Project description:BackgroundCoevolution between pathogens and their hosts decreases host morbidity and mortality. Bats host and can tolerate viruses which can be lethal to other vertebrate orders, including humans. Bat adaptations to infection include localized immune response, early pathogen sensing, high interferon expression without pathogen stimulation, and regulated inflammatory response. The immune reaction is costly, and bats suppress high-cost metabolism during torpor. In the temperate zone, bats hibernate in winter, utilizing a specific behavioural adaptation to survive detrimental environmental conditions and lack of energy resources. Hibernation torpor involves major physiological changes that pose an additional challenge to bat-pathogen coexistence. Here, we compared bat cellular reaction to viral challenge under conditions simulating hibernation, evaluating the changes between torpor and euthermia.ResultsWe infected the olfactory nerve-derived cell culture of Myotis myotis with an endemic bat pathogen, European bat lyssavirus 1 (EBLV-1). After infection, the bat cells were cultivated at two different temperatures, 37 °C and 5 °C, to examine the cell response during conditions simulating euthermia and torpor, respectively. The mRNA isolated from the cells was sequenced and analysed for differential gene expression attributable to the temperature and/or infection treatment. In conditions simulating euthermia, infected bat cells produce an excess signalling by multitude of pathways involved in apoptosis and immune regulation influencing proliferation of regulatory cell types which can, in synergy with other produced cytokines, contribute to viral tolerance. We found no up- or down-regulated genes expressed in infected cells cultivated at conditions simulating torpor compared to non-infected cells cultivated under the same conditions. When studying the reaction of uninfected cells to the temperature treatment, bat cells show an increased production of heat shock proteins (HSPs) with chaperone activity, improving the bat's ability to repair molecular structures damaged due to the stress related to the temperature change.ConclusionsThe lack of bat cell reaction to infection in conditions simulating hibernation may contribute to the virus tolerance or persistence in bats. Together with the cell damage repair mechanisms induced in response to hibernation, the immune regulation may promote bats' ability to act as reservoirs of zoonotic viruses such as lyssaviruses.

Project description:Staphylococcus aureus (S. aureus) is a Gram-positive opportunistic bacterium commonly isolated on the skin and in the nasal cavity of many healthy individuals. It can cause infections, from local skin and soft tissue infections (SSTIs) like wound infections, abscesses, and cellulitis, to more severe conditions such as endocarditis, pneumonia, sepsis, and toxic shock syndrome. Its pathogenicity is due to its numerous virulence factors and its adaptability to environmental changes, including pH, oxygen, CO2, and temperature during host infections. Understanding metabolic adaptation is a complex step for identifying new therapeutic targets to combat antibiotic-resistant pathogens. Proteomic approaches are essential for studying microbial protein expression during environmental changes to understand metabolic pathways and antibiotic resistance mechanisms. We investigated the protein expression of two MRSA strains under three experimental conditions (ECs) to simulate the variations in pH, temperature, and NaCl concentration expected during a wound infection. One strain is Livestock-Associated (LA-MRSA) ST398, which colonizes the nasal cavity of pigs and has also been reported in other animals and used in the food industry. It poses a risk for farmers and people in direct contact with animals, such as veterinarians. The other strain is Community Associated (CA-MRSA), JE2 strain (USA300 isolate), one of the major causes of SSTI and the main cause of CA infections in the USA. The selected ECs mimic the stress conditions occurring on the skin during infection with S. aureus. These ECs were named experimental condition 1 (EC1), characterized by pH 7 at 37°C and considered as a control condition; experimental condition 2 (EC2), characterized by pH 6 at 35°C; and experimental condition 3 (EC3), characterized by pH 6 at 35°C with 5% NaCl added to the medium. The comparative analysis of bacterial proteomes under these diverse in vitro exposures provided evidence of metabolic reshaping and highlighted strain-specific and environmental-specific variations in critical bacterial functions.

Project description:We analyzed the transcriptional profile of P.aeruginosa PA14 grown under 14 different environmental conditions. These included conditions of growth within biofilms, at various temperatures, osmolarities and phosphate concentrations, under anaerobic conditions, attached to a surface and conditions encountered within the eukaryotic host. We found that >30% of the PA14 genome was differentially regulated at least under one of the 14 environmental conditions (referred to as the adaptive transcriptome). Most of the genes were also differentially regulated upon sigma factor hyper expression and/or inactivation (GEO accession number GSE54999) and many of those belonged to primary alternative sigma factor regulons.

Project description:Real-time reverse transcription PCR (RT-qPCR) has emerged as an accurate and widely used technique for expression profiling of selected genes. However, obtaining reliable measurements depends on the selection of appropriate reference genes for gene expression normalization. The aim of this work was to assess the expression stability of 15 candidate genes to determine which set of reference genes is best suited for transcript normalization in citrus in different tissues and organs and leaves challenged with five pathogens (Alternaria alternata, Phytophthora parasitica, Xylella fastidiosa and Candidatus Liberibacter asiaticus). We tested traditional genes used for transcript normalization in citrus and orthologs of Arabidopsis thaliana genes described as superior reference genes based on transcriptome data. geNorm and NormFinder algorithms were used to find the best reference genes to normalize all samples and conditions tested. Additionally, each biotic stress was individually analyzed by geNorm. In general, FBOX (encoding a member of the F-box family) and GAPC2 (GAPDH) was the most stable candidate gene set assessed under the different conditions and subsets tested, while CYP (cyclophilin), TUB (tubulin) and CtP (cathepsin) were the least stably expressed genes found. Validation of the best suitable reference genes for normalizing the expression level of the WRKY70 transcription factor in leaves infected with Candidatus Liberibacter asiaticus showed that arbitrary use of reference genes without previous testing could lead to misinterpretation of data. Our results revealed FBOX, SAND (a SAND family protein), GAPC2 and UPL7 (ubiquitin protein ligase 7) to be superior reference genes, and we recommend their use in studies of gene expression in citrus species and relatives. This work constitutes the first systematic analysis for the selection of superior reference genes for transcript normalization in different citrus organs and under biotic stress.

Such consideration on control systems design, specifically on AGV position control, can improve the controller accuracy. Project description:The present experimental study on permeability characteristics for raw coal under different stress states is implemented by applying the triaxial self-made 'THM coupled with servo-controlled seepage apparatus for gas-containing coal'; the result indicates that the flow rate of gas in the coal sample gradually decreases with the nonlinear loading of axial pressure and increases with the nonlinear unloading of axial stress and confining pressure. The flow rate, axial stress and confining pressure curves all satisfy the negative exponential function relation. When the sample reaches the peak intensity, the sample will be destroyed and the stress will drop rapidly; then the flow rate of the sample will increase rapidly. At this stage, the flow rate and axial strain show an oblique 'v' pattern. The flow rate of the coal sample increases nonlinearly with the increase of gas pressure; the relation curve between flow rate and gas pressure satisfies the power function relation. Under the same confining pressure and gas pressure conditions, the larger the axial stress, the smaller the flow rate of the coal sample. Under the same axial stress and gas pressure conditions, the flow rate of the coal sample will first decrease, but then increase as the confining pressure decreases. During the post-peak loading and unloading process, the flow rate of the coal sample will decrease with the loading of confining pressure but increase with the unloading of confining pressure, and there will be an increase in wave shape with the increase in axial strain. The flow rate of each loading and unloading confining pressure is higher than that of the previous loading and unloading confining pressure. At the post-peak stage, the relation curve between the flow rate of the coal sample and the confining pressure satisfies the power function relation in the process of loading and unloading confining pressure.

Project description:This study uncovers the molecular processes governing the adaptive evolution of multidrug-resistant (MDR) pathogens without antibiotic pressure. Genomic analysis of MDR Acinetobacter baumannii cells cultured for 8000 generations under starvation conditions (EAB1) or nutrient-rich conditions (EAB2) revealed significant genomic changes, primarily by insertion sequence (IS)-mediated insertions and deletions. Only two Acinetobacter-specific prophage-related deletions and translocations were observed in the EAB1 strain. Both evolved strains exhibited higher virulence in mouse infection studies, each with different modes of action. The EAB1 strain displayed a heightened ability to cross the epithelial barrier of human lung tissue, evade the immune system, and spread to lung tissues, ultimately resulting in cellular mortality. In contrast, the EAB2 strain strongly attached to epithelial cells, leading to increased synthesis of proinflammatory cytokines and chemokines. The genomic alterations and increased virulence observed in evolved strains during short-term evolution underscore the need for caution when handling these pathogens, as these risks persist even without antibiotic exposure.
