ABSTRACT: MicroRNA-455 is a key microRNA in cartilage, strongly induced by Sox9. Overexpression of miR-455-3p in the developing chick limb bud at HH18-19 causes a reduction in limb bud size at HH24-25.

Stage-specific microRNA profiling of FACS-sorted Math1-positive commissural neurons in developing chick embryos

Project description:This study aims to systematically identify miRNAs and characterize microRNA (miRNA) expression in a specific population of Math1-positive commissural neurons in the developing chick spinal cords. To achieve this, chick embryos were electroporated with constructs encoding a Venus-tagged, enhancer-driven Math1, followed by FACS isolation of Venus-Math1-positive neurons from developing spinal cords at pre-crossing (HH18–20) stages. By performing miRNA sequencing on these sorted populations, the study seeks to uncover stage-specific miRNA expression signatures that may play critical roles in the differentiation, identity, and maturation of Math1-positive commissural neurons during spinal cord development.

Project description:During limb development, fibroblast growth factors (FGFs) govern proximal-distal outgrowth and patterning. FGFs also synchronize developmental patterning between the proximal-distal and anterior-posterior axes by maintaining sonic hedgehog (SHH) expression in cells of the zone of polarizing activity (ZPA) in the distal posterior mesoderm. SHH, in turn, maintains FGFs in the apical ectodermal ridge (AER) which caps the distal tip of the limb bud. Crosstalk between FGF and SHH signaling is critical for patterned limb development, but the mechanisms underlying this feedback loop are not well characterized. Implantation of FGF beads in the proximal posterior limb bud can maintain SHH expression in the former ZPA domain (evident 3hrs after application), while prolonged exposure (24hrs) can induce SHH outside of this domain. Although temporally and spatially disparate, comparative analysis of transcriptome data from these different populations enriched molecules required for FGF-mediated SHH regulation.

RNA-Seq analysis miR-455-3p overexpression effect on breast cancer cell line

Project description:MiR-455-3p has been reported to suppress the expression of ROCK2, HOXB5, hTERT, and RUNX2, which are involved in multiple biological processes including renal fibrosis, chondrogenic development and differentiation, Alzheimer’s disease, preeclampsia, and tumorigenesis. Here, we showed that miR-455-3p broadly inhibited TGF-beta-induced EMT by repressing the expression of Smad2, ZEB1, and HDAC2 in breast cancer cells, and that miR-455-3p loss-of-function promotes the acquisition of cell invasive properties and the ability to metastasize. These results indicate that miR-455-3p represents an important node that inhibits metastasis of breast cancer.
