Project description:Drought is a major cause of crop yield loss worldwide. Plants can adopt different mechanisms to survive under water deficit, where stomatal closure leads to CO2 limitation. This requires increased dissipation of light energy as heat by non-photochemical quenching (NPQ), changes in the structural configuration of light-harvesting complexes, or changes in the mode of photosynthetic electron flow (i.e. linear vs. cyclic). Under field conditions, where light intensity can change rapidly, this problem becomes even more challenging. Particularly the slow release from photoprotection (NPQ relaxation) was recently shown to cause significant loss of potential yield. We analyzed the acclimation of two different wheat varieties and compared their molecular acclimation strategy to Arabidopsis under moderate drought stress at a slow soil dehydration rate, mimicking realistic field conditions. We monitored their photosynthetic activity under fluctuating light intensities and integrated functional and structural data, based on a detailed analysis of photosynthetic parameters combined with biochemical analysis and unbiased shot-gun proteomics. Contrary to previous models based on the damage of photosynthetic complexes as consequence of drought, we found that plants actively preserve and fine-tune their photosynthetic machinery under drought to adjust the photosynthetic electron transfer to fast changes of light intensity. Strikingly, Arabidopsis and wheat use different molecular strategies for this acclimation. While clear differences in NPQ relaxation and phosphorylation levels of photosynthetic proteins were observed in wheat, Arabidopsis modified the light energy distribution among photosynthetic complexes without changing PSII-LHCII phosphorylation. The fine-tuning of NPQ relaxation can therefore represent a potential trait for crop breeding.

Project description:Elevated atmospheric CO2 can influence the structure and function of rhizosphere microorganisms by altering root growth and the quality and quantity of compounds released into the rhizosphere via root exudation. In these studies we investigated the transcriptional responses of Bradyrhizobium japonicum cells growing in the rhizosphere of soybean plants exposed to elevated atmospheric CO2. The results of microarray analyses indicated that atmospheric elevated CO2 concentration indirectly influences on expression of large number of Bradyrhizobium genes through soybean roots. In addition, genes involved in C1 metabolism, denitrification and FixK2-associated genes, including those involved in nitrogen fixation, microanaerobic respiration, respiratory nitrite reductase, and heme biosynthesis, were significantly up-regulated under conditions of elevated CO2 in the rhizosphere, relative to plants and bacteria grown under ambient CO2 growth conditions. The expression profile of genes involved in lipochitinoligosaccharide Nod factor biosynthesis and negative transcriptional regulators of nodulation genes, nolA and nodD2, were also influenced by plant growth under conditions of elevated CO2. Taken together, results of these studies indicate that growth of soybeans under conditions of elevated atmospheric CO2 influences gene expressions in B. japonicum in the soybean rhizosphere, resulting in changes to carbon/nitrogen metabolism, respiration, and nodulation efficiency.

Project description:Increasing photosynthetic efficiency remains a major goal across the globe given its central role in CO2 assimilation. Here, we identified a nuclear genome-residing orphan gene, comprised of 3 exons, two with apparent endophytic origins and the third being a highly conserved fragment of the RIBULOSE BISPHOSPHATE CARBOXYLASE/ OXYGENASE LARGE SUBUNIT (RuBisCo). This novel gene, here-on referred to as Populus RuBisCo-like (PRL-1) gene localizes in chloroplast, endoplasmic reticulum and nuclear compartments of plant cells, acts as a transcriptional repressor and modulates adaptation to fluctuating light. P. trichocarpa genotypes with high PRL-1 expression levels quickly dissipated non-photochemical quenching (NPQ) upon transitioning from high to low light and rapidly induced NPQ upon returning to high light. The dynamic modulation of NPQ yields high quantum efficiency of linear electron transport (PSII) and 10-20% increased quantum efficiency of CO2 assimilation (CO2). The enhanced photosynthesis efficiency corresponded with increased plant height and biomass in P. trichocarpa with up to 35 % and 100% increases under field and greenhouse conditions. Transgenic Arabidopsis plants heterologously expressing the PRL-1 gene gained up to 200% in biomass and 97% in seed yield under greenhouse conditions. Taken together, PRL-1 presents a novel and trackable target for increasing photosynthetic efficiency in plants.

Project description:This study compared the photosynthetic performance and the global gene expression of the winter hardy wheat Triticum aestivum cv Norstar grown under non-acclimated (NA) or cold-acclimated (CA) condition at either ambient CO2 or elevated CO2 (EC). CA Norstar maintained comparable light saturated and CO2 saturated rates of photosynthesis but lower quantum requirements for photosystem II and non photochemical quenching relative to NA plants even at EC. Neither NA nor CA plants were sensitive to feedback inhibition of photosynthesis at EC. Global gene expression using microarray combined with bioinformatics analysis revealed that genes affected by EC were 3 times higher in NA (1022 genes) compared to CA (372 genes) Norstar. The most striking effect was the down-regulation of genes involved in the plant defense responses in NA Norstar. In contrast, cold acclimation reversed this down regulation due to the cold induction of genes involved in plant pathogenesis resistance, and cellular and chloroplast protection. These results suggest that EC have less impact on plant performance and productivity in cold adapted winter hardy plants in the northern climates compared to warmer environments. Selection for cereal cultivars with constitutively higher expression of biotic stress defense genes may be necessary under EC during the warm growth period and in warmer climates.

Project description:PsbS Protein Interactions during Non-photochemical quenching The non-photochemical quenching of excitation energy (NPQ) describes a photoprotective mechanism in the antenna of PSII which dissipates excess excitation energy as heat at the level of 1Chl* and by that prevents the formation of singlet oxygen in PSII. Four different components contribute to NPQ, namely qT, qE, qZ and qI. Under saturating light conditions, the qE represents the dominant NPQ component; qE is based on a complex mechanism which strictly depends on the ΔpH across the thylakoid membrane, the PsbS protein and the xanthophyll zeaxanthin (Zx). The central role of PsbS in these processes is related to the function of PsbS as sensor of the lumen pH. However, the molecular basis of this central function and particularly the underlying interactions of PsbS with PSII antenna proteins that lead to energy quenching are largely unclear. In this work, we present an in vitro approach to identify protein transient interactions involving the PsbS protein during NPQ induction and relaxation in Arabidopsis thaliana, revealing its direct role in the formation of quenching sites in the antenna complexes of photosystem II.

Limited CO2 availability will cause higher rates of photorespiration and affect the plant redox homeostasis. We studied the transcriptomic impact of exposing plants to a lower CO2 environment to further eliculidate the signaling pathways during photorespiratory stress.

Project description:Plant respiration responses to elevated growth [CO2] are key uncertainties in predicting future crop and ecosystem function. In particular, the effects of elevated growth [CO2] on respiration over leaf development are poorly understood. This study tested the prediction that, due to greater whole-plant photoassimilate availability and growth, elevated [CO2] induces transcriptional reprogramming and a stimulation of nighttime respiration in leaf primordia, expanding leaves, and mature leaves of Arabidopsis thaliana. In primordia, elevated [CO2] altered transcript abundance, but not for genes encoding respiratory proteins. In expanding leaves, elevated [CO2] induced greater glucose content and transcript abundance for some respiratory genes, but did not alter respiratory CO2 efflux. In mature leaves, elevated [CO2] led to greater glucose, sucrose and starch content, plus greater transcript abundance for many components of the respiratory pathway, and greater respiratory CO2 efflux. Therefore, growth at elevated [CO2] stimulated dark respiration only after leaves transitioned from carbon sinks into carbon sources. This coincided with greater photoassimilate production by mature leaves under elevated [CO2] and peak respiratory transcriptional responses. It remains to be determined if biochemical and transcriptional responses to elevated [CO2] in primordial and expanding leaves are essential prerequisites for subsequent alterations of respiratory metabolism in mature leaves. Arabidopsis plants were grown in either ambient (370 ppm) or elevated (750 ppm) CO2. Leaf number 10 was harvested when it was a primordia, expanding, or mature in each of the CO2 treatments.

Project description:There are seedling samples (high CO2 exposure of 0h, 2h, 6h, 12h, 1d, 3d, 7d, 14d) with duplicates in two chambers. Arabidopsis WT (Col-0) seeds were plated on Murashige and Skoog plates and placed at 4°C in darkness for at least 2 d to synchronize germination. Plants were grown at 22C under long-day conditions (16-h light and 8-h dark) in two atmospheric CO2 environments: ambient (CO2: 390 μmol molâ??1) or elevated (CO2: 780 μmol molâ??1). Plants were grown in ambient atmospheric CO2 concentration and then exposed to elevated CO2 for 0 h, 2 h, 6 h, 12 h, 1 d, 3 d, 7 d and 14 d. 14-d-old seedlings were sampled at the same time. Treatments for elevated CO2 were carried out two times using two different chambers. Duplicate samples were collected from each chamber experiment. Totally, four biologically replicates were prepared in each condition.
