Molecular mechanism of Forsythia suspensa glycoside promoting innate immunity by activating ER stress.

ABSTRACT: Forsythia suspensa glycoside is a phenylethanoid glycoside extracted from the bark, leaves, flowers, fruits, and seeds of the Oleaceae plant Forsythia suspensa. As a major active ingredient in the Chinese herbal medicine Forsythia suspensa, it has a wide range of pharmacological activities. It not only inhibits various Gram positive and negative bacteria that cause infections, but also has strong antiviral effects. However, the role of Forsythia suspensa glycosides in innate immunity is not yet clear.

Effects of Cardiac Glycosides on RNA Expression in Prostate Cancer LNCaP-abl Cells

Project description:Prostate cancer is the most common cancer in men and cardiac glycosides inhibit prostate cancer cell proliferation. In order to investigate the mechanism by which cardiac glycosides inhibit prostate cancer cells, we observed genome-wide RNA expression in prostate cancer LNCaP-abl cells, hormone resistant cells, after the cardiac glycoside treatment using RNA-Seq. In addition, we profiled LNCaP-abl cells after androgen receptor (AR) knockdown to observe whether cardiac glycoside effect on RNA expression is similar to that of AR knockdown. Observation of three cardioglycosides, Digoxin, Peruvoside and Strophanthidin, and AR knockdown regulated RNA expression in LNCaP-abl with RNA-Seq (each triplicates)

Functional exploration of novel glycoside hydrolases in Fervidibacter sacchari PD1T

Project description:Few aerobic hyperthermophiles degrade polysaccharides. We describe the genome-enabled enrichment and isolation of an aerobic hyperthermophile, Fervidibacter sacchari, which was originally ascribed to candidate phylum Fervidibacteria. F. sacchari uses polysaccharides and monosaccharides as sole carbon sources from 65-87.5 °C, and its genome encodes 117 glycoside hydrolases (GHs) spanning 49 GH families, including 31 homologs of understudied GH109, GH177, and GH179 domains. Here, we analyzed the transcriptomes of F. sacchari cells grown on eight different sole carbon and energy sources (beta-glucan, chondroitin sulfate, corn stover, gellan gum, locust bean gum, starch, xanthan gum, and xyloglucan) to link glycoside hydrolase substrate to function, as well as identify potential regulatory mechanisms. These data will provide preliminary characterization of novel carbohydrate-active enzymes at high temperatures.

Comparative transcriptome analyses of three medicinal Forsythia species and prediction of candidate genes involved in secondary metabolisms [Forsythia viridissima]

Project description:Here, de novo transcriptome assemblies for leaf and flower tissues of Forsythias were performed, resulting in 81913 unigenes of F. suspensa, 88491 unigenes of F. viridissima and 69458 unigenes of F. koreana (F. viridissima var. koreana). Classification of the annotated unigenes by gene ontology terms and KEGG pathways was used to explore transcriptomic differences among the Forsythias. Orthogroup was introduced to compare expression levels of unigenes in tissues from different species, which unveiled that three leaf tissues of Fosythias were closely correlated based on expression values of orthologous unigenes. Showing high expression mainly in leaves of F. viridissima and F. koreana, candidate homologs for genes involved in the biosynthetic pathway of lignans and phenylethanoid glycosides were determined in these transcriptome assemblies.

Efficacy and molecular mechanism of a glycoside compound inhibiting abnormal prion protein formation in prion-infected cells

Project description:This report describes our study of the efficacy and the potential mechanism underlying the anti-prion action of a new anti-prion compound having a glycoside structure in prion-infected cells. The study revealed involvements of two factors in the mechanism of the compound action: interferon and a microtubule nucleation activator, phosphodiesterase 4D interacting protein. In particular, phosphodiesterase 4D interacting protein was suggested to be important in regulating the trafficking or fusion of prion protein-containing vesicles or structures in cells. The findings of the study are expected to be useful not only for the elucidation of cellular regulatory mechanisms of prion protein, but also for the implication of new targets for therapeutic development. Prion-infected N167 cells were treated with either anti-prion glycoside compound (Gly-9) or control glycoside compound (Gly-14) at a dose of 5 M-NM-<g/mL for three days. Then, gene expression profiles were analyzed by DNA microarray analysis. Experiments were performed in quadruplicate.

Transcriptome profiling of ovaries in aged mice administered with individual ingredients of Samul-tang

Project description:Samul-tang (SM) is a traditional multi-ingredient treatment widely used in clinical practice for improving ovarian function in women with diminished ovarian reserve. Despite its therapeutic benefits, the complexity of its mechanisms of action due to the combination of multiple compounds has limited its research integration into modern pharmacological science. To address this challenge, we identified 39 herbal compounds as major ingredients in SM and generated transcriptome data from the mice ovaries after administrating with individual ingredients. This dataset aims to provide insights into the mechanisms of action of SM and to aid in the identification of effective ingredient combinations.
