other endogenous lectins.

Burn Injury-Induced G-CSF Secretion Depletes Spi-C+ Erythroblastic Island Macrophages and Impairs Medullary Erythropoiesis.

Project description:The erythroblastic island (EBI) functions as a niche where erythroblastic island macrophages (EBIMφ) are positioned within rings of erythroblasts, providing support and signals that orchestrate efficient erythropoiesis. Herein, we postulated burn injury impacts the EBI, given the nearly universal presence of anemia and inflammation in burn patients, and a divergent myeloid transcriptional signature we observed in murine bone marrow following burn injury, where G-CSF secretion increased neutrophil and monocyte transcripts but reduced EBIMφ transcripts. We utilized flow cytometry to investigate EBIMφ after imaging flow cytometry confirmed the heme induced transcription factor Spi-C as a robust marker of EBIMφ in Spicigfp/igfp mice. Two populations referred to as Spic+ macrophages and Spic+ Gr1 low monocytes possessed cell intrinsic GFP, with Spic+ macrophages distinguished by higher levels of GFP, F4/80, autofluorescence, and negligible CD115 staining compared to Gr1 low monocytes. Further, at least 50% of the Spic+ macrophages stained for CD163 while staining was negligible on Gr1 low monocytes. Since CD163 distinguished the populations and macrophage remnants could convolute flow cytometry analysis, total CD163 protein was quantified in marrow aspirates to corroborate results. Application of Spicigfp/igfp mice in studies revealed a G-CSF dependent reduction of Spic+ macrophages in post burn marrow that coincided with a loss of erythroid cells and that recombinant G-CSF alone was sufficient to reduce Spic+ macrophages in the marrow. These results provide the first evidence that burn injuries impact the EBI niche through G-CSF dependent depletion of EBIMφ and support the use of Spicigfp/igfp mice in investigation of EBIMφ.
