Project description:Tiller angle is a key factor determining rice plant architecture, planting density, light interception, photosynthetic efficiency, disease resistance, and grain yield. The distribution of auxin and shoot gravitropism play important roles in regulating tiller angles of rice. Several tiller angle-associated genes have been cloned. However, the mechanisms underlying tiller angle control are far from clear. In this study, we isolate bta1-1, a mutant with an enlarged tiller angle throughout its life cycle. A detailed analysis reveals that BTA1 has multiple functions because several major agronomic traits, including tiller and panicle number, biomass production, secondary branch number per panicle, panicle weight, grain size, and grain weight, are increased in bta1-1 plants. Moreover, BTA1 is a positive regulator of shoot gravitropism in rice. Shoot responses to gravistimulation are disrupted in bta1-1 under both light and dark conditions. Gene cloning reveals that bta1-1 is a novel mutant allele of LA1. LA1 is able to rescue the tiller angle and shoot gravitropism defects observed in bta1-1. BTA1/LA1 is required to regulate the expression of auxin transporters and signaling factors that control shoot gravitropism and tiller angle. High-throughput mRNA sequencing is performed to elucidate the molecular and cellular functions of BTA1/LA1. The results show that BTA1/LA1 may have multiple functions in regulating nucleosome and chromatin assembly, and protein and DNA interactions. Our results provide new insight into the mechanisms whereby BTA1/LA1 controls shoot gravitropism and tiller angle in rice.

Project description:Leaf angle is mainly determined by the lamina joint (LJ), and contributes to ideal crop architecture for high yield. Here, we dissected five successive stages with distinct cytological features of LJs spanning organogenesis to leaf angle formation, and obtained the underlying stage-specific mRNAs and small RNAs, which well explained the cytological dynamics during LJ organogenesis and leaf angle plasticity. Combining the gene coexpression correlation with high-throughput promoter analysis, we identified a set of transcription factors determining the stage- and/or cytological structure-specific profiles. The functional studies of these TFs demonstrated that cytological dynamics determined leaf angle, and the knockout rice of these TFs with erect leaves significantly enhanced yield by maintaining the proper tiller number under dense planting. This work revealed the high-resolution mechanisms how the cytological dynamics of LJ determined the leaf erectness, and served as a valuable resource to remodel rice architecture for high yield via controlling population density.

Project description:Flowering of rice is triggered by transcriptional reprogramming at the shoot apical meristem (SAM) mediated by florigenic proteins produced in leaves in response to changes in the photoperiod. Florigens are more rapidly expressed under short day (SD) and include the HEADING DATE 3a (Hd3a) and RICE FLOWERING LOCUS T1 (RFT1) Phosphatidyl Ethanolamine Binding Proteins (PEBP). Hd3a and RFT1 are largely redundant at converting the SAM into an inflorescence, but whether they activate the same target genes and convey all photoperiodic information that modifies gene expression at the SAM is currently unclear. We uncoupled the contribution of Hd3a, RFT1 and SD to transcriptome reprogramming at the SAM, by RNA-sequencing of dex-inducible over-expressors of single florigens and wild type plants exposed to photoperiodic induction. Fifteen highly differentially expressed genes common to Hd3a, RFT1 and SD were retrieved, ten of which still uncharacterized. Detailed functional studies on some candidates revealed a role for LOC_Os04g13150 in determining tiller angle and bract development and the gene was renamed BROADER TILLER ANGLE 1 (BRT1). We identified a core set of genes common to florigenic and photoperiodic induction and defined the function of a novel florigen target controlling tiller angle.

Project description:Tiller angle, a key agronomic trait for achieving ideal plant architecture and increasing grain yield, is regulated mainly by shoot gravitropism. Strigolactones (SLs) are a group of newly identified plant hormones that are essential for shoot branching/rice tillering and have further biological functions as yet undetermined. Through screening for suppressors of lazy1 (sols), a classic rice mutant exhibiting large tiller angle and defective shoot gravitropism, we identified multiple SOLS that are involved in the SL biosynthetic or signaling pathway. We show that SL biosynthetic or signaling mutants can rescue the spreading phenotype of lazy1 (la1) and that SLs can inhibit auxin biosynthesis and attenuate rice shoot gravitropism, mainly by decreasing the local indoleacetic acid content. Although both SLs and LA1 are negative regulators of polar auxin transport, SLs do not alter the lateral auxin transport of shoot base, unlike LA1, which is a positive regulator of lateral auxin transport in rice. Genetic evidence demonstrates that SLs and LA1 participate in regulating shoot gravitropism and tiller angle in distinct genetic pathways. In addition, the SL-mediated shoot gravitropism is conserved in Arabidopsis. Our results disclose a new role of SLs and shed light on a previously unidentified mechanism underlying shoot gravitropism. Our study indicates that SLs could be considered as an important tool to achieve ideal plant architecture in the future.

Project description:WRKY transcription factors constitute a crucial protein superfamily that plays a vital role in orchestrating diverse responses to environmental stimuli in plants. Although considerable research has been conducted on the stress resistance mechanisms associated with WRKYs, little is known about the roles of WRKYs in regulating plant growth and development. Here, we identified a key regulator of the rice leaf angle and plant architecture, OsWRKY36, from a rice oswrky mutant library. OsWRKY36 is highly expressed in the leaf lamina joint and promotes cell growth and expansion in adaxial parenchyma cells, leading to a greater leaf angle. A mechanistic investigation revealed that OsPUB24, an E3 ubiquitin ligase that is inhibited by brassinosteroid (BR), interacts with and facilitates the ubiquitination and degradation of OsWRKY36. Genetic evidence indicated that OsWRKY36 acts downstream of OsPUB24 to regulate the BR-induced increase in the leaf angle. Furthermore, OsWRKY36 physically interacts with DLT, a positive regulator of BR, and cooperatively activates the expression of the BR response gene OsBZR4. Thus, BR facilitates the degradation of OsPUB24, leading to enhanced stability of the OsWRKY36 protein. Consequently, a substantial quantity of OsWRKY36 interacts with DLT, thereby inducing the expression of OsBZR4 and ultimately exerting a positive regulatory effect on BR-induced leaf inclination. Collectively, our findings reveal the mechanism underlying the regulation of leaf inclination and plant architecture by the BR-OsPUB24-OsWRKY36 and DLT-OsWRKY36-OsBZR4 modules and thus provide an innovative strategy for optimizing crop production by manipulating the BR signalling pathway.
