AMIGO2 overexpression effect on colon cancer cell line

ABSTRACT: Analysis of SW480 colon cancer cells overexpressing AMIGO2. Result provide insight into the role of AMIGO2 in colon cancer. Amphoterin-inducible genes and open reading frame 2 (AMIGO2) have been reported to play a role in promoting malignant transformation in several cancer types, including human colon cancer. However, the exact role of AMIGO2 in human colorectal cancer invasiveness remains unclear. We established an AMIGO2 overexpression line (SW480_A9) and a vector control line (SW480_E11) using a human colon cancer cell line (SW480) and analyzed differential expression of AMIGO2 on a microarray (Toray 3D-Gene).

Amphoterin-Induced Gene and Open Reading Frame 2 (AMIGO2) Interacts with CAPN2 and Promotes the Proliferation and Migration of Hypopharyngeal Cancer Cell

Project description:Background: Amphoterin-induced gene and open reading frame 2 (AMIGO2) has been reported to play a detrimental role in multiple cancer types. However, the precise role of AMIGO2 in hypopharyngeal squamous cell carcinoma (HPSCC) remains elusive. Methods: Bioinformatic analysis was employed to identify the differential expression and prognostic value of AMIGO2 in the head and neck cancer. Proliferation, apoptosis, and migration assays were then conducted after short hairpin RNA-mediated knockdown of AMIGO2 in FaDu cell line and in xenograft mouse model. Co-immunoprecipitation and mass spectrometry analysis were performed to determine the protein interaction between AMIGO2 and CAPN2. Rescue experiments with respect to the overexpression of CAPN2 were also performed. RNA sequencing after AMIGO2 knockdown with and without CAPN2 overexpression were conducted, followed by pathway enrichment analysis. Results: AMIGO2 expression was up-regulated in head and neck cancer and correlated with a poor prognosis. Functionally, HPSCC cell proliferation and migration were significantly inhibited in vivo and in vitro upon AMIGO2 knockdown. AMIGO2 binds to CAPN2 in cell context, and overexpression of CAPN2 could effectively reverse the inhibition from AMIGO2 knockdown. Bioinformatics analysis based on RNA sequencing indicated that the interaction between AMIGO2 and CAPN2 might be involved in multiple signaling pathways. Conclusions: Our work suggests that AMIGO2 promotes the proliferation and migration of HPSCC by interacting with CAPN2, implying that AMIGO2 can be therapeutically targeted to treat HPSCC.

Project description:The role of SPROUTY2 (SPRY2) in human colon cancer is controversial. Our data support a tumorigenic action of SPRY2. We use microarrays to identify SPRY2 target genes in human SW480 ADH colon carcinoma cell line. RNA was obtained from SW480 ADH cells, Mock transfected or transfected with WT or two differents SPRY2 genes mutants. Hybridization was performed on Affymetrix microarrays. We performed three independent experiments. Each experiment was conducted in triplicate.

Gene expression data of QRsP-11 mouse fibrosarcoma cell line and its liver metastatic subline

Project description:Mouse QRsP-11 fibrosarcoma cells were injected into the spleens of mice to isolate sub-populations of tumor cells that colonize to the liver. Cells from liver metastatic nodules were established in vitro and subsequently injected intrasplenically for further cycles of selection. After 12 cycles of in vivo selection, the cell subline LV12 was established. Intravenous injection of LV12 cells produced liver metastases than QRsP-11 cells, whereas the incidence of lung metastases was similar to that of QRsP-11 cells. LV12 cells exhibited adhesiveness to liver-derived but not to lung-derived endothelial cells. DNA chip analysis indicated that an amphoterin-induced gene and open reading frame 2 (Amigo2) was overexpressed in LV12 cells. Knockdown of Amigo2 expression by siRNA in LV12 cells attenuated liver endothelial cell adhesion. Ex vivo imaging showed that siRNA-mediated suppression of Amigo2 in luciferase-expressing LV12 cells reduced attachment to liver tissue to the same level as that observed with QRsP-11 cells. Moreover, Amigo2-knockdown by siRNA inhibited liver metastasis. Amigo2 expression in human cancer cells was higher in liver metastatic lesions than in primary lesions of colon and gastric cancer specimens. These data indicated that Amigo2 is involved in tumor cell adhesion to liver endothelial cells and formation of liver metastases.

The colon cancer cell line SW480 does not express endogenous ER but is made ERb-expressing by lentiviral transduction of an ERb expression cassette. Introduction of ERb makes it possible to study the role and function of ERb in colon cancer as well as the impact ERb has on its own (in the absence of ERa).

Transcription profiling by array of human colon cancer stem cells derived from human colon cancer line cell, SW480

Project description:Cancer stem-like cells are defined as small population of cancer cells which has, higher tumor-initiating ability, self-renewing ability and differentiation ability. In this experiment, transcription profile of cancer stem-like cells derived from human colon cancer line cell SW480 was investigated. Cancer stem-like cells were isolated as side population (SP) cells by Hoechst33342 dye staining from human colon cancer line cell SW480. Non-cancer stem-like cells were isolated as main population (MP) cells. RNAs were isolated from SP cells and MP cells derived from SW480 cells and array experiment was perfromed in dye swaped fashion.

MicroRNA expression profile between human colorectal cancer SW480 and SW620 cell line

Project description:Two isogenic human colorectal cancer cell lines (primary SW480 cell line and its lymph node metastatic variant SW620 cell line),as an in vitro metastatic model. We have demonstrated that SW620 cell line possesses high metastasis potential and SW480 cell linepossesses low metastatic potential. We want to compare the whole cell microRNAs profiles of two isogenic colorectal cancer cell lines (SW480 and SW620 cell line), to gain an insight into the molecular events of colon cancer metastasis.

Diverse effect of WWOX overexpression in SW480 colon cancer cell line.

Project description:The WWOX gene is a tumor suppressor probably involved in regulation of cell cycle and apoptosis and downregulated in variety of cancer types.However, its role in colon cancerogenesis is unknown. The aim of this study was to characterize how WWOX may be involved in colon cancerogenesis or cancer progression, how it influences the basic cancer cell features and modifies cell expression profile.Our observations suggest that in HT29 colon cancer cell line increased expression of WWOX may result in transition of cancer cells into more normal- like colon epithelium phenotype, on the other hand in SW480 WWOX revealed the well-known tumour suppressor properties. However, as the colon cancer is very heterogeneous disease, obtained discrepancies may reflect the known differences between cell lines and cancerogenesis pathway, which they undergone. SW480 colon cancer cells were stably transfected with WWOX cDNA. SW480 cells transfected with an empty vector served as a control. Total mRNA was isolated to look for gene-expression differences induced by the WWOX overexpression.
