Project description:Aspergillus fumigatus mutant strains were collected from bronchoalveolar lavage fluids (BALFs) during acute mouse infection (4 and 12 hours). Transcriptional analysis were conducted by comparison of each mutant with the wild type strain (CEA17) in a dye swap experiment. Besides, we performed the transcriptional profile of akuB(KU80) mutant compared to CEA17 wild type strain. All the strains were recovered from bronchoalveolar lavage fluid from CD1 mice infected with 10e8 spores for 4 and 12 hours.

Proteomic analysis of humoral immune components in bronchoalveolar lavage of patients infected or colonized by Aspergillus fumigatus

Project description:Humoral immune factors have been individually studied in the context of host-Aspergillus fumigatus interaction. However, a global view of the multitude and complex nature of humoral factors is needed to bring new insight into the pathobiology of aspergillosis. Therefore, we undertook proteomic analysis of A. fumigatus-infected or colonized bronchoalveolar lavage fluid versus controls, to identify alveolar humoral factors affected upon A. fumigatus infection. Complement proteins C1q, C8 beta chain, factor-H, surfactant protein-D, ficolin-1, ficolin-2, mannan binding lectin serine peptidase 2 and pentraxin-3 were identified as the major candidates. We hypothesize that crosstalk between these factors is essential during host-Aspergillus interaction.

Project description:To investigate the influence of Aspergillus fumigatus on iron regulation in macrophages, we obtained macrophages in culture from human derived monocytes and co-cultured the monocyte-derived macrophages with Aspergillus conidia at a 1:1 ratio. We collected samples at 0, 2, 4, 6 and 8 hours and extracted RNA. Gene expression profiling of bronchoalveolar lavage cells during Aspergillus colonization of the lung allograft

Project description:Background: Aspergillus colonization after lung transplant is associated with an increased risk of chronic lung allograft dysfunction (CLAD). We hypothesized that gene expression during Aspergillus colonization could provide clues to CLAD pathogenesis. Methods: We examined transcriptional profiles in 3 or 6-month surveillance bronchoalveolar lavage fluid cell pellets from recipients with A. fumigatus colonization (n=12) and without colonization (n=10). Among the Aspergillus colonized, we also explored profiles in those who developed CLAD (n=6) or remained CLAD free (n=6). Transcription profiles were assayed with the HG-U133 Plus 2.0 microarray (Affymetrix). Statistical Analysis: Differential gene expression analyses were performed to select candidate lists of genes. Functional analyses on these selected genes were explored.
