ABSTRACT: Pokmeon is an oncogenic transcription factor involved in cell growth, differentiation, and oncogenesis,but its regulatory network in breast cancer cells remians elusive . We used microarray to determine the down stream target genes of pokemon by using the MCF-7 cell line with stable ectopic expression of pokemon.

Project description:Erythroid differentiation-associated gene (EDAG), a hematopoietic tissue-specific transcription regulator, plays a key role in maintaining the homeostasis of hematopoietic lineage commitment. However, the mechanism and genes regulated by EDAG remain unknown.Here, we performed genome-wide parallel expression analyses of 32D cells stably transfected with EDAG. Total RNA from the control 32D cells and 32D/EDAG cells were used to generate target cDNA, and then hybridized to 36k Mouse Genome Array Genechips, representing about 25000 characterized murine genes.

Transcriptome analysis of gene expression in MCF-7 cell upon circFAT1(e2) depletion

Project description:In previous study, we found circFAT1(e2) was highly expressed in breast cancer tissues and cell lines. And this factor is also a crucial role in breast cancer genesis and development. So in this study, we knocked down circFAT1(e2) in MCF-7 cells. The viability and metastasis was significantly inhibited with circFAT1(e2) depletion. In order to explore the potential down-stream gene of circFAT1(e2) in breast cancer cells. We extracted the total RNA from circFAT1(e2) knockdown MCF-7 cells and negative control group by using TriZol reagent. Then, toal RNA was subjected to transcriptome sequencing to determine the potential down-stream targets of circFAT1(e2). We expect to reveal the underlied mechanism of the regulatory effect on breast cancer of circFAT1(e2).

Development of the cell lines has been previously reported by Zhou et al. (PMID: 15448698). Dataset includes 3 replicate cultures of MCF-7-Snail cells and 3 replicate cultures of MCF-7-control cells
