Project description:Obese women who develop gestational diabetes show lower adiponectin levels across pregnancy than obese euglycemic women suggesting that obese women with low adiponectin levels have an impaired capacity to handle the metabolic changes during pregnancy. Adiponectin acts on the placenta during pregnancy; this fact allows for the interesting possibility that adiponectin can exert endocrine effects on the developing fetus. The aim was to investigate how adiponectin affects fetal growth, placenta function and metabolic functions during pregnancy. Wild-type (wt) and adiponectin transgenic (APNtg) mice were fed normal chow or a high fat/high sucrose (HF/HS) diet for 8 weeks before mating. Dams and fetuses were dissected at gestational day 18.5. Maternal adiponectin overexpression decreased fetal body weight in dams on normal chow, and even more in dams on HF/HS diet. Pools of two placentas or two livers from one male and one female fetus with the same genotype (wt or APNtg) from the same dam on HF/HS diet were used in proteomic analysis, and a total of five pools per group (wt-wt, wt-APNtg, APNtg-wt, APNtg-APNtg) were included in the liquid chromatography-tandem mass spectrometry (LS-MS/MS) analysis. In total, 7290 proteins were identified in placenta, and 7301 proteins in fetal liver. Next, we analyzed phosphosites, in total,19264 sites were identified in placenta proteins and 15334 sites were identified in fetal liver proteins. We then applied pathway analysis to the total protein and phosphopeptide data using PRISM and Enrichr.

Project description:Analysis of gene expression in uterine natural killer (NK) cells purified from either lean (control) or obese pregnanct women. The studys' aim is to identify differentially expressed genes and pathways in natural killer cells that are affected by maternal obesity. Results provide mechanitic insight into gene pathways altered in the condition of obeisty. Total RNA obtained from immuno-magnetic bead purified natural killer cells from uterine decidual mucosa from 7 to 10 week old pregnancies from 13 lean (BMI 20-24.9) and 11 obese (BMI >30) women. All women were between 19 and 35 years of age. In addition to BMI, blood serum CRP (hsCRP), a measure of inflammation, was measured via ELISA.

Project description:This study assesses fatty acid composition of lipids in matched maternal plasma and placental tissue from lean, obese and gestational diabetes (GDM) women stratified by fetal sex. Obesity and GDM are associated with adverse pregnancy outcomes and program the offspring for cardiometabolic disease in a sexually dimorphic manner. We collected maternal plasma and placental villous tissue following elective cesarean section at term from women who were lean (pre-pregnancy BMI 18.5-24.9), Obese (BMI>30) and type A2 GDM (matched to obese BMI) with either a male or female fetus (n=4 each group). Lipids were extracted and fatty acid composition of different lipid classes were analyzed by LC-MS/MS to find significant changes in GDM vs Obese, GDM vs Lean, and Obese vs Lean.

Project description:Maternal obesity has both a direct effect on the outcome of pregnancy but also programs the fetus for obesity and metabolic syndrome in later life. Due to its roles at the interface between mother and fetus, the placenta transduces and mediates the effects of the obese intrauterine environment on the fetus. Here we have generated genome-scale high-resolution maps of 5mC and 5hmC in human placentas of obese and healthy weight pregnancies. Widespread alterations to the placental epigenome identified across the genome in the setting of maternal adiposity are primarily characterized by increase in 5mC and reciprocal decrease in 5hmC. This is also evident at the two pregnancy-associated gene clusters on chromosomes 17 and 19. In addition, maternal adiposity is associated with downregulation of ten-eleven translocation (TET) and isocitrate dehydrogenase (IDH) family genes and is accompanied by decreased levels of α-ketoglutarate (αKG) in the placenta. Of note, αKG levels exhibit a close positive correlation with both ascorbate and aconitate levels and less substantial but significant association with global 5hmC levels and placenta-specific gene expression. These observations suggest a mechanistic linkage between obesogenic environment and conversion efficiency of 5mC to 5hmC and highlight the idea that TET-mediated active DNA demethylation pathway can sense and respond to metabolic derangements and plays a role in subsequent cellular adaptation. Collectively, this study demonstrates that altered DNA demethylation process is likely one of the mechanisms underlying metabolic modulation of the placental epigenome with maternal adiposity. Project description:Context: An increase in maternal insulin resistance (IR) during pregnancy is essential for normal fetal growth. The mechanisms underlying this metabolic adaptation to pregnancy are poorly understood. Placenta factors are believed to instigate and maintain these metabolic changes, as IR decreases shortly after delivery. Methylation of placental gene loci that are common targets for miRNAs, are associated with maternal IR. Objective: We hypothesized that placental miRNAs targeting methylated loci are associated with maternal IR during late pregnancy. Methods: Placental miRNA expression was measured via small RNA-sequencing in a subset of 40 placentas selected by maternal pre-gravid BMI and neonatal adiposity. Results: Median maternal age was 27.5 years, with a median pre-pregnancy BMI of 24.7 kg/m2, and median HOMA-IR of 2.9. Among the five selected miRNA, near delivery maternal HOMA-IR correlated with the placental expression of miRNA-371b-3p (r=0.25; p=0.008) and miRNA-3940-3p (r=0.32; p=0.0004) across the 132 individuals. After adjustment for confounding variables, placental miRNA-3940-3p expression remained significantly associated with HOMA-IR (β=0.16; p=0.03). Conclusion: Placental miRNA may have an autocrine or paracrine effect - regulating placental genes which play a role in modulating maternal IR.

Project description:The aim of the project was to compare global gene expression in adipocytes from obese patients and lean controls. Subcutaneous adipose tissue was collected from severely obese patients undergoing bariatric surgery (average body-mass index (BMI) of 45.5 kg/m2 (n = 12, thereof 4 men) and healthy lean patients undergoing hernia repairs (average BMI of 24.2 kg/m2 (n = 12, thereof 7 men), between 27 and 56 years of age. Adipocytes were isolated by collagenase treatment of adipose tissue, followed by filtering and centrifugation. Floating adipocytes were lysed in Qiazol before RNA purification and microarray analysis.
