Project description:In order to Sp1 and EZH2-specific gene expression pattern in breast cancer, small interfering RNA for Sp1 and EZH2 was transfected in MDA-MB-231 cells. Four groups of samples are included: 1. si control (siCT_1, siCT_2, siCT_3). 2. siSp1 (siSp1_1, sisSp1_2, siSp1_3). 3. si control (siCT_A, siCT_B, siCT_C). 4. siEZH2 (siZEH2_1, siZEH2_2,siEZJ2_3). Gene expression profiles of Sp1 and EZH2 depleted cells were compared to that of siRNA controls. Experiments were performed in MDA-MB-231 cells.

Project description:Epigenetic modifications modulate gene expression and, when deregulated, contribute to the development and progression of cancer. G9a (EHMT2) and EZH2 are two histone methyltransferases commonly upregulated in several cancer types, leading to the silencing of tumor suppressor genes. We performed high throughput sequencing of a breast cancer cell line (MDA-MB-231), an ovarian cancer cell line (CAOV3) and a melanoma cell line (D14) treated with a G9a inhibitor (UNC0642), EZH2 inhibitor (GSK126) and their combination.

Genome-wide maps of chromatin state in MDA-MB-231 cells [ChIP-seq]

Project description:We report a series of ChIP-Seq results in MDA-MB-231 cells. Briefly, we performed ChIP-Seq experiments with antibodies specific for PHF20L1, MTA1, EZH2, H3K27me2, H3K27me3 and H3K27ac in normal or knockdown of PHF20L1 MDA-MB-231 cells . We found that knockdown of PHF20L1 reduced the enrichment of MTA1, EZH2, H3K27me2, and H3K27me3 on the PHF20L1 target gene, and the enrichment of H3K27ac increased. This study gave us a new understanding of the role of PHF20L1 in chromatin status and gene transcription.
