Effect of MbovP475 on gene expression in BoMac cells

ABSTRACT: MbovP475 of M. bovis is a nucleomodulin and has the ability to enter into cells nucleus during infection. Recently, we verifed that MbovP475 can regulate the hostone PTMs. To detect the effect of MbovP475 of M. bovis on gene expression in BoMac cells, we constructed BoMac cells stablely expressing MbovP475. Our data provide globaly change of mRNA profile caused by MbovP475.

Project description:MbovP475 of M. bovis is a nucleomodulin and has the ability to enter into cells nucleus during infection. Recently, we verifed that MbovP475 can regulate the hostone PTMs. We found MbovP475 promoted the H3K4me3 and H3K27ac. So we proposed MbovP475 regulated gene expression in BoMac cells by promoting H3K4me3 and H3K27a, we constructed BoMac cells stablely expressing MbovP475. Our data provide globaly genes marked by H3K4me3 and H3K27ac enhanced by MbovP475.

Effect of M . bovis NX2-EVs on gene expression in BoMac cells

Project description:Our research shows that EVs derived from M. bovis-infected BMEC cells (EVs-M. bovis NX2) could regulate BoMac cell injury by inducing inflammation. To detect the effect of EVs-M. bovis NX2 on gene expression in BoMac cells, we incubated Ctrl-EVs and EVs-M. bovis NX2 with BoMac cells for 12h, 24h. Our data provide globaly change of mRNA profile caused by EVs-M. bovis NX2.

Mycoplasma bovis nucleomodulin MbovP202 modulates gene expression in BoMac cells

Project description:We identified a novel M. bovis nucleomodulin, MbovP202, which functions as a 5-methylcytosine-specific DNA methyltransferase that localizes to the nucleus of the bovine macrophage cell line (BoMac). To investigate its regulatory role, we performed RNA sequencing to compare the mRNA expression profiles between BoMac and BoMacMbovP202 cells. A total of 2,790 differentially expressed genes (DEGs) were identified between the two groups. Integrated analysis of Whole Genome Bisulfite Sequencing (WGBS) and RNA-Seq data revealed that genes with promoter hypermethylation and concomitant down-regulated expression were predominantly enriched in metabolic and inflammatory pathways.

Genome-wide maps of MbovP475-binding sits in BoMac cells.

Project description:We used Next-generation sequencing (NGS) to reveal MbovP475-binding sites in BoMac cells. We extracted the genome of BoMac infected with mycoplasma bovis by sonication, then the sheared chromatin in the supernatant was incubated with purified rabbit polyclonal IgG against MbovP475 and protein A/G magnetic beads. The DNA was eluted from the beads and subject to making sequencing library. We totally got 2648 potential MbovP475-binding sites by ChIP-seq. Among them, CRYAB and MCF2L2 gene were further verifed to be down-reguated by MbovP475 through binding to the promoter region of two genes.

Project description:In this work, we detect the effect of EVs derived from M. bovis-infected BMEC cells (M. bovis NX2-EVs) on regulating the inflammatory response of BoMac. The results indicate M. bovis-infected BMEC cells promote the inflammatory response in BoMac cells by releasing EVs. We furtherly identify the miRNAs in EVs involved in promoting inflammatory response. Understanding their cargo content will provide new insights into the molecular mechanisms of these respective EVs.
