Monitor RNA synthesis in single cells (K562) by using NASC-seq

ABSTRACT: We applied the new transcriptome alkylation-dependent single-cell RNA sequencing (NASC-seq), to monitor newly synthesized and pre-existing RNA in single cells for K562. We tested the best s4U labeling concentration and duration for K562. We found that the tailored s4U concentration for K562 cell line is 50 μM. The proportion of new RNA increased with the longer labeling time.
