Project description:Study the effects of 3 weeks training exercise in chronic obstructive pulmonary disease (COPD) patients and healthy individuals. Skeletal muscle biopsies of 5 COPD patients (aged 63±2 year) and 3 age-matched healthy individuals were studied before and after 3 weeks of a supervised endurance training program. Samples were collected from open muscle biopsies of the vastus lateralis of 5 COPD patients and 3 age-matched healthy individuals before and after 3 weeks of a supervised endurance training program. Patients and controls underwent an endurance exercise program on a cycloergometer 5 days a week for three consecutive weeks. One 1 h session/day was established in all subjects, with effective exercise periods of at least 30 min in each case.

Project description:Study the training exercise effects in chronic obstructive pulmonary disease (COPD) patients and aged-matched healthy individuals. Skeletal muscle biopsies from 9 stable COPD patients with normal fat free mass index (FFMI, 21Kg/m2) (COPDN), 6 COPD patients with low FFMI (16Kg/m2) (COPL), and 12 healthy sedentary subjects (FFMI 21Kg/m2) before and after 8 weeks of a supervised endurance exercise program were analyzed. Samples were collected from open biopsies from the musculus vastus lateralis of COPD patients and healthy individuals before and after 8 weeks of exercise training. Constant-work rate exercise at 70% of pre-training Watts peak (Wpeak) (CardiO2 cycle Medical Graphics Corporation, USA) was carried out before and after 8 weeks of supervised interval training with a cycloergometer until pre-training endurance time exhaustion. Measurements before and after training were obtained at isowork-rate and iso-time.

Plasma metabolic profile in COPD patients: effects of exercise and endurance training

Project description:The study examines plasma metabolic profiles of patients with chronic obstructive pulmonary disease (COPD) to prove whether the disease influences metabolism at rest and after endurance training. This is based on the hypothesis that metabolome levels should reflect impaired skeletal muscle bioenergetics in COPD. The study aims to test this hypothesis by evaluating plasma metabolic profiles in COPD patients before and after 8 weeks of endurance exercise training. We studied blood samples from 18 COPD patients and 12 healthy subjects. Pre- and post-training blood plasma samples at rest and after constant-work rate exercise (CWRE) at 70% of pretraining Watts peak were analyzed by 1H-nuclear magnetic resonance spectroscopy to assess metabolite profiles. The two groups presented training-induced physiological changes in the VO2 peak and in blood lactate levels (P0.01 each). Before training, the two groups also showed differences in metabolic profiles at rest (P0.05). Levels of valine (r = 0.51, P0.01), alanine (r = 0.45, P0.05) and isoleucine (r = 0.51, P0.01) were positively associated with body composition (Fat Free Mass Index). While training showed a significant impact on the metabolic profile in healthy subjects (P0.001), with changes in levels of amino acids, creatine, succinate, pyruvate, glucose and lactate (P0.05 each), no equivalent training-induced effects were seen in COPD patients in whom only lactate decreased (P0.05). This study shows that plasma metabolic profiling contributes to the phenotypic characterization of COPD patients.

Endurance training remodels sperm-borne small RNA expression and methylation at neurological gene hotspots [sncRNA]

Project description:Remodelling of the sperm epigenome by lifestyle factors before conception could account for altered metabolism in the next generation offspring. Here, we hypothesized that endurance training changes the epigenome of human spermatozoa. Using small RNA (sRNA) sequencing and Reduced Representation Bisulfite Sequencing (RRBS), we respectively investigated sRNA expression and DNA methylation in pure fractions of motile spermatozoa collected from young healthy individuals before, after 6 weeks of endurance training and after 3 months without exercise. Expression of 8 piwi interacting RNA were changed by exercise training. RRBS analysis revealed 330 differentially methylated regions (DMRs) after training and 303 DMRs after the detraining period which were, in both conditions, enriched at close vicinity of transcription start sites. Ontology analysis of genes located at proximity of DMRs returned terms related to neurological function at the trained state and, to a much lesser extent, at the detrained state. Our study reveal that short-term endurance training induces marked remodelling of the sperm epigenome, and identify genes related to the development of the central nervous system as potential hot spots for epigenetic variation upon environmental stress.

Project description:The leg of healthy volunteers was locally deconditioned using three weeks of unilateral lower limb suspension (ULLS). The extremely deconditioned legs of subjects with a spinal cord injury (SCI) were trained using eight weeks of functional electrical stimulation (FES) exercise, 2-3 times per week (total 20 sessions).
