Gene expression in lung resident macrophages and recruited macrophages in pulmonary hypertension

ABSTRACT: We generated a Lineage tracing system to trace the fate of resident macrophages and recruited macrophages using GFP and RFP fluorescence respectively, in a SuHx-induced mouse PH model. Cx3cr1+GFP+resMФs and RFP+recMФs was achieved through bone marrow transplantation system and flow cytometry sorted. Gene-expression data was collected via microarray analysis.

Time-resolved single-cell RNAseq profiling identifies a novel Fabp5-expressing subpopulation of inflammatory myeloid cells in chronic spinal cord injury

Project description:Traumatic spinal cord injury (SCI) is a debilitating central nervous system (CNS) pathology that lacks effective restorative treatments. The mechanisms of tissue recovery after SCI are dysregulated in comparison to normal wound healing, leading to a chronic wound state, rather than a return to homeostasis. While this state is characterized by persistent inflammation driven by CNS resident microglia (MG) and infiltrating myeloid cells, the precise roles of myeloid cell subsets after SCI remains unclear. In particular, upon crossing the blood-brain barrier, tissue infiltrating monocyte-derived macrophages (MCd) take on the morphology of MG, and upregulate canonical MG markers2, making the two populations practically indistinguishable. Here, we first employed a Cx3cr1-CreERT2 mouse model to label Cx3cr1+ cells for FACS isolation. Then we utilized fate-mapping approach (see methods) to investigate the ontology and temporal dynamics of Cx3cr1+ spinal cord MG (RFP+/YFP+) and Cx3cr1+ infiltrating myeloid cells (RFP-/YFP+) via FACS isolation and single-cell RNA sequencing (scRNAseq) in a mouse model of thoracic contusion SCI. Transcription profiling of mouse peripheral blood and alveolar recruited mononuclear phagocytes in Pam3CSK4 treated animals

Project description:Aim of this project was to examine the global gene expression profiles of mononuclear phagocytes recruited from peripheral blood to the alveolar space following alveolar deposition of the TLR2-ligand Pam3CSK4 in transgenic CX3CR1+/GFP mice. Experiment Overall Design: Alveolar macrophages (AM) and peripheral blood monocytes (PBM) were isolated from broncho-alveolar lavages and from blood, respectively, using FACS. Expression profiles from AM and PBM of the same mice were compared on the same slides. Each labeled RNA sample contained RNA pooled from six individuals. Four pairs of RNA from corresponding AM and PBM pools were hybridized, giving a total amount of 24 individual mice analyzed. Two hybridizations were performed with AM labeled with Cy3 and PBM with Cy5, two hybridizations were performed with swaped dyes.

Project description:We performed molecular phenotyping of RV remodelling in pulmonary hypertension (PH), using transcriptome analysis of RV tissue obtained from SuHx-induced male rats.When compared to human RV decompensated PH, the molecular characterisation of these rats showed that ECM and collagen fibrillar organisation were highly upregulated in SuHx rats and human profiles along with responses to cytokines. Moreover, growth differential factor was found common in SuHx and RV human samples.

Inhibition of Intimal CX3CR1+ Macrophages Mitigates Thoracic Aortic Aneurysm Progression in Marfan Syndrome

Project description:Current understanding on the pathogenesis of thoracic aortic aneurysms (TAAs) in Marfan Syndrome (MFS) mainly focuses on vascular smooth muscle cell (VSMC) pathologies. Whether immune cell-mediated inflammation drives the progression of TAAs is still elusive. Using single-cell RNA sequencing, a subset of macrophages highly expressing CX3CR1 and mainly locating in the aortic intima was identified in aortic root and ascending aortas from Fbn1C1041G/+ mice, and further validated in MFS patients. Specific elimination of CX3CR1+ macrophages by diphtheria toxin in Cx3cr1-CreERT2iDTRF/+Fbn1C1041G/+ mice efficiently ameliorated TAA progression, suggesting their pathogenic action. Using monoclonal antibodies Adalimumab and Teprotumumab to respectively neutralize the proinflammatory cytokines TNFalpha and IGF1 produced by CX3CR1+ macrophages from MFS patients substantially suppressed CX3CR1+ macrophage-mediated inflammation in MFS patient-specific induced pluripotent stem cell (iPSC)-derived VSMCs. Furthermore, bone marrow transplantation and parabiosis using Cx3cr1GFP/+ mice revealed the intimal CX3CR1+ macrophages derived from circulating monocytes. Administration of CCR2 antagonist RS504393 to inhibit monocyte infiltration significantly reduced intimal CX3CR1+ macrophages and subsequently alleviated TAA development in Fbn1C1041G/+ mice. In conclusion, intimal CX3CR1+ macrophages mediated TAA formation through paracrinally causing VSMC inflammation. Targeting intimal CX3CR1+ macrophages would be a promising anti-inflammatory strategy in TAA therapy for MFS.

DNA methylome and transcriptome sequencing to define pathways involved in the activation of microglia

Project description:Microglia cells are macrophages which reside in the brain, and become activated during the inflammatory processes involved in neurodegenerative and autoimmune disorders. An understanding of the molecular processes involved in this activation process could lead to the development of new neuroprotective therapies. To reveal molecular mechanisms involved in microglia activation, we performed RNA-seq and MBD-isolated genome sequencing (MiGS or MBD-seq) on both naïve and peak microglia derived from the mouse experimental autoimmune encephalomyelitis (EAE) model. The EAE model reflects some of the histological and immunological features of multiple sclerosis (MS) in humans. Mice genetically engineered to label Ccr2 with red fluorescent protein (rfp) and Cx3cr1 with green fluorescent protein (gfp) (“red-green mice” as described in Saederup et al., 2010 and Mizutani et al., 2012) were employed and microglia (positive for gfp) were separated from infiltrating monocytes using flow cytometry.
