Project description:Dogs frequently develop glaucoma, a disease that leads to vision loss due to loss of retinal ganglion cells and degeneration of axons within the optic nerve. We used Affymetrix Gene chips to characterize transcriptional changes between healthy and glaucomatous retinas. These data describe gene expression changes in the canine retina with glaucoma. RNA was isolated from the retinas of 5 dogs with advanced glaucoma and from 5 normal individuals.

Local and systemic changes in lipid profile as potential biomarkers for canine atopic dermatitis.

Project description:Lipids play a critical role in the skin as components of the epidermal barrier and as sig-naling molecules. Atopic dermatitis in dogs is associated with changes in the lipid composition of the skin, but whether these precede the onset of dermatitis or occur secondary to the dermatitis is unclear. We applied rapid lipid profiling mass spectrometry methods to skin and blood samples of dogs and determined changes following systemic treatment. Thirty control dogs and 30 atopic dogs with mild to moderate dermatitis were enrolled. Marked differences in lipid profiles were observed between control, nonlesional and lesional skin of dogs. Additionally, there were significant altera-tions in the lipid composition of the blood samples indicating systemic changes in lipid metabolism. Treatment with oclacitinib or lokivetmab resulted in a significant decrease of the disease clinical severity associated with changes in skin and blood lipids. A set of lipid features of the skin were selected as biomarkers that classified samples as control or atopic dermatitis with 95% accuracy, whereas blood lipids discriminated between control and atopic dogs with 82% accuracy. These data suggest that atopic dermatitis is a systemic disease and support the use of rapid lipid profiling to identify novel biomarkers.

Quantitative label-based canine faecal proteomics

Project description:In small animals, diarrhoea is a regular presentation, the aetiology of which can be varied, from an isolated self-limiting event to more serious episodes requiring symptomatic and supportive treatment. Chronic diarrhoea, defined as diarrhoea lasting longer than 3 weeks, is a relatively common problem often prompting referral for more detailed investigation. Although the potential cause of chronic diarrhoea includes chronic intestinal parasitism and alimentary neoplasia (specifically lymphoma), the majority of these dogs will be diagnosed with idiopathic canine chronic enteropathy (CCE), previously referred to as idiopathic inflammatory bowel disease (IBD). Of those cases diagnosed with CCE, further subdivision based on response to treatment trials leads to a final diagnosis of antibiotic responsive diarrhoea (ARD), food responsive diarrhoea (FRD) or idiopathic inflammatory bowl disease (IBD). The aim of this study was to compare the proteomes of faeces of dogs with chronic enteropathies and a population of healthy dogs, and furthermore to identifying proteins that might be useful in distinguishing FRD from ARD from IBD.

Retinal Gene and Protein Expression Associated with CNTF-Induced Deconstruction of Photoreceptor Outer Segments in normal and CNGB3-achromatopsia dogs

Project description:Background: Intravitreal injection of CNTF leads to deconstruction and regeneration of photoreceptor outer segments in normal dogs, and improves the success rate of viral CNGB3 gene replacement therapy in dogs (> 1 year) with CNGB3-achromatopsia. Objectives: The goal of this study was to examine the changes caused by intravitreal CNTF injection to the retinal gene expression profiles and the retinal function of normal and CNGB3-achromatopsia affected dogs. Retinal gene expression profiles were evaluated with canine specific Agilent Oligo Microarray containing 42,034 60-mer oligonucleotide probes. Expression of eleven selected genes (BCL2, CCL2, STAT3, TNFRSF1A, ARR3, RHO, TYROBP, COL1A1, C3, CNGA1, and CNGB3) was quantified in the same RNA samples by quantitative real-time PCR. The results confirmed the differential expression levels observed with the microarray analysis.

Expression data from adult mouse normal and damaged retina from B6 and 129 mouse strains

Project description:Retinal damage causes proliferation of Muller glia, but the degree of proliferation depends on mouse strains. Muller glial proliferation was significantly promoted by the addition of GSK3 inhibitor in 129, but not in B6. We used retinal explant culture as a model for retinal damage which caused preferential photoreceptor death in a few days. We used microarrays to detail the global programme of gene expression regulating the proliferative potential of Muller glia after retinal damage. Total RNA from intact whole retina, retinal tissue cultured for three days, and retinal tissue cultured with chir99021 for three days was used. Retinal tissues from 10 weeks old mice were used.
