Project description:Proteomic characterization of skeletal muscle biopsies collected from the vastus lateralis of 44 male and female human subjects. Research subjects were divided in three different groups based on their individual exercise backgrounds and physical performance testing: 1) endurance trained (males; ME, n = 9 and females; FE, n = 9, with at least 15 years’ of regular training experience), 2) strength-trained males (MS, n = 9, with at least 15 years’ of regular training experience), and 3) age-matched healthy untrained controls (males, MC, n = 9 and females FC, n = 8, with a self-reported history of <2 exercise bouts per week over the past 15 years).

Project description:Phosphorylation is important in skeletal muscle development, growth, regeneration, and contractile function. Alterations in the skeletal muscle phosphoproteome due to aging have been reported in males; however, studies in females are lacking. We have demonstrated that estrogen deficiency decreases muscle force which correlates with decreased myosin regulatory light chain phosphorylation. Thus, we questioned whether the decline of estrogen in females that occurs with aging might alter the skeletal muscle phosphoproteome. C57BL/6J female mice randomly assigned to a sham-operated (Sham) or ovariectomy (Ovx) group to investigate the effects of estrogen deficiency on skeletal muscle protein phosphorylation in a resting, non-contracting condition. After 16 weeks of estrogen deficiency, the tibialis anterior muscle was dissected and prepped for label-free nano-liquid chromatography tandem mass spectrometry phosphoproteomic analysis. We identified 4,780 phosphopeptides in tibialis anterior muscles of ovariectomized (Ovx) and Sham-operated (Sham) control mice. Further analysis revealed 647 differentially regulated phosphopeptides (Benjamini – Hochberg adjusted p-value < 0.05 and 1.5-fold change ratio) that corresponded to 130 proteins with 22 proteins differentially phosphorylated (3 unique to Ovx, two unique to Sham, six upregulated, and 11 downregulated). Differentially phosphorylated proteins associated with the sarcomere, cytoplasm, and metabolic and calcium signaling pathways were identified. Our work provides the first global phosphoproteomic analysis in females and how estrogen deficiency impacts the skeletal muscle phosphoproteome.

Project description:Global microarray (HG U133 Plus 2.0) was used to investigate the basal level skeletal muscle transcriptome profile of young and old adults. One vastus lateralis muscle biopsy was obtained in the basal state from 36 different subjects. A total of 36 subjects were included in this investigation. The young (n=15, 25±1y) participants included 7 males and 8 females. The old (n=21, 78±1y) participants included 10 males and 11 females. All subjects were healthy and had never been involved in any formal exercise. Skeletal muscle biopsies were obtained from the vastus lateralis in the basal state. The RNA integrity of all 36 samples was very good with an average RIN number of 8.2. All samples were analyzed on a separate microarray chip, and samples were not pooled between subjects.

Project description:Females with heart failure and reduced ejection fraction (HFrEF) have greater physical limitations and a lower quality of life compared to males, however, the role of non-cardiac mechanisms remains poorly resolved. We hypothesized that differences in skeletal muscle pathology between males and females with HFrEF may explain clinical heterogeneity. In this study, we performed an unbiased RNA sequencing of 5 male controls, 5 female controls, 6 male HFrEF patients, and 5 female HFrEF patients.

Project description:Skeletal muscle is an important organ for health and movement, largely driven by specific muscle fibres. However, the assessment of fibre-type-specific DNA methylation in human skeletal muscle has been limited. We generated methylome profiles of Type I muscle fibres (T1), Type II muscle fibres (T2) and whole skeletal muscle (WM) from males and females before and after 12 weeks high intensity interval training. We compared the TI and TII pooled fibre samples (per individual) and whole muscle samples at the baseline timepoint and generated TI, TII and WM profiles of human skeletal muscle from both males and females.

Project description:The molecular mechanisms underlying the sex differences in human muscle morphology and function remain to be elucidated. The purpose of the study was to detect the sex differences in the skeletal muscle transcriptome in both the resting state and following anabolic stimuli, resistance exercise. We used microarrays to profile the transcriptome of the biceps brachii of young men and women who underwent an acute unilateral RE session following 12 weeks of progressive training. Bilateral muscle biopsies were obtained either at an early (4h post-exercise) or late recovery (24h post-exercise) time point. Muscle transcription profiles were compared in the resting state between men (n=6) and women (n=8), and in response to acute RE in trained exercised vs. untrained non-exercised control muscle for each sex and time point separately (4h post-exercise, n=3 males, n=4 females; 24h post-exercise, n=3 males, n=4 females). A logistic regression-based method (LRpath), following Bayesian moderated t-statistic (IMBT), was used to test gene functional groups and biological pathways enriched with differentially expressed genes.

Project description:We simultaneously extracted DNA and protein from >200 single fibres per participant (7 males, 5 females) to directly compare methylome and proteome profiles of Type I and Type II human skeletal muscle fibres. Using the EPICv2 array and LC-MS/MS, we identified 5,689 robust DMRs (FDR <0.001) and 338 DEPs (FDR <0.001), revealing strong inverse correlations between methylation and protein abundance in key contractile and metabolic genes. We also built a TI/TII DNA methylation reference matrix and implemented fibre-type deconvolution (MyoTYPE) for whole-muscle samples, all accessible via the MyoMETH web resource.

Project description:Aging impairs skeletal muscle insulin sensitivity, yet the underlying molecular mechanisms remain incompletely understood. Here, we examined how short-term caloric restriction (CR) alters muscle insulin action and signaling in aged rats (22 months old) of both sexes. After 8 weeks of CR or ad libitum feeding, we assessed insulin-stimulated glucose uptake (ISGU) ex vivo and performed deep phosphoproteomic profiling. CR enhanced ISGU in both sexes, with a more pronounced effect in females. Insulin-regulated phosphoproteomic responses were concordant between males and females, yet females exhibited more robust and widespread phosphorylation changes, potentially explaining their greater ISGU. CR induced extensive remodeling of basal muscle phosphorylation, particularly within structural and contractile pathways. Personalized phosphoproteomic analysis identified insulin-responsive sites on Leiomodin-1 (Lmod1) and EH domain-binding protein 1-like protein 1 (Ehbp1l1) that correlated with ISGU across individuals. Notably, Lmod1demonstrated strong genetic association with glycemic traits in humans, and several regulated sites overlapped with phosphosites observed in human clamp studies, reinforcing their translational relevance. Collectively, these findings reveal sex-dependent and diet-sensitive phosphosignaling mechanisms that modulate muscle insulin responsiveness during aging and nominate candidate regulatory nodes for future study.
