Terminal nucleotidyl transferases govern secondary siRNA production at distinct steps in plants [RACE]

ABSTRACT: In this study, we aim at understanding the contributions from TUTases, HESO1 and URT1, in miRNA-targeted secondary siRNA production in Arabidopsis thaliana. We have produced 3'RACEseq libraries from various T-DNA insertation single mutants and one double mutant, and compared their capability of uridylating AFB2 and MYB33 5'cleavage fragments. We also compared their uridylyl transfer activity of the AFB3 3' end extremity. Here, we have sequenced biological replicates from Col-0 WT, urt1-1, heso1-4, urt1-1/heso1-4 (set1 and set3) and biological replicates from Col-0 WT, urt1-1, mir393b and rrp4 (set2).
