Project description:Mesenchymal stem/stromal cells (MSCs) have great potential in regenerative medicine owing to their multilineage differentiation capacity. However, tissue-derived MSCs (tMSCs) exhibit inconsistent characteristics. Although induced pluripotent stem cell (iPSC)-derived MSCs (iMSCs) are a potential solution, the effect of different embryonic lineages on their properties remains unknown. We generated MSCs from human iPSCs via five lineage-specific routes: cranial neural crest, trunk neural crest, paraxial mesoderm (somite), lateral plate mesoderm, and limb mesenchyme. All types met established MSC criteria yet differed in morphology, proliferation, and differentiation capacity. Somite-, cranial neural crest–, and limb mesenchyme–derived MSCs showed higher osteogenic potential, whereas somite-derived MSCs also showed high chondrogenic potential but were prone to hypertrophy. Limb mesenchyme–derived MSCs showed the highest adipogenic potential. Transcriptomic profiles indicated distinct clusters within iMSCs. Despite variances, a high correlation level existed between iMSCs and tMSC. Therefore, iMSCs are potential alternatives to tMSCs in regenerative medicine.

Project description:Mesenchymal stem cells (MSCs) participate in the repair/remodeling of many tissues, where MSCs commit to different lineages dependent on the cues in the local microenvironment. Here we show that TGFβ-activated RhoA/ROCK functions as a molecular switch regarding the fate of MSCs in arterial repair/remodeling after injury. MSCs differentiate into myofibroblasts when RhoA/ROCK is turned on, endothelial cells when turned off. The former is pathophysiologic resulting in intimal hyperplasia, whereas the latter is physiological leading to endothelial repair. Further analysis revealed that MSC RhoA activation promotes formation of an extracellular matrix (ECM) complex consisting of connective tissue growth factor (CTGF) and vascular endothelial growth factor (VEGF). Inactivation of RhoA/ROCK in MSCs induces matrix metalloproteinase-3-mediated CTGF cleavage, resulting in VEGF release and MSC endothelial differentiation. Our findings uncover a novel mechanism by which cell-ECM interactions determine stem cell lineage specificity and offer additional molecular targets to manipulate MSC-involved tissue repair/regeneration. Mouse bone marrow MSCs were purchased from Texas A&M Health Science Center College of Medicine Institute for Regenerative Medicine. All MSCs were cultured in αMEM supplied with 10% FCS. The transfection of DNA plasmids were performed with Lipofectamine® LTX with Plus Reagent (Life Technologies). Myc-L63RhoA or empty vector (control) was transfected into mouse bone marrow MSCs. Three days later, the cells were harvested and total RNA was isolated using RNeasy Mini kit (Qiagen). A total of four independent experiments were performed. RNA samples were assessed for quality and integrity using Synergy HT (Biotek, Winooski, VT). Microarray expression profiles were generated using the Illumina MouseRef-8 v2.0 Expression BeadChip (Illumina, San Diego, CA). Biotin-labeled cRNA was synthesized by the total prep RNA amplification kit from Ambion (Austin, TX). cRNA was quantified and normalized to 75 ng/µl, and then 1µg was hybridized to Beadchips. The hybridized chip was scanned using the Illumina iScan system and background corrected signal intensities were extracted using the GenomeStudio software (Illumina). The lumi R package was used to transform the data using a Variance Stabilizing Transformation (VST) and normalized using quantile normalization. Differential expression analysis was performed using the R/Mannova package. P-values were calculated by performing 1000 permutations, then corrected for multiple comparisons by false-discovery rate (FDR) transformation, using a 20% FDR cutoff.

Here we demonstrate that we can expand cells with characteristics of cardiovascular progenitor cells from the MSC population of human fetal hearts with only minor fluctuations over time in culture (from day 15 to day 48). We used microarray to compare the gene-expression profile of cultured human fetal cardiac MSCs over time (from day 15 to day 48). MSCs from human fetal hearts were cultured on GelTrex in a defined medium stimulating the canonical Wnt/beta-catenin pathway. Samples from three different time points (day 15, 27 and 48) were compared on microarray.

Project description:Mesenchymal stem cells (MSCs) are widely used in regenerative medicine, including the treatment of pulmonary fibrosis. However, implanted MSCs disappear within days, constraining therapeutic efficacy, which is largely attributed to nutrient deprivation. In this study, we established glycogen metabolism engineering strategies in mammalian cells. By expressing a functionally optimized glycogen synthase (GYSmut), MSCs could accumulate large amounts of glycogen rapidly as a reserve substance. We performed RNA-seq on engineered MSCs to understand its effects.

Project description:This study aimed to investigate the effects of electrical stimulation (ES) on the secretome of mesenchymal stem cells (MSCs). Using quantitative proteomics, we identified a total of 435 proteins secreted by MSCs and ES-treated MSCs (epMSCs), among which 418 were common between groups, with 14 and 8 unique to the epMSC and MSC groups, respectively. Differential expression analysis revealed that 42 proteins were upregulated and 49 were downregulated in the epMSC secretome. Functional enrichment analyses indicated that ES enhances the pro-angiogenic properties of MSCs by modulating pathways such as VEGFA-VEGFR2 signaling and the complement cascade. These findings provide insight into how ES alters MSC-derived secretomes to promote vascularization, which may have therapeutic relevance in regenerative medicine.

Project description:Human mesenchymal stromal cells (MSCs) hold great regenerative medicine potential due to their pluripotency and paracrine functions. However, MSCs lose stemness and immunomodulatory capabilities in two-dimensional (2D) culture, which differs from the natural MSC niche. We investigated methods to enhance MSC characteristics by employing an in vivo-like culture using functional polymers. Implementing FP003 polymer-based 3D culture, we observed increased expression of stemness markers (Oct4, Nanog), in hMSCs compared to 2D culture on regular plastic. This 3D environment also enhanced expression COX2 and HO1, known for their immunomodulatory functions. MSCs cultured in 3D exhibited higher secretion of PGE2 and effectively suppressed TNF- release from LPS-stimulated splenocytes, surpassing 2D-cultured MSCs. To explore therapeutic potential in vivo, we administered 3D-cultured MSCs via intravenous injection in a mouse model of neuroinflammation. Remarkably, this approach significantly reduced the expression of Iba1 and GFAP compared to 2D-cultured MSC injection. RNA-seq analysis revealed upregulated adhesion-related genes, including ITGA2, in MSCs cultured in 3D. Ectopic ITGA2 expression notably enhanced immunomodulatory function. In summary, our findings demonstrate that an in vivo mimetic 3D culture provides a beneficial microenvironment for MSCs, enhancing their immunomodulatory function through ITGA2 expression. Engineered MSCs can thus be utilized as an effective cell therapy source.

Project description:Mesenchymal stromal cells (MSCs) have been introduced as promising cell source for regenerative medicine. Besides their multilineage differentiation capacity, MSCs release a wide spectrum of bioactive factors. This secretome holds immunomodulatory and regenerative capacities. In intervertebral disc (IVD) cells, application of MSC secretome has been shown to decrease the apoptosis rate, induce proliferation and promote production of extracellular matrix (ECM). For clinical translation of secretome-based treatment, characterization of the secretome composition is needed to better understand the induced biological processes and identify potentially effective secretomes. Methods: This study aimed to investigate the proteome released by bone marrow derived MSCs following exposure to a healthy, traumatic, or degenerative human IVD environment by mass spectroscopy and quantitative immunoassay analyses. Exposure of MSCs to the proinflammatory stimulus interleukin 1β (IL-1b) was used as control.

Project description:Human mesenchymal stem cell-derived small extracellular vesicles(MSC-sEVs) have exhibited immunomodulatory and pro-regenerative properties. However, the absence of quantifiable and specific markers for defining and qualifying human MSC-sEVs impedes their translation into clinical practice. Here, we ranked the relative abundances of proteins representing unique signature for MSCs in the proteomic datasets of MSC-sEVs and chose several proteins with high relative abundances as candidate markers for MSC-sEVs. This work would facilitate the establishment of quantifiable, reproducible and standardized assays for defining MSC-sEV preparations, ultimately accelerating the translation of MSC-sEVs into clinical practice.

Project description:Therapeutic benefits of mesenchymal stem/stromal cells (MSCs) are now widely believed to come from their paracrine signalling, i.e. secreted factors such as cytokines, chemokines, and extracellular vesicles (EVs). Cell-free therapy using EVs is an active and emerging field in regenerative medicine. The cellular environment of MSCs is of critical importance when directing paracrine activity. Typical 2D cultivation of stem cells on tissue culture plastic is far removed from the physiological environment of MSCs. The application of 3D cell culture allows MSCs to adapt to their cellular niche environment which, in turn, influences their paracrine signalling activity. In this study we evaluated the impact of 3D MSCs culture on EVs secretion and cargo proteome composition and functional assessment. The outcome highlights critical differences between MSC-EVs obtained from different culture microenvironments, which should be considered when scaling up MSC culture for clinical manufacturing.
