Erianin inhibits the progression of pancreatic cancer by directly targeting AKT and ASK1

ABSTRACT: Erianin, a natural product extracted from Dendrobium, possesses multiple pharmacological properties, including antioxidative and antitumor activities. The objective of this study was: to evaluate the anti-pancreatic cancer activity of erianin and explore its underlying mechanisms.To further explore the potential molecular mechanisms of erianin in pancreatic cancer, we performed RNA Sequencing analysis to identify DEGs under erianin stimulation in SW1990 cells.

We carried out microarray analyses using the CD166+ and CD166- separated cells from both Panc-1 and SW1990. The quality of the RNA samples was evaluated using a 2100 Bioanalyzer (Agilent Technologies, Waldbronn, Germany). We used a Human HT-12v4 Expression BeadChip (Illumina, San Diego, CA) for these analyses. The data were analyzed using BeadStudio software version 3.2.3 (Illumina).

SIRT5 Directly Inhibits the PI3K/AKT Pathway in Prostate Cancer Cell Lines.

Project description:Background/aimProstate cancer (PCa) is the most commonly diagnosed genital cancer in men globally. Among patients who develop advanced PCa, 80% are affected by bone metastasis, with a sharp drop in survival rate. Despite efforts, the details of mechanisms of metastasis of PCa remain unclear. SIRT5, an NAD+-dependent deacylase, is hypothesized to be a crucial regulator of various cancers. The role of SIRT5 in cancer has not been extensively studied compared to other SIRTs. In this study, we showed significantly decreased levels of SIRT5 in PC-3M, a highly aggressive PC-3 cell variant.Materials and methodsWe characterized the differentially expressed proteins between parental and SIRT5 KO PC-3 cells using quantitative proteomics analysis.ResultsA significant increase in expression of interleukin-1β (IL-1β) in SIRT5 KO cells was observed, and the PI3K/AKT/NF-ĸB signaling pathway was found significantly elevated in SIRT5 KO cells by the Gene Ontology annotation and KEGG pathway functional enrichment analysis. Moreover, we confirmed that SIRT5 can bind PI3K by immunoprecipitation analysis.ConclusionThis study is the first to demonstrate a relationship between SIRT5 and PCa metastasis, suggesting that SIRT5-mediated inhibition of the PI3K/AKT/NK-kB pathway is reduced for secondary metastasis from bone to other tissues.

Gene expression data from SW1990 cells stably expressing Fbw7

Project description:Fbw7 plays a negative role in pancreatic cancer tumorigenesis and progression. To further clarify the function and mechanism that Fbw7 plays in pancreatic cancer,mRNA microarray assays were performed to identify the genes and signaling pathways that were changed upon Fbw7 overexpression. Fbw7 overexpressing SW1990 cells were cultured for RNA extraction and hybridization on Affymetrix mRNA microarrays. These were compared against the control,which were infected by empty control lentiviral particles.
