Transcript analysis in two alfalfa salt tolerance selected breeding populations relative to a non-tolerant population

Project description:With the growing limitations on arable land, alfalfa (a widely cultivated, low-input forage) is now being selected to extend cultivation into saline lands for low-cost biofeedstock purposes. Here, minerals and transcriptome profiles were compared between two new salinity-tolerant North American alfalfa breeding populations and a more salinity-sensitive Western Canadian alfalfa population grown under hydroponic saline conditions. All three populations accumulated two-fold higher sodium in roots than shoots as a function of increased electrical conductivity. At least 50% of differentially expressed genes (p < 0.05) were down-regulated in the salt-sensitive population growing under high salinity, while remaining unchanged in the saline-tolerant populations. In particular, most reduction in transcript levels in the salt-sensitive population were observed in genes specifying cell wall structural components, lipids, secondary metabolism, auxin and ethylene hormones, development, transport, signalling, heat shock, proteolysis, pathogenesis-response, abiotic stress, RNA processing, and protein metabolism. Transcript diversity for transcription factors, protein modification, and protein degradation genes was also more strongly affected in salt-tolerant CW064027 than in salt-tolerant Bridgeview and salt-sensitive Rangelander, while both saline-tolerant populations showed more substantial up-regulation in redox-related genes and B-ZIP transcripts. The report highlights the first use of bulked genotypes as replicated samples to compare the transcriptomes of obligate out-cross breeding populations in alfalfa.

Genome-wide DNA methylation profiles of Arsenic exposed subjects through drinking water in Pakistan

Project description:Ground water Arsenic (As) toxicity is a global problem and millions of people are exposed to elevated levels (more than WHO advised maximum limit of 10µg/L) through drinking water. The exposure is associated with various cancerous and non-cancerous diseases. It may alter DNA methylation profiles of inviduals and suppress the activity of various genes giving rise to different diseases. Pakistan, a developing country in South Asian region, also has reported elevated ground water As levels in various investigations since 2005. However, a very limited biomonitoring studies have been conducted in this context while no study reports molecular changes associated with drinking water As exposure in Pakistan. Within this context, the present study aimed to investigate genome-wide DNA methylation profiles of the exposed subjects in two districts of Punjab Province Pakistan, i.e Lahore and Kasur. The population was stratified into three exposure groups comprising Low, Medium and High exposure based on their urinary arsenic levels. Genome-wide DNA methylation profiles were obtained using MeDIP in combination with NimbleGen 2.1M Deluxe Promotor arrays.

Exploiting Rye in Wheat Quality Breeding: The Case of Arabinoxylan Content.

Project description:Rye (Secale cereale subsp. cereale L.) has long been exploited as a valuable alternative genetic resource in wheat (Triticum aestivum L.) breeding. Indeed, the introgression of rye genetic material led to significant breakthroughs in the improvement of disease and pest resistance of wheat, as well as a few agronomic traits. While such traits remain a high priority in cereal breeding, nutritional aspects of grain crops are coming under the spotlight as consumers become more conscious about their dietary choices and the food industry strives to offer food options that meet their demands. To address this new challenge, wheat breeding can once again turn to rye to look for additional genetic variation. A nutritional aspect that can potentially greatly benefit from the introgression of rye genetic material is the dietary fibre content of flour. In fact, rye is richer in dietary fibre than wheat, especially in terms of arabinoxylan content. Arabinoxylan is a major dietary fibre component in wheat and rye endosperm flours, and it is associated with a variety of health benefits, including normalisation of glycaemic levels and promotion of the gut microbiota. Thus, it is a valuable addition to the human diet, and it can represent a novel target for wheat-rye introgression breeding.

Mechanisms of Salt Tolerance and Molecular Breeding of Salt-Tolerant Ornamental Plants.

Project description:As the area of salinized soils increases, and freshwater becomes more scarcer worldwide, an urgent measure for agricultural production is to use salinized land and conserve freshwater resources. Ornamental flowering plants, such as carnations, roses, chrysanthemums, and gerberas, are found around the world and have high economic, ornamental, ecological, and edible value. It is therefore prudent to improve the salt tolerance of these important horticultural crops. Here, we summarize the salt-adaptive mechanisms, genes, and molecular breeding of ornamental flowering crops. We also review the genome editing technologies that provide us with the means to obtain novel varieties with high salinity tolerance and improved utility value, and discuss future directions of research into ornamental plants like salt exclusion mechanism. We considered that the salt exclusion mechanism in ornamental flowering plants, the acquisition of flowers with high quality and novel color under salinity condition through gene editing techniques should be focused on for the future research.
