Effect of knockdown of RPLP0P2 on gene expression in RKO cells

ABSTRACT: We found that RPLP0P2 is subject to m6A modification. In addition, knockdown of METTL3 decreased the RNA level of RPLP0P2, suggesting that m6A modification may promote RPLP0P2 RNA stability. Furthermore, RNA pulldown assay indicated that the reader of m6A modified RPLP0P2 was IGF2BP2, and mutation of m6A modified site of RPLP0P2 destroyed its interaction with IGF2BP2. Finally, RPLP0P2 activated Notch signaling pathway and promoted the stemness of CRC cells.
