CCL17-expressing dendritic cells are crucial for the development of atherosclerosis

Project description:Dendritic cells (DCs) are essential for priming of immune responses. Although immune mechanisms are known to control the pathogenesis of atherosclerosis, the role of DCs remains elusive. Here we show that Ccl17 expressing mature, myeloid DCs accumulate within atherosclerotic lesions. Deletion of Ccl17 in apolipoprotein E-deficient (Apoe-/-) mice reduces the development and progression of atherosclerosis in several disease models. While Ccl17 expression by DCs dampened antigen-specific T cell proliferation, it is required for efficient polarization of T helper type 1 (Th1) and Th17 as reflected by a preponderance of Th2 cytokines in Ccl17-/- Apoe-/- mice. In line with these findings, only transfer of T cells from Apoe-/-, but not from Ccl17-/- Apoe-/- precipitated atherosclerosis in T cell depleted Apoe-/- recipients. These findings identify Ccl17+ DCs as central immune regulators in atherosclerosis and Ccl17 as a potential target in the treatment of this disease. There were two samples analyzed with no replicates included. The two samples should be compared with each other.

Tetramerization of Transcription Factor STAT5 in Monocytes Promotes Autoimmune-mediated Neuroinflammation via the Regulation of CCL17

Project description:STAT5 plays a critical role in mediating cellular responses following cytokine stimulation. The activated STAT5 proteins can form dimers and tetramers with distinct biological functions. The role of STAT5 tetramerization in autoimmune-mediated neuroinflammation has not been investigated. Using the STAT5 tetramer-deficient Stat5a-Stat5b N-domain double knock-in (DKI) mouse strain, we report here that STAT5 tetramers promote the pathogenesis of experimental autoimmune encephalomyelitis (EAE). The mild EAE phenotype observed in DKI mice correlates with the impaired extravasation of pathogenic Th17 cells and interactions between Th17 cells and monocyte-derived cells (MDCs) in the meninges. We further demonstrated that STAT5 tetramerization regulates the GM-CSF-dependent production of CCL17 by MDCs. Importantly, DKI Th17 cells expanded with CCL17 exhibit more severe EAE. Mechanistically, the effect of CCL17 is dependent on the activity of the integrin VLA-4. Thus, our study uncovered a novel GM-CSF-STAT5 tetramer-CCL17 pathway that promotes autoimmune neuroinflammation via the regulation of Th17 cell migration.

ScRNA-seq of cell fractions from LNs of CCL17-competent and CCL17-deficient mice on chew diet and western diet

Project description:The deficiency in CCL17 decreased atherosclerotic plaque burden by facilitating Treg maintenance and survival. cDC-derived CCL17 plays a crucial role in the recruitment and migration of various T cell subsets, including a subpopulation of CD4+ T cells, regulatory T cells (Tregs). To evaluate whether CCL17-deficient cDCs harbor an unique expression profile. we FACS-sorted CD45+CD11c+CD3-CD19- cells from lymph nodes (LNs) of Apoe-/-Ccl17wt/e or Apoe-/-Ccl17e/e mice on chew diet (CD) and performed single cell RNA sequencing (scRNAseq). We also performed scRNAseq to detect the expression of some chemokine receptors in sorted CD45+CD19-MHC+ cells from aortic LNs of Apoe-/-Ccl17wt/e or Apoe-/-Ccl17e/e mice on 6-week western diet (WD).
