Single cell RNA-sequencing of testes from control and Brca1 germ cell-specific knockout mice

ABSTRACT: To examine the role of BRCA1 during the formation of undifferentiated spermatogonia, we generated Brca1 germ cell-specific knockout mice, in which BRCA1 was undetectable when undifferentiated spermatogonia started to form at PD1. To analyze the defects comprehensively, we isolated testes from one pair of control and Brca1 germ cell-specific knockout mice male mice at PD7 and performed single-cell RNA sequencing (scRNA-seq) analyses using the 10× Genomics platform. In the study, we found that BRCA1 loss indeed disrupts early postnatal germ cell development.

Regulation of gene expression in undifferentiated spermatogonia by DDX5

Project description:The maintenance of spermatogenesis in adult males is dependent on a population of mitotic germ cells with self-renewal potential known as undifferentiated spermatogonia. Regulation of undifferentiated spermatogonia function is dependent on transcriptional and post-transcriptional mechanisms. We have identified an essential role for the RNA helicase DDX5 in undifferentiated spermatogonia through generation of a UBC-CreERT2;Ddx5flox/flox mouse model that allows tamoxifen-dependent Ddx5 ablation. To identify genes regulated by DDX5, we generated lines of cultured undifferentiated spermatogonia from these mice and treated cells with tamoxifen (TMX) to induce Ddx5 knockout or vehicle (VEH) as a control, and performed RNA-sequencing analysis to compare these conditions.

RNA-binding protein IGF2BP1 is required for spermatogenesis in an age-dependent manner

Project description:Post-transcriptional regulation mediated by RNA binding proteins is crucial for male germline development. Insulin-like growth factor 2 mRNA-binding protein 1 (IGF2BP1), an RNA binding protein, is specifically expressed in human and mouse male gonads and is involved in manifold biological processes and tumorigenesis. However, the function of IGF2BP1 in mammalian spermatogenesis remains poorly understood. Herein, we generated an Igf2bp1 conditional knockout mouse model using Nanos3-Cre. Germ cell deficiency of Igf2bp1 in mice caused spermatogenic defects in an age-dependent manner, resulting in decreased numbers of undifferentiated spermatogonia and increased germ cell apoptosis. Immunoprecipitation-mass spectrometry analysis revealed that ELAV-like RNA binding protein 1, a well-recognized mRNA stabilizer, interacted with IGF2BP1. Single cell RNA-sequencing showed distinct mRNA profiles in spermatogonia from conditional knockout versus wide type mice. Further research showed that IGF2BP1 plays a vital role in the modulation of spermatogenesis by regulating Lin28a mRNA, which is essential for clonal expansion of undifferentiated spermatogonia. Thus, our results highlight the crucial effects of IGF2BP1 on spermatogonia for the long-term maintenance of spermatogenesis

RNA-seq analysis of Control and Mps1sKO testis at P10

Project description:Monopolar spindle 1 (Mps1), which plays a critical role in somatic mitosis, has also been revealed to be essential for meiosis I in oocytes. Spermatogenesis is an important process involving successive mitosis and meiosis, but the function of Mps1 in spermatogenesis remains unclear. Here, we generated Mps1 conditional knockout mice and found that Ddx4-cre-driven loss of Mps1 in male mice resulted in depletion of undifferentiated spermatogonial cells and subsequently of differentiated spermatogonia and spermatocytes. In addition, Stra8-cre-driven ablation of Mps1 in male mice led to germ cell loss and fertility reduction. Spermatocytes lacking Mps1 were blocked at the zygotene-to-pachytene transition in prophase of meiosis I, and the expression of many meiotic genes was decreased, while that of apoptotic genes was increased. Moreover, we also detected increased apoptosis in spermatocytes with Mps1 knockout, which may have been the reason why germ cells were lost. Taken together, our findings indicate that Mps1 is required for mitosis of gonocytes and spermatogonia, differentiation of undifferentiated spermatogonia, and progression of meiosis I in spermatocytes.

Chromatin remodeler CHD4 establishes chromatin states required for ovarian reserve formation, maintenance, and germ cell survival [ATAC-seq]

Project description:To understand the functional role of CHD4, a member of the Nucleosome Remodeling and Deacetylase (NuRD) complex, in establishing chromatin states in the formation and maintenance of ovarian reserve and the maintenance of female and male germ cells, we eliminated CHD4 function in germ cells using Ddx4-Cre and Gdf9-iCre to generate CHD4 conditional knockout (Chd4 DcKO and GcKO) mice. We discover that CHD4 defines the chromatin state that ensures germ cell survival, thereby enabling the long-term maintenance of female and male germ cells. We then performed ATAC-seq in Chd4 Dctrl and Chd4 DcKO non-growing oocytes and undifferentiated spermatogonia.

Murine Spermatogonial Stem Cells: Rapamycin- vs Vehicle-exposed in vivo (2-wk treatment)

Project description:Male FVB strain mice aged 12-days-old through 26-days-old were administered daily intraperitoneal injections of rapamycin (4mg/kg body weight) or control vehicle (5% Tween-80, 5% PEG-400), beginning at postnatal day (P)12. Mice were euthanized at P26 and their testes were isolated for germ cell enrichment. Single cell suspensions of germ cells were prepared from isolated testes and subjected to magnetic-activated cell sorting (MACS). This procedure enriches the undifferentiated spermatogonia fraction, which represents the spermatogonial stem cell (SSC) population. Total RNA from cells double-positive for the SSC surface markers thymus cell antigen 1, theta (THY1) and glial cell line-derived neurotrophic factor family receptor alpha 1 (GFRA1) was isolated for gene expression microarray analysis. Magnetic-activated cell sorting (MACS) was used to enrich undifferentiated spermatogonia from a pool of primary testicular cells isolated from 5 littermate rapamycin (RM)-treated male mice and from 5 littermate control vehicle (VEH)-treated male mice. With MACS-enriched cells from pool generating RNA, no technical replicates performed.

Identification of gene targets of the transcription factor SALL4 in undifferentiated spermatogonia

Project description:Sustained spermatogenesis in adult males and recovery of fertility following germ cell depletion are dependent on undifferentiated spermatogonia with self-renewal potential. We have previously demonstrated a critical role for the transcription factor Spalt-like 4 (SALL4) in spermatogonial differentiation. However, it remains unclear whether SALL4 has broader roles within the spermatogonial pool despite its ability to co-regulate genes with PLZF, a transcription factor required for undifferentiated cell maintenance. To identify genes regulated by SALL4 in the male germline, we established cultures of undifferentiated spermatogonia from a Sall4 inducible knockout mouse model. Cells were treated with vehicle (as control) or tamoxifen to induce gene deletion, then cells harvested and analysed by microarray to identify genes mis-expressed upon loss of SALL4.

Self-renewal of undifferentiated spermatogonia is dependent on a complex network of transcriptional and post-transcriptional regulatory factors. DDX5 is known to act as a transcriptional co-regulator in other systems through interactions with key transcription factors. Here, we performed ChIP-Seq to identify target genes of DDX5 in cultured undifferentiated spermatogonia in order to elucidate its role in maintenance of the male germline.

Non-cell Autonomous Regionalization of Sertoli Valve Epithelia in the Terminal Segment of the Seminiferous Tubules of Mouse Testes

Project description:In mammalian testes, undifferentiated spermatogonia undergo meiotic differentiation in high retinoic acid (RA) signaling states, while their undifferentiated states are maintained by fibroblast growth factors (FGFs) and glial cell line-derived neurotrophic factor (GDNF), the major niche factors for spermatogonial stem/progenitor cells, in the basal compartment of the convoluted seminiferous tubules (STs). In the valve-like terminal end of the STs adjacent to the rete testis (RT), the Sertoli valve (SV) epithelia constitutively lack most spermatogenesis activity due in part to high GDNF expression, leading to the SV-specific enrichment of undifferentiated spermatogonia; however, the molecular and cellular characteristics of the SV epithelia are unclear. By performing a Sertoli cell ablation/replacement experiment using a mouse diphtheria toxin receptor-mediated system, we show here that the SV epithelia are non-cell autonomously specified, together with having high AKT phosphorylation, in the Sertoli cells located adjacent to the RT epithelia. RNA analyses revealed constitutively high expression of Cyp26a1, which encodes an RA-degrading enzyme, in the SV region, together with high expression of Fgf9 in the adjacent RT. Exogenous in vivo RA treatment induced meiotic differentiation of the spermatogonia located within the SV region, leading to disruption of the SV structure by the ectopic appearance of adluminal meiotic germ cells in some severely affected STs. We therefore propose that the regionalization and inactive spermatogenesis of SV epithelia are mediated in part by low RA signaling, together with high FGF9 signals in the terminal end of the STs.
