Development of mRNA expression signatures for Nao Yi Kang decoction in treating intracerebral hemorrhage

ABSTRACT: To identify the potential mRNA asscociated with Nao Yi Kang decoction (NYKD) in treating intracerebral hemorrhage (ICH), we have employed mRNA expression profiling. 2405 and 1806 mRNAs were identified in the ICH vs sham group and the NYKD vs ICH group, respectively. Specifically, 294 DEmRNAs were intersected among the three groups.

Project description:To identify the potential lncRNA and mRNA asscociated with Buyang Huanwu decoction (BYHWD) in treating intracerebral hemorrhage (ICH), we have employed lncRNA microarray expression profiling. 1596 and 1114 mRNAs were identified in the ICH vs sham group and the BYHWD vs ICH group, respectively. Specifically, 180 DEmRNAs were intersected among the three groups. 2287 and 2245 lncRNAs were identified in the ICH vs sham group and the BYHWD vs ICH group, respectively. Specifically, 342 DElncRNAs were intersected among the three groups. Expression of four mRNA (Spata2, Nkiras, Mef2c, Sh2b3) and lncRNA (Gm16045, Gm14005, Gm41118, 493304012Rik) from this signature was quantified by qPCR.

Project description:To identify the potential miRNA asscociated with Buyang Huanwu decoction (BYHWD) in treating intracerebral hemorrhage (ICH), we have employed miRNA expression profiling. 126 and 38 miRNAs were identified in the ICH vs sham group and the BYHWD vs ICH group, respectively. Specifically, 15 DEmiRNAs were intersected among the three groups. Expression of 14 miRNA (miR-7b-5p, miR-770-3p,miR-760-3p, miR-376c-5p, miR-1224-5p, miR-298-5p, miR-541-3p, miR-344d-3-5p, miR-653-5p, miR-7a-5p, miR-6540-5p,miR-146a-5p, miR-375-3p, and miR-3962) from this signature was quantified by RT-qPCR.

Project description:To identify the potential lncRNA and mRNA post intracerebral hemorrhage (ICH), we have employed lncRNA microarray expression profiling. 570 mRNAs and 313 lncRNAs were identified in the ICH vs sham group .Expression of six mRNA (Saa3, Col10a1, Mmp13, Oxt, Pcdhb6 and Resp18) and lncRNA (ENSMUT00000141700, AK143011, ENSMUT00000128306, AK030988, ENSMUST00000187076 and ENSMUST00000134129) from this signature was quantified by qPCR.

TMT-based quantitative proteomics reveals the protective mechanism of Tenuigenin after experimental intracerebral hemorrhage in mice

Project description:Tenuigenin (TNG), an extract obtained from Polygalae Radix, possesses anti-inflammatory,anti-oxidant, neuro-protective properties. However, the potential mechanism of TNG on intracerebral hemorrhage (ICH) has not been well studied. Therefore, in this study, we aimed to identify the prospective mechanism of TNG in treating ICH with the help of TMT-based quantitative proteomics. Our results showed that the optimal dose of TNG was 16mg/kg, which could markedly improve neurological functions, reduce cerebral edema, hematoma volume and hemoglobin level 72h after ICH. Furthermore,our proteomic results identified that a total of 404 DEPs (353 up and 51 down regulated) were identified in ICH+vehicle vs Sham group, while 342 DEPs (306 up and 36 down regulated) and 76 DEPs (28 up and 48 down regulated) were quantified in TNG vs Sham group and TNG vs ICH+vehicle group respectively. In addition, a total of 26 DEPs were selected out according to strict criteria. Complement and coagulation cascades was the most significantly enriched pathway and two proteins (MBL-C and Car1) were further validated as hub molecules.

RNA-seq of perihaematomal and anatomically matched contralateral brain tissue after intracerebral hemorrhage

Project description:Secondary injury causes death and dependence after spontaneous intracerebral haemorrhage (ICH). Having found that ICH is associated with activation of genes regulated by the transcription factor NRF2, we performed bulk RNA sequencing of perihaematomal and anatomically matched contralateral brain homogenate obtained at autopsy from a cohort of patients who died either within 24h of ICH or between 4-14 days of ICH onset, to investigate the spatiotemporal evolution of this Nrf2 activation.

Examining Transcriptomic Alterations in Rat Models of Intracerebral Hemorrhage and Severe Intracerebral Hemorrhage

Project description:Intracerebral hemorrhage (ICH) is a life-threatening condition associated with significant morbidity and mortality. Understanding the molecular mechanisms underlying ICH and its severe form is crucial for developing effective therapeutic strategies. This study investigates transcriptomic alterations in rodent models of ICH and severe intracerebral hemorrhage to shed light on the genetic pathways involved in hemorrhagic brain injury. We performed principal component analysis, revealing distinct principal component segments of normal rats compared to intracerebral hemorrhage and severe intracerebral hemorrhage rats. We further employed heatmaps and volcano plots to identify differentially expressed genes and utilized bar plots and KEGG pathway analysis to elucidate the different molecular pathways involved. Using comprehensive RNA sequencing and bioinformatics analyses, we identified a multitude of differentially expressed genes in both the ICH and severe ICH models. Our results revealed 5679 common genes among the normal, intracerebral hemorrhage, and severe intracerebral hemorrhage groups in the upregulated genes group, and 1196 common genes in the downregulated genes. A volcano plot comparing the groups further highlighted common genes, including PDPN, TIMP1, SERPINE1, TUBB6, and CD44. These findings underscore the complex interplay of genes involved in inflammation, oxidative stress, and neuronal damage. Furthermore, pathway enrichment analysis uncovered key signaling pathways, including the TNF signaling pathway, protein processing in the endoplasmic reticulum, MAPK signaling pathway, and Fc gamma R-mediated phagocytosis, implicated in the pathogenesis of ICH.

Altered Expression Profiles of Transfer-RNA-Derived Small RNAs Reveal the Regulating Functions in Rat Brain After Intracerebral Hemorrhage (ICH) and the Potential Therapeutic Targets of Buyang Huanwu Decoction in ICH

Project description:Transfer-RNA-Derived Small RNA (tsRNA) is s a novel class of short non-coding RNA including stress-induced tRNA fragments (tiRNA) and tRNA-derived fragments (tRF). Using RNA sequencing, we evaluated the tsRNA expression profiles in the brain of intracerebral hemorrhage (ICH) and sham rats at days 21. Meanwhile, tsRNA levels in ICH treated with the Traditional Chinese Medicine named Buyang Huanwu Decoction (BYHWD) were detected. Bioinformatics analyses indicated that tsRNAs were the important regulators in ICH and potential new therapeutic targets of BYHWD.

Identification of inflammation-related microRNAs deregulated in the plasma from intracerebral hemorrhage patients

Project description:We tested the hypothesis that circulating microRNAs (miRNAs) present in plasma might display a specific signature in patients with intracerebral hemorrhage (ICH). Global miRNA profiles were determined with the Agilent Human miRNA Microarray platform, 027233. ICH patients display a characteristic inflammation-related miRNA profile as compared to healthy controls. Plasma samples were collected from the following 6 subject groups: male ICH patients (n=8), female ICH patients (n=7), male healthy control (n=4), female healthy control (n=4), male ischemic stroke patients (n=8) and female ischemic stroke patients (n=8). Total RNAs isolated from 1 ml plasma were pooled for each group. A fixed volume of RNA sample was withdrawn from each pool and used for microarray detection.

Genome-wide DNA methylation pattern in whole blood associated with primary intracerebral hemorrhage

Project description:To reveal the role of DNA methylation in peripheral blood from primary intracerebral hemorrhage(ICH) ,DNAs from 30 ICH patients and 34 matched controls were analyzed by methylation microarray. In total, we identified 1377 hypomethylated and 153 hypermethylated differentially methylated positions (DMPs) in peripheral blood from ICH patients compared to healthy controls.
