Project description:Microplastics are emerging environmental pollutants with potential biological effects on human health. However, the molecular responses of colorectal cancer tissues following microplastic exposure remain poorly characterized. In this study, we performed bulk RNA sequencing to investigate transcriptomic alterations associated with polystyrene microplastics (PS-MPs) exposure in a murine colorectal cancer model. CT26 colorectal cancer cells were subcutaneously implanted into BALB/c mice to establish xenograft tumors. After tumor formation, mice received daily intratumoral injections of 1 μm PS-MPs suspension (10 μg/mL, 50 μL/day), while control mice received no PS-MPs treatment. Tumor tissues were collected for RNA extraction and high-throughput sequencing. Differential gene expression analysis and downstream bioinformatic analyses were performed to characterize molecular changes associated with PS-MPs exposure. This dataset provides transcriptomic profiles of CT26 colorectal cancer xenograft tumors following PS-MPs exposure and may facilitate further investigation of the biological responses associated with environmental microplastic exposure.

Project description:Microplastics (MPs) have become a serious global environmental threat that causes damage to mammalian organs. In this work, we investigated the potential molecular mechanism underlying the development of liver fibrosis induced by long-term exposure to three different sized PS-MPs (80 nm, 0.5 µm and 5 µm) in mice. Liver fibrosis levels were evaluated in mice after chronic exposure to PS-MPs. Liver inflammation was mainly increased in chronic exposure to 80 nm and 0.5 µm PS-MPs. Liver lipid deposition was significantly enhanced after PS-MP exposure. However, oxidative stress was not changed under PS-MP exposure. GO enrichment and KEGG pathway analyses revealed that the DEGs and shared DEGs were mainly enriched in the metabolism of lipids. The mRNA expression levels of genes related to fatty acid oxidation, synthesis and transport were dramatically induced by PS-MP exposure. Four hub genes, Acot3, Abcc3, Nr1i3 and Fmo2, were identified by CytoHubba analysis of shared DEGs. The mRNA expression levels of three hub genes, Acot3, Abcc3 and Nr1i3, were significantly augmented under chronic PS-MP exposure. Our results suggest that Acot3, Abcc3 and Nr1i3 are potential molecules involved in the development of liver fibrosis under chronic exposure to PS-MPs.

Accumulating evidence suggested that MNPs can bioaccumulate in gonads and impair fertility in animals, yet the underlying cellular mechanisms and tissue-specific responses remain poorly understood. In this study, we employed in vivo and in vitro models to systematically investigate the impact of polystyrene microplastics (PS-MPs; 100 nm and 5 µm) on ovarian development and function in pubertal female mice. Following 35 days of exposure, we observed size-dependent reproductive toxicity, with 100 nm PS-MPs causing reduced body weight gain and ovarian size, and compromised ovarian structures and functions, including altered follicle and corpora lutea counts, alongside disrupted hormone levels. Leveraging single-cell RNA-sequencing (scRNA-seq), we uncovered profound alterations in intracellular communication networks and signaling flow patterns across seven identified ovarian cell types. Granulosa cells (GCs) were identified as the primary target PS-MPs, exhibiting marked transcriptional changes, including dysregulation of FSCN1, a critical actin cytoskeleton regulator. In vitro experiments confirmed that only 100 nm PS-MPs were internalized by GCs, leading to cell cycle arrest, necroptosis and hormonal dysfunction. Mechanistically, PS-MPs triggered F-actin cytoskeleton remodeling, increasing cell stiffness and reducing histone marks (H3K4me3, H3K27ac) associated with chromatin accessibility. Integrated ATAC-seq and RNA-seq analyses implicated STAT1 as a key transcriptional regulator driving PS-MP-induced epigenetic and transcriptional changes. Overall, this study first revealed PS-MP-mediated reproductive toxicity from a single-cell perspective, identifying granulosa cells as particularly vulnerable targets. It uncovered novel mechanisms involving cytoskeletal disruption and epigenetic reprogramming, while demonstrating size-dependent internalization patterns that directly correlate with reproductive impairment. These findings provide unprecedented insights into the molecular and epigenetic consequences of MNPs in mammalian reproduction, emphasizing the potential health risks of environmental MNPs exposure.

Project description:Aims: Polystyrene microplastics (PS-MPs) are emerging environmental pollutants, but their impact on lung cancer treatment remains unclear. This study investigates how PS-MPs affect radiotherapy efficacy in lung cancer, focusing on their role in ferroptosis regulation and NF-κB pathway activation. Results: PS-MPs were rapidly internalized by lung cancer cells and remained detectable across multiple passages. Exposure to PS-MPs promoted lung cancer cell proliferation, increased mitochondrial length, and elevated Ki67 and c-Myc expression. Following ionizing radiation, PS-MPs significantly attenuated radiation-induced ferroptosis, as evidenced by reduced mitochondrial damage, lipid peroxidation, and GSH depletion. Transcriptomic analysis revealed that PS-MPs activated the NF-κB pathway, leading to increased phosphorylation of IKKβ, IκBα degradation, and enhanced nuclear translocation of NF-κB. In vivo, PS-MPs accumulated in lung tumor-bearing mice, reducing radiotherapy efficacy by increasing tumor volume and weight while decreasing survival rates. Knockdown of NF-κB restored ferroptosis sensitivity and mitigated PS-MPs-induced radioresistance, confirming the NF-κB-dependent inhibition of ferroptosis. Innovation and Conclusion: This study provides the first evidence that PS-MPs impair radiotherapy efficacy in lung cancer by suppressing ferroptosis via NF-κB activation. Unlike previous research focusing on microplastic toxicity in normal tissues, our findings highlight their oncological impact and potential role as an environmental factor influencing cancer therapy resistance. These results emphasize the need for further investigation into microplastics as emerging disruptors of redox homeostasis in oncology and their broader implications for environmental and cancer research.

Project description:The toxic mechanisms of microplastics (MPs), which are emerging environmental pollutants, remain uncertain. MPs have been linked to severe liver metabolic disorders; however, the underlying mechanisms remain unclear. In this study, we found that the plasma concentrations of MPs, especially polystyrene MPs (PS-MPs), were higher in patients with metabolic dysfunction–associated steatotic liver disease (MASLD) than in healthy controls and showed a positive correlation with liver enzymes and serum lipids, as detected by pyrolysis gas chromatography–mass spectrometry. Exposure to PS-MPs of 2 μm in size resulted in a significant increase in the severity of fibrosis and inflammation and exacerbated mitochondrial dysfunction and mitophagy disorders in livers of db/db mice and palmitic acid (PA)–treated HepG2 cells. RNA sequencing and proteomics analyses demonstrated that the mitophagy and the autophagy-related protein tyrosine kinase 6 (PTK6) underwent significant changes. Molecular docking, surface plasmon resonance (SPR), and molecular dynamics studies confirmed that PS-MPs stably interacted with PTK6. Furthermore, when PTK6 was silenced, PS-MPs treatment failed to exacerbate mitophagy in PA-treated HepG2 cells and PS-MPs inhibited mitophagy in mitophagy agonist CCCP-stimulated HepG2 cells. Collectively, these findings suggest that PS-MPs exacerbate liver damage and mitochondrial damage via PTK6-mediated mitophagy in MASLD, providing valuable insights into the hepatotoxic effects of MPs on human liver health and suggesting potential therapeutic targets for MASLD

Project description:Human ingestion of microplastics (MPs) is common and inevitable due to the widespread contamination of food items, but implications on the gastric digestion of food proteins are still unknown. In this study, the interaction between pepsin and polystyrene (PS) MPs was uncovered by investigating its effect on enzyme activity and conformation while in a simulated human gastric environment. The impact on food digestion was also determined by monitoring the kinetics of protein hydrolysis through static in vitro gastric digestion of cow’s milk contaminated with PS. The binding of pepsin to PS showed that surface chemistry of MPs dictates binding affinity. The key contributor to pepsin adsorption seems to be π−π interactions between the aromatic residues and the PS phenyl rings. During quick exposure (10 minutes) of pepsin to increasing concentrations (222, 2219, 22188 particles/mL) of 10 μm PS (PS10) and 100 μm PS (PS100), total enzymatic activities were not affected remarkably. However, upon prolonged exposure at 1 and 2 hours, preferential binding of pepsin to the small, low zeta-potential PS caused structural changes in the protein which led to a significant reduction of its activity. Digestion of cow’s milk mixed with PS10 resulted in transient accumulation of larger peptides (10-35 kDa) and reduced bioavailability of short peptides (2-9 kDa) in the gastric phase. This, however, was only observed at extremely high PS10 concentration (0.3 mg/mL or 5.46E+05 particles/mL). The digestion of milk peptides, bound preferentially over pepsin on the hard corona around PS10, was delayed up to 15 minutes in comparison to bulk protein digestion. Intact caseins, otherwise rapidly digested, remained bound to PS10 in hard corona for up to 15 minutes. This work presents valuable insights regarding the interaction of MPs, food proteins, and pepsin, and their dynamics during gastric digestion which provides new knowledge and understanding on the potential risks of MPs to human health.

However, current understanding of MP toxicities are based on cell population-averaged measurements. Here we used single-cell RNA sequencing to provide the transcriptome heterogeneity of 12000 intestinal cells obtained from zebrafishes exposed to 100nm, 5μm and 200μm polystyrene MPs (PS-MPs) for 21 days. Eight intestinal cell populations were identified. We found that all the three sizes of PS-MPs induced dysfunction of intestinal immune cells (including phagosome and regulation of immune system process).
