Loss of SPHK1 recruits pro-tumorigenic macrophagese to drive lung adenocarcinoma

ABSTRACT: Altered lipid metabolism contributes to lung inflammation and drives lung tumorigenesis. A key factor maintaining homeostasis is Sphingosine Kinase 1 (SPHK1), which phosphorylates sphingosine to the bioactive lipid sphingosine-1-phosphate. Interruption of this synthesis pathway in lung tumor cells promotes a pro-tumorigenic microenvironment and hence drives tumor progression. To better understand this process, we performed RNAseq analysis of SPHK1 proficient and deficient mouse lung tumor cells harboring KrasG12D mutations.
