ZEB1-mediated Epithelial to Mesenchymal Transition in Colorectal cancer HT-29 cell line

ABSTRACT: We report the induction of epithelial-mesenchymal transition (EMT) in the epithelial colorectal cancer (CRC) cell line HT-29. ZEB1 expression is sufficient to induce the downregulation of epithelial markers such as E-cadherin, claudins, keratins and occludin, while upregulating the expression of several mesenchymal markers such as vimentin and fibronectin.

Expression profiling of a prostate cancer cell line(OPCT1) and its clonal progenies with different functional characteristics

Project description:We have developed a novel spontaneous model of epithelial-to-mesenchymal transition (EMT) which involves four phenotypically distinct clones derived from a primary tumour-derived human prostate cancer cell line (OPCT-1), and its use to explore relationships between EMT and the generation of cancer stem cells (CSCs) in prostate cancer. Expression of epithelial (E-Cadherin) and mesenchymal markers (vimentin, fibronectin) revealed that two of the four clones were incapable of spontaneously activating EMT, whereas the others contained large populations of EMT-derived, vimentin-positive cells having spindle-like morphology. One of the two EMT-positive clones exhibited aggressively and stem cell-like characteristics, whereas the other was non-aggressive and had no stem cell phenotype. One of the two EMT-negative clones exhibited aggressive stem cell-like properties, whereas the other was the least aggressive of all clones. To understand this phenotypic differences between the clones we have conducted a microarray expression profiling experiment using Affymetrix platform.

Knockdown of laminin alpha 5 chain in colorectal cancer cell line HT-29

Project description:Laminin-mediated signaling via membrane receptors actively participate in most normal biological process and pathologies. Laminins involve in all step of metastatic cascade including tumor cell proliferation, migration, anoikis resistance, extravasation, epithelial to mesenchymal transition etc. The 5-containing laminins (LAMA5) are one of the three types of laminins predominantly expressed in the epithelial basement membrane in the adult intestine as well as during crypt-villus development. Likewise laminins 511 and 521 (laminin-10 and laminin-11) have been shown as important matrix component to facilitate stem cell survival and to maintain their stem cell properties. Thus, it is assuming that laminins play a role in the formation of premetastatic niches. To investigate the role of 5-containing laminins in colorectal cancer we performed the knockdown of LAMA5 gene expression in HT-29 cell line. Here we present the results of comprehensive gene expression analysis using Affymetrix microarrays in cells transduced by shRNA to LAMA5, control nonsense shRNA, and parental cells as well.

Gene expression in an EMT model using an immortalized bronchial epithelial cell line induced by TGF-β1

Project description:This study aimed to establish an epithelial-mesenchymal transition (EMT) model with an immortalized human bronchial epithelial cell line, M-BE, and to identify an EMT signature gene set. The TGF-β1-induced M-BE cells got spindle-shaped fibroblast-like morphology and lost the cell-cell contact, with down-regulated expression of epithelial marker E-cadherin and up-regulated expression of mesenchymal markers N-cadherin and Vimentin. Examined by microarray, there were 2628 genes identified as significant EMT-related, including 1490 up-regulated genes (FC > 2, fdr < 0.01) and 1138 down-regulated genes (FC < 0.5, fdr < 0.01) in TGF-β1-induced M-BE cells.
