RNA-seq analysis of the brain of mice following recombinant HSV-1 carrying the S302A mutation in vUNG and the repaired HSV-1 strain

ABSTRACT: To investigate the relationship between the host factor APOBEC1 and the viral factor vUNG in the pathogenesis of herpes encephalitis, wild-type and Apobec1-deficient mice were infected with vUNG mutants and their repaired strains. Brain tissues were collected, and host mRNA expression was analyzed using RNA sequencing.

Raw RNAseq data of HSV-1-infected mouse skin fibroblasts

Project description:Herpes simplex viruses (HSV) are responsible for a broad variety of human diseases, including cold sores, ocular herpes and herpetic encephalitis, leading to a great burden worldwide. However, the role for sex in herpes infection is controversial and unclear due to the complexity of factors involving in the formation of HSV diseases. Herein, we ask whether there is a sexual dimorphism based on the chromosomal rather than hormonal differences in the primary skin cells during low exposure of HSV. Profiling of male and female transcriptional programs reveals that cytosolic sensing pathway is induced to a greater degree in female cells, correlated with higher yields of infectious virions in male counterparts. Female skin cells distinctively reactivate Xist, a critical component of X inactivation, to silence the expression of the transcriptional repressor on X chromosome, which thereby maintains higher viperin level and further explains the different growth phenotypes between two sexes. Collectively, we propose a model that HSV-1 triggers a sex-specific regulation of antiviral response in the cytosolic sensing signaling via the control of Xist.

Raw RNA-seq data of HSV-1-infected human cells (HFF and MRC5)

Project description:We report transcriptomic data from HSV-1-infected human cells (HFF and MRC5) Herpes simplex virus type I (HSV-1) is a common human pathogen causing cold sores, and in rare cases, severe keratitis and encephalitis. Mouse models are commonly used to study pathogenesis of HSV-1 infection due to the neurotropic properties of HSV make it hard to reach information from infected humans, but mice are not a natural host for this virus. Therefore, it is important to have insights into transcriptional regulation in human cell cultures, which gave us more information before we interpret experimental results from humans and mouse models. Herein, we provide overall transcriptomic data from two HSV-1infected cells, HFF and MRC5. We found that these two human cells downregulated many genes in an antiviral pathway characterized by interferon-stimulated genes.

Project description:Dendritic cells (DCs) regulate both innate and adaptive immune responses. The role of CD11c-plus DCs in the corneal response to to herpes simplex virus-1 (HSV-1) infection was investigated by depleting them prior to infection. Bone marrow cells from CD11c-DTR (C.FVB-Tg(Itgax-DTR/EGFP)57Lan/J) mice were transferred intravenously to irradiated BALB/cJ host mice. After 6 weeks, the bone marrow chimeric host mice were injected with diphtheria toxin (DT) to deplete CD11c-positive cells. Two days after injection, the mice were subjected to a standard corneal infection protocol using HSV-1.

Worldwide circulation of HSV-2 × HSV-1 recombinant strains.

Project description:Homo sapiens harbor two distinct, medically significant species of simplexviruses, herpes simplex virus (HSV)-1 and HSV-2, with estimated divergence 6-8 million years ago (MYA). Unexpectedly, we found that circulating HSV-2 strains can contain HSV-1 DNA segments in three distinct genes. Using over 150 genital swabs from North and South America and Africa, we detected recombinants worldwide. Common, widely distributed gene UL39 genotypes are parsimoniously explained by an initial >457 basepair (bp) HSV-1 × HSV-2 crossover followed by back-recombination to HSV-2. Blocks of >244 and >539 bp of HSV-1 DNA within genes UL29 and UL30, respectively, have reached near fixation, with a minority of strains retaining sequences we posit as ancestral HSV-2. Our data add to previous in vitro and animal work, implying that in vivo cellular co-infection with HSV-1 and HSV-2 yields viable interspecies recombinants in the natural human host.

Response to HIV-1 gp160-carrying recombinant virus HSV-1 and HIV-1 VLP combined vaccine in BALB/c mice.

Project description:Human immunodeficiency virus (HIV) induced AIDS causes a large number of infections and deaths worldwide every year, still no vaccines are available to prevent infection. Recombinant herpes simplex virus type 1 (HSV-1) vector-based vaccines coding the target proteins of other pathogens have been widely used for disease control. Here, a recombinant virus with HIV-1 gp160 gene integration into the internal reverse (IR) region-deleted HSV-1 vector (HSV-BAC), was obtained by bacterial artificial chromosome (BAC) technology, and its immunogenicity investigated in BALB/c mice. The result showed similar replication ability of the HSV-BAC-based recombinant virus and wild type. Furthermore, humoral and cellular immune response showed superiority of intraperitoneal (IP) administration, compared to intranasally (IN), subcutaneous (SC) and intramuscularly (IM), that evidenced by production of significant antibody and T cell responses. More importantly, in a prime-boost combination study murine model, the recombinant viruses prime followed by HIV-1 VLP boost induced stronger and broader immune responses than single virus or protein vaccination in a similar vaccination regimen. Antibody production was sufficient with huge potential for viral clearance, along with efficient T-cell activation, which were evaluated by the enzyme-linked immunosorbent assay (ELISA) and flow cytometry (FC). Overall, these findings expose the value of combining different vaccine vectors and modalities to improve immunogenicity and breadth against different HIV-1 antigens.
