Transcriptome analysis of the CA1 region of the rat hippocampus reveals the signature of experience-induced gene expression in the left and right hemispheres

ABSTRACT: We aimed to characterize experience-induced gene expression in lateralization and to assess the sensitivity of the left and right CA1 regions of the rat hippocampus to an enriched environment. RNA sequencing was performed on the left and right CA1 region tissues dissected from the rats, which lived in isolated or enriched conditions. Transcriptomic analysis was used to characterize the gene expression profiles of the left and right CA1 regions and to determine the effects of different environmental conditions. Hippocampal CA1 lateralization was observed at the gene expression level in both housing conditions.

FMRP cTag CLIP from mouse CA1 pyramidal neurons

Project description:Loss of the neuronal RNA binding protein FMRP causes Fragile X Syndrome (FXS), the most common cause of inherited intellectual disability, yet it is unknown which brain regions and cell types within them contribute to disease pathophysiology. We used conditional tagging of FMRP and CLIP (cTag FMRP CLIP) to examine FMRP targets specifically in CA1 hippocampal neurons, a critical cell type for learning and memory known to have altered synaptic function in FXS. Integrating this data with analysis of ribosome-bound transcripts from the same neuronal population revealed CA1-enriched binding of autism-relevant mRNAs, and unexpected CA1-specific regulation of transcripts encoding circadian proteins.

Project description:Aortic calcification is essential for atherosclerosis (AS). Carbonic anhydrase I (CA1) can promote calcification in aortic tissues to stimulate AS plaque formation, and methazolamide (MTZ), a CA inhibitor, can be used to treat AS by inhibiting CA1-mediated calcification. This study used an established a CA1 [+/+] KI Apoe [-/-] mice to confirm stimulatory role of CA1 on AS and the therapeutic effect of MTZ and investigate the mechanism. In this study, Apoe [-/-] mice with CA1 overexpression were induced to AS by administrating high-fat food. The present study confirmed that CA1 is AS-therapeutic target by mediating the proportions of B1/MZB-1 cells, CD8+T cells, CD11c+ macrophages and CD11c- macrophages as well as their AS-related gene expressions and focal adhesion pathway. MTZ can be used for AS therapy.

Expression data from left versus right mouse entorhinal cortex (EC).

Project description:The entorhinal cortex of the mouse seems to be sensitive to molecular mechanisms that have been linked to the pathology of Alzheimer's disease. In this microarray study we are interested in comparing the expression profile of the left versus the right EC of the mouse, in order to understand if there is a significant difference in gene expression that might reveal any insights into the differential activation of these areas. We used microarrays to detail the global programme of gene expression underlying a possible lateralization of the EC in the mouse brain (left versus right EC). The left and the right entorhinal cortices of 6 month-old C57BL/6 wild-type mice was dissected out by following anatomical landmarks and guided by the Mouse Brain Paxinos and Franklin’s atlas. Samples were immediately processed for RNA extraction by using the RNeasy Kit from QIAGEN according to manufacturer’s instructions. Before running the microarrays, RNA quality and integrity was monitored on an Agilent BioAnalyzer. Samples were then run on Affymetrix Genechip Mouse gene 2.0 ST arrays following Affymetrix’s standard procedures (n=3 per cortical hemisphere). Microarrays data were analyzed through the use of Ingenuity® iReport (Ingenuity® Systems, www.ingenuity.com).
